An experimental study of scavenging oxygen free radicals with garlicin

摘要 目的 :观察大蒜素是否具有直接清除氧自由基的作用 ,以进一步解释大蒜素治疗缺血性心脑血管疾病的作用机制。方法 :应用黄嘌呤、黄嘌呤氧化酶等建立超氧阴离子自由基 (·O2 - )产生体系。应用过氧化氢(H2 O2 )及FeSO4 (NH4 ) 2 SO4 建立羟自由基 (·OH )产生体系。以电子顺磁共振仪分别测试大蒜素对·O2 - 及·OH的清除效果 ,并与川芎嗪比较。结果 :大蒜素在 8.42 7μmol/ml及 4.2 14 μmol/ml浓度的情况下对·O2 - 的清除率分别为 2 9.0± 8.83 %、19.8± 4.3 5 % ,川芎嗪的清除率分别为 2 2 .4± 7.47%、19.0± 12 .1% ,两种药物间差异不显著 (P >0 .0 5 )。大蒜素在 84.2 7μmol/ml、8.42 7μmol/ml、1.3 3 7μmol/ml浓度下对·OH的清除率分别为 10 0 %、65 .4± 6.3 2 %、42 .1± 5 .67% ,川芎嗪的清除率分别为 (高浓度不溶 ,未测 ) 4.48± 16.5 %和 6.2 2±10 .7% ,两种药物间差异显著 (P <0 .0 1)。结论 :大蒜素可直接清除氧自由基 ,可减轻缺血 再灌注损伤 。 Objective:To investigate whether garlicin could scavenge oxygen free radicals directly and further to explain the mechanisms of garlicin working on ischemic cardiovascular and cerebrovascular diseases.Methods:Superoxide radical production system was established with xanthine and xanthine oxidase.Hydroxy radical production system was established with H 2O 2 and FeSO 4 (NH 4) 2SO 4.The effect of scavenging free radicals by garlicin was detected with electron spin resonance instrument,and its effect was compared with ligustrazine.Results:When the concentrations of garlicin were 8.427 μmol/ml and 4.214 μmol/ml,the scavenging rates of garlicin to superoxide were 29.0±8.83% and 19.8±4.35%,respectively.And to ligustrazine,they were 22.4±7.47% and 19.0±12.1%,respectively.The comparison between garlicin and ligustrazine showed no significance( P >0.05).When the concentrations of garlicin were 84.27 μmol/ml,8.427 μmol/ml and 1.337 μmol/ml,the scavenging rates of garlicin to hydroxy radical were 100%,65.4±6.32% and 42.1±5.67%,respectively.And to ligustrazine,they were 4.48±16.5% and 6.22± 10.7% ,respectively.The comparison between garlicin and ligustrazine showed significance( P <0.01).Conclusion:Garlicin can directly scavenge oxygen free radicals and improve ischemia reperfusion injury in the cardiovascular and cerebrovascular diseases.
