马钱子碱对一氧化氮诱导软骨细胞凋亡的影响 被引量:40

摘要 目的:观察中药马钱子碱对一氧化氮(nitricoxide,NO)在体外诱导的兔软骨细胞凋亡的影响。方法:取体外培养的新西兰兔软骨细胞,分别加入硝普钠(SNP)培养12,24,48h,用透射电镜和流式细胞仪观察软骨细胞凋亡的变化,并以不同剂量马钱子碱加入NO诱导的软骨细胞凋亡体系培养24h,采用annexinV/PI方法检测凋亡情况。结果:在体外培养兔软骨细胞加入硝普钠24h,流式细胞仪检测早期凋亡率为(38.7±6.5)%。空白对照组为(2.4±1.4)%(t=10.90,P<0.01),透射电镜可见典型软骨细胞早期凋亡形态改变,并有一定时间依赖性,加入不同剂量马钱子碱,其凋亡率明显下降,并以高剂量最为显著为(2.9±1.1)%。马钱子碱高剂量组加入硝普钠后与正常软骨细胞组比较,二者凋亡率无显著差异性(t=0.51,P>0.05)。结论:硝普钠在体外能诱导软骨细胞凋亡,马钱子碱能明显降低NO诱导的软骨细胞早期凋亡,提示马钱子碱能有效治疗骨关节炎,可能与抑制软骨细胞早期凋亡有关。 AIM:To observe the effects of brucine on chondrocyte apoptosis of rabbit induc ed by nitric oxide (NO) in vitro. METHODS:The chondroytes obtained from New Zealand rabbits were cultured in vit ro with Sodium Nitroprusside (SNP) for 12, 24 and 48 hours respectively.Transmis sion electron microscope (TEM) and flow cytometer were employed to observe the c ell apoptosis. Then the chondrocytes were cultured with different dosage of bruc ine for 24 hours in vitro, and the annexin V/pI method was conducted to detect t he early chondrocyte apoptosis induced by NO. RESULTS:The flow cytometer showed that the apoptosis rate of chondrocytes cult ured in SNP for 24 hours was (38.7±6.5)%, and that of the control group was (2 .4±1.4)%(t=10.90,P< 0.01).Through TEM, the early typical morphological change of chondrocytes was seen with the time dependence. After cultured with different dosage of brucine, the apoptosis rate declined significantly, especially with t he high dosage of brucine(2.9±1.1)%. There was an insignificant difference in the apoptosis rate between the high dose group with SNP and normal control group (t=0.51,P >0.05).
