摘要
目的 :观察α_半乳糖苷酶对猕猴类人B抗原的酶解效果及α_半乳糖苷酶酶解后的红细胞进行异种血型动物输血的安全性。方法 :用 10 0U/mlα_半乳糖苷酶酶解猕猴红细胞表面的类人B血型抗原 ,吸收放散法检测猕猴血型及α_半乳糖苷酶酶解后猕猴红细胞类人B血型抗原清除情况 ;同时检测酶解对红细胞结构与功能的影响 ;回输实验检测红细胞存活率以及观察酶解后红细胞的安全性。结果 :α_半乳糖苷酶可以清除猕猴红细胞表面类人B抗原 ,而不影响红细胞结构、功能及其在体内存活 ;α_半乳糖苷酶酶解的类人B型红细胞输给类人A型血的猕猴 ,未发生输血反应 ;受血猴的血液生化及尿液各项指标与输血前相比 ,无明显变化。结论 :α_半乳糖苷酶可以清除猕猴红细胞表面类人B抗原 ,酶解后的红细胞输给类人A型猕猴是安全的。
Objective:To observe the enzymatic treatment effects of α_galactosidase on human_like B antigens on the surface of rhesus monkey red blood cell (rRBC) as well as to study the safety of transfusion with enzyme_treated rRBC.Methods:rRBC were treated with 100?U/ml α_galactosidase. Human_like B antigens on enzyme_treated cells were analyzed by means of absorbing and resuspending examination. The effects of enzyme treatment on the rRBC structure and function were observed. The enzyme_treated rRBC were labeled with fluorescin and injected into recipient monkey (blood groups like human A). The in vivo survival fraction of enzyme_treated cells was tested with flow cytometry. The biochemical and physiological properties of recipient blood and urine samples were investigated. Results and Conclusions:Human_like B antigens on rRBC surface disappeared by treatment with 100?U/ml α_galactosidase. The enzyme_treated cells could survive normally in recipient monkey with type A_like blood. The enzyme treatment had no clinical effect on biochemical and physiological properties of recipient monkey blood and urine samples. The results suggest that there may be great possibility of using α_galactosidase treated_group B red blood cells for clinical transfusion.
