应用非离子去垢剂抽提和扫描电镜样品制备、观察相结合的方法 ,显示了镰游仆虫的腹面皮层细胞骨架 ,详细描述了处于毛基体和毛基体下水平的口围带、口侧膜、额腹横棘毛骨架 ,以及口围带小膜托架、口侧膜托架、额腹横棘毛托架的主要附属纤维和非纤毛区表膜下皮层骨架的立体图形。作者据所述各种纤毛器托架附属纤维的定位和分布特征推测 ,这些附属结构可能与细胞内各种纤毛器间的联系 ,以及包括纤毛器运动在内的整个细胞运动的协调等有关.
The ventral cortical cytoskeleton of the ciliated protozoan Euplotes harpa, obtained with the non-ionic detergent extraction method, was observed with scanning electron microscopy. This revealed that: (1) In the adoral zone of membranelles (AZM), paraoral membrane- and fronto-ventral cirri cytoskeleton, the transverse link fiber connects two kinetosomes within the same row of kinetosomes while the longitudinal link fiber connects two kinetosomes of the anterior and posterior rows of kinetosomes on the kinetosomal level. The fiber is the structural component in every ciliature bracket beneath the kinetosomes. The fibers are woven into the shape of a network. Each bracket of the AZM membranelle and the fronto-ventral cirri are a sheet-like structure of fibers in which each fiber is similar in thickness. The sheet-like bracket of the paraoral membrane consists of two groups of fibers, one group parallel to each other transversely and the other group longitudinally. The regulated pores of the bracket are the place of attachment of the kinetosome. The three-dimensional morphology of the ciliature skeleton has not been reported in other species of Euplotes. (2)The post-bracket root fibers are associated with the AZM membranelle bracket, the post-oral fibers are associated with the paraoral membrane bracket, the anterior and posterior longitudinal fibers are associated with the fronto-ventral cirri bracket, and the anterior longitudinal fibers are associated with the transverse cirri bracket. These fibers are primary bracket-associated fibers. Besides shaping the cortex, support, movement of cilia, etc., the attached fibers were evaluated for possible connective functions between the various tracts of cilia and coordination in cellular motion, including cilia motion. (3) Beneath the pellicle of the fronoto-ventro-transverse cirri region, there are, consistent with previous observations on Euplotes sp., three layers of fibers, i.e. longitudinal fibers, spherule-coiled fibers and deep fibers.
