摘要
研究了热水法及复合酶法提取百合多糖 ( LBP)的最佳条件 ,对两种方法得到的多糖的生物活性进行了比较分析 .发现复合酶法能显著提高百合多糖的提取率 ,可达 31 .0 3% ,是热水法的 2 .85倍 .复合酶法提取的多糖在体外抗氧化活性如对 O- 2 ·及· OH的抑制作用、对 H2 O2 诱导的人血红细胞氧化溶血的抑制作用及人红细胞膜脂质过氧化的抑制作用均显著高于热水法提取的多糖 .经 Sephadex G- 75柱层析对粗多糖进行分离 ,得到 3种组分 ,其中活性组分 L BP- 3主要存在于酶法提取的多糖中 ,测得该活性组分的分子量为 1 340 0 .
Two methods of extracting Lilium brownii polysaccharide, hot-water method and combined-enzyme method, were studied and the optimum conditions of the two methods were found. The yield of the polysaccharides extracted by combined enzymes was (2.8) times that of polysaccharides extracted by hot-water. It was also found that the antioxidative activity of the polysaccharides, such as inhibiting effects on O^-_2·, ·OH, the hemolysis of human RBC induced by (H_2O_2) and the formation of MDA, extracted by combined-enzymes were much greater than that of those extracted by hot-water. After extracted by the two methods, the polysaccharides were separated by sephedex G-75 gel filtration chromotography and three fractions were eluted out. LBP-3, whose molecular weight was about (13 400) and had the highest antioxidative activity of the three fractions, was found to exist in polysaccharides extrated by combined enzymes much more than that to exist in those extracted by hot-water.
