摘要
目的 :研究冬凌草甲素诱导白血病 K5 6 2细胞凋亡的机制。方法 :以不同浓度的冬凌草甲素作用于体外培养的 K5 6 2细胞 ,检测细胞生长抑制率 ,观察细胞凋亡时的形态学变化 ,对细胞凋亡前后的端粒酶活性进行检测。结果 :冬凌草甲素可显著降低 K5 6 2细胞的端粒酶活性 ,抑制细胞的生长 ,诱导细胞发生凋亡。并呈现出明显的量 -效与时 -效关系。结论 :冬凌草甲素能抑制 K 5 6 2细胞的生长并诱导细胞发生凋亡 ,降低细胞端粒酶活性可能是其重要作用机制之一。
Objective:To study the mechanism of apoptosis induced by Orindonin in K562 cells.Methods:The variation of both morphology and inhibitory rate of K562 cells were observed in culture medium with various concentrations of Oridonin at different time in vitro. The activity of telomerase was detected before and after apoptosis occurred.Results:Oridonin could decrease the activity of telomerase, as well as cause apoptosis and inhibit the growth of K562 cells significantly.The suppression was both time and dose dependent.Conclusion:Orindonin can inhibit the growth of K562 cells and induce apoptosis , decreasing the activity of telomerase may be one of the most important biochemical mechanisms.
