摘要
以禽流感H5和H9亚型病毒分别免疫Balb/c小鼠 ,取其脾细胞与SP2 / 0的骨髓瘤细胞融合 ,用血凝抑制试验(HI)检测细胞培养上清 ,结果获得了 6株特异性单克隆抗体 ,其中抗禽流感H5亚型病毒血凝素特异性单克隆抗体细胞株 3株 ,分别命名为 4B6、4A3、3H1;抗H9亚型病毒血凝素单克隆抗体细胞株 3株 ,命名为 6E6、6B6和 5B4。这些单克隆抗体小鼠腹水HI效价为 2 13~ 15,细胞培养上清抗体HI效价为 2 7~ 8。研究结果表明 ,所有这些单克隆抗体仅与试验的相应H5或H9亚型病毒株发生特异性反应 ,而不能与鸡新城疫病毒、鹅新城疫病毒、鹅腺病毒和鸡产蛋下降综合征病毒(EDS76 )等反应。实验室检测结果证明 ,应用这些单克隆抗体能在 2 4h内迅速检测出相应的禽流感病毒。
A panel of monoclonal antibodies specific to hemagglutinin of subtype H5 and H9 avian influenza virus (AIV_H5 and AIV_H9) were developed by fusing between SP2/0 myeloma cells and spleen cells of Babl/c mice immunized with AIV_H5 and AIV_H9.Three of them were designated as 4B6,4A3,and 3H1 respectively,which were specific to AIV_H5,but not reacted with AIV_H9,Newcasttlet Disease virus from chickens(NDV),NDV from geese,adenovirus from geese and EDS 76 by hemagglutination inhibition test (HI) and immunofluorensense assay.Others were named as 6E6 and 5B4 respectively,which were against AIV_H9 but not reacted with other tested viruses.Chicken embryo fibroblast(CEF) cells infected with 10 TCID 50 of AIV_5 could been detected in 24 hours by these monoclonal antibodies in indirect immunofluorescent assay (IFA).The results suggested that these monoclonal antibodies will be very useful in control of avian influenza.
