摘要
目的 :探讨哮喘大鼠CRHmRNA(促肾上腺皮质激素释放激素信使核糖核酸 )、血浆CORT(皮质酮 )变化及补肾定喘汤 (鹿角、肉苁蓉、巴戟天、熟地等 )对它们的作用。方法 :采用放射免疫、半定量RT PCR(逆转录 聚合酶链反应 )技术 ,分别观察血浆CORT、下丘脑CRHmRNA水平的变化。结果 :哮喘大鼠血浆CORT水平降低 (P <0 .0 1) ,下丘脑CRHm RNA水平明显降低。补肾定喘汤治疗能明显促进CRH基因表达 ,升高血浆CORT水平。结论 :哮喘模型大鼠存在着HPA轴抑制 ,补肾定喘汤能有效地保护和提高受抑的HPA轴的功能。结果
Objective: To study the change of the blood level of CORT and expression of CRHmRNA in asthma-tic rats and the effects of Bushendingchuan Decoction(Cornu Cervi, Herba Cistanches, Radix Morindae Officinalis, Radix Rehmanniae Preparata, etc.) on them. Methods: The change of level in CORT was measured by radioimmunoassay, the change of level in hypothalamic CRHmRNA was measured by RT-PCR(Reverse transcription and polymerase chain reaction by the semi-quantitative determination). Results: The blood level of CORT decreased(P<0.01) and the level of hypothalamic CRHmRNA decreased obviously in asthmatic rats. The treatment of BSDCT obviously promoted the mRNA level of CRH, increased the blood level of CORT. Conclusion: There is suppression on the HPA axis in acute asthmatic rats. BSDCD can preserve and increase the function of the suppressed HPA axis effectively. The probable mechanism has been discussed in this paper.
