糖尿病大鼠血管损伤后纤连蛋白的表达

Expression of fibronectin mRNA and protein in injured vessels for diabetic rats

摘要 目的 :观察糖尿病大鼠血管损伤后纤连蛋白 (FN)mRNA和蛋白的表达。方法 :Wistar大鼠 32只 ,随机分为正常对照组 (n =8) ,手术对照组及糖尿病组 (均n =12 ) ,糖尿病组单次腹腔注射链脲佐菌素 (STZ)制成糖尿病模型。后 2组大鼠均行胸主动脉至左髂总动脉球囊损伤后 ,每组选 8只用于试验。术后 14d用Northern印迹杂交测定血管平滑肌细胞FNmRNA的表达 ,用免疫组化检测FN在血管壁的表达。结果 :与手术对照组相比 ,糖尿病组FNmRNA和蛋白表达均明显增高 (P <0 .0 1) ,而此 2组又均高于对照组 (P <0 .0 1)。结论 Aim:To investigate the expression of fibronectin(FN)mRNA and protein in injured vessels in diabetic rats.Methods:Wistar rats were randomly allocated into control group,angioplasty group,and diabetic plus angioplasty group (diabetic group).Diabetic group was served with stretozotocin (STZ) 60 mg/kg for each rat by intraperitoneal injection.The angioplasty group and diabetic group were treated with balloon denudation from thoracic to left iliac aorta.Fourteen days after the operation,thoracic aortae for each group were collected to detect vascular smooth muscle cells(VSMCs)FN mRNA expression with Northern blot and the protein expression of vessel wall FN with immunohistochemistry. Results: Expression of FN mRNA and protein were significantly higher in diabetic group than angioplasty group ( P <0.01) and those in these 2 groups were both higher than control group. Conclusion:These findings indicates that the pathogenesis of restenosis in injured vessels is related to the higher expression of FN mRNA and protein in diabetic rats.
