长期应用吗啡对大鼠性腺轴β-内啡肽及其受体mRNA的影响 被引量:5

The Effect of Morphine on the Expression of β-EP and μ-opioid Receptor mRNA in Hypothalamic-Pituitary-Ovarian Axis of Rats

摘要 目的:初步探讨长期应用吗啡导致月经周期紊乱乃至闭经的发生机制。方法:建立长期应用吗啡成瘾大鼠模型,证实动情周期已紊乱。免疫组织化学方法及图像分析系统观察下丘脑、垂体和卵巢茁-内啡肽含量,原位杂交方法测定下丘脑、垂体和卵巢的滋-阿片受体mRNA的表达。结果:长期应用吗啡时,下丘脑、垂体和卵巢茁-内啡肽的含量下降,而滋-阿片受体mRNA表达增强。结论:吗啡竞争性结合茁-内啡肽结合位点,对性腺轴产生抑制是引起月经周期紊乱的机制之一。 Objective: To study injury mechanism of morphine on hypothalamic-pituitary-ovarian axis (H-P-O axis) of female rats. Methods:The model rats were made by injecting morphine. Using immuno- histochemistry technique and image analysis system,authors observed the effect of morphine on the expression of β -EP on H-P-O axis.And using in situ hybridization authors observed the effect of morphine on expression of μ -opioid receptor mRNA.Results:It was detected to have μ -opioid receptor mRNA hybridization signal and β -endorphine immune activity in H-P-O axis. It was showed that injecting morphine for long time reduces the positive substance of β -EP on H-P-O axis. But the expression of opioid receptor mRNA was increased significantly than the control. Conclusion:It suggests that morphine injuries H-P-O axis and reduces the secretion of β -EP by combining with μ -opioid receptor.
