摘要 目的 :了解NT - 3在挤压伤脊髓背角表达的变化 .方法 :制作成年SD大鼠T13 -L1节段脊髓挤压伤模型并设正常对照组 ,分别于术后 2 4h ,7d ,2 1d处死动物 ,取L3 节段脊髓制作 2 0 μm厚冰冻连续切片 ,用抗NT - 3抗体行免疫组织化学ABC法染色 .观察NT - 3在背角的表达 ,记录其免疫反应强度 ,计数其免疫阳性神经元数和胶质细胞数 .结果 :NT - 3的阳性反应物主要分布于脊髓背角的神经元和胶质细胞 .挤压伤后NT - 3的表达从术后 2 4h至术后 7d进行性减少 ,而术后 2 1d时则增加并高过正常者水平 .结论 :NT - 3不仅参与脊髓的正常生理活动 。 Objective: The aim of this study is to explore the expression of NT-3 in spinal dorsal horn following crushed spinal cord injury (T 13 -L 1). Methods: The adult srague-daweley rat model of crushed spinal cord injury established in our laboratory was used. The rats were sacrificed respectively after 24?hours, 7?days, and 21?days of operation, and the L 3 spinal segments were removed out sectioned into sections of 20?μm in thickness . The sections were stained with anti-NT-3 antibody by ABC method of immmunohistochemishtry technique. The immunoreactive intensity of NT-3 and the number of positive neurons and glial cells in the dorsal horn were observed and counted under light microscope. Results: The intensity and positive cells of NT-3 in the dorsal horn decreased from 24?hours to 7 days at first and then increased at 21?days after operation. Conclusion: It indicated that NT-3 be related to the postoperative reaction during the early period of the crushed spinal cord injury.
