摘要
目的 构建以尿路致病性大肠杆菌Ⅰ型菌毛 fimH基因为靶位的真核表达质粒 ,并使其在小鼠肌肉中表达。 方法 应用PCR方法及基因重组技术 ,克隆了尿路致病性大肠杆菌标准菌株J96 fimH基因 ,并将fimH基因插入真核表达载体pcDNA3中。免疫BALB/c小鼠 ,用免疫组化染色法观察表达情况。结果与结论 成功地构建了真核表达质粒pcDNA3-fimH ,序列分析显示 ,fimH基因核苷酸序列与GenBank上登录的一致 ,fimH蛋白在小鼠肌肉组织细胞内呈分散表达。
