摘要
目的 观察联用阿米洛利与姜黄素预防大鼠肝纤维化的疗效。方法 建立二甲基亚硝胺大鼠肝纤维化模型 ,观察阿米洛利与姜黄素预防后 ,肝 /体和脾 /体比值、血及肝组织生化、羟脯氨酸、透明质酸及层黏连蛋白含量变化。结果 除阿米洛利组 ,各预防组较模型组肝功能得到改善 (P <0 0 0 1) ;阿米洛利和姜黄素预防组较模型组血清透明质酸、层黏连蛋白和肝组织羟脯氨酸含量降低 (P <0 0 5) ;预防组肝组织内SOD、GSH_PX较模型组升高 ,而MDA低于模型组 (P <0 0 1) ;尤以合用组效果最佳。结论 阿米洛利与姜黄素对二甲基亚硝胺诱导的肝纤维化均有预防作用 。
Objective To investigate the preventive effect of combination of amiloride and curcumin o n experimental liver fibrosis. Methods The rat liver fibrosis was induced by dimethlnitrosamine(DMN) and liver/weight index,spleen/we ight index,serum and tissue biochemical indexes,liver hydroxyproline (Hyp) co ntent,serum Hyaluronic Acid (HA) and Laminin (LN) content were detected after tr eating by amiloride and curcumin. ResultsSerum AST and ALP were decreased in all preventive grou ps,while serum A/G were increased in all preventive groups,liver function was am eliorative in preventive groups compared with those of DMN group except amiloride group ( P <0 001).Serum H A,LN and liver Hyp con tent were declined in all preventive groups compared with those of DMN group ( P <0 05).Liver SOD,GSH_PX were higher in preventive groups than those in the model group,while MDA was reduced ( P <0 01). Conclusion Both amiloride and curcumin show prophylactic effect on DMN induced rat liver fi brosis.The coeffects on rat liver fibrosis induced by DMN were observed in combi nation of amiloride with curcumin.
