摘要
建立了溴代十六烷基吡啶 (CPB)存在下 ,用间甲酚紫 (CP)为探针测定核酸的新方法 .在HMTA-HCl缓冲溶液 (pH =4 .5 )中 ,DNA的加入使CP -CPB体系的共振光散射 (RLS)增强 ,此RLS信号与DNA浓度成正比 ,fsDNA和ctDNA的线性范围分别为 0 .1~ 2 0 μg/mL和 0 .1~ 5 0 μg/mL ,检出限分别为 10ng/mL和 2 6ng/mL .
In the presence of CPB and CP as probe,a new determination method of nucleic acids was developed.In the HMTA-HCl buffer(pH4.5),the weak light scattering of m-cresol purple and CPB were enhanced greatly by addition of nucleic acids and the enhanced intensity of RLS was in proportion to the concentration of nucleic acids.The linear range is 0.1 to 20 μg/mL for fsDNA,and 0.1 to 50 μg/mL for ctDNA.The effective factors and the optimum conditions have been studied and the detection limits of fsDNA and ctDNA are 10ng/mL and 26 ng/mL.
