摘要
目的 建立了用RP HPLC对赤芍中芍药苷、芍药内酯苷以及苯甲酸同时定量的方法 ,并对不同产地赤芍进行考察。方法 高效液相色谱法 ,色谱柱Hypersil C18(2 0 0mm× 4 .6mm ) ,流动相 :乙腈 水 (12∶88) ,磷酸调 pH =3.0 ,流速为 0 .8mL·min-1,检测波长为 2 30nm ,柱温为室温。用内标法同时测定了赤芍中芍药苷、芍药内酯苷及苯甲酸的含量。结果 芍药苷、芍药内酯苷及苯甲酸浓度与峰面积呈良好的线性关系。平均回收率芍药苷为 98.6 % ,RSD为 3.0 % ;芍药内酯苷为 98.6 % ,RSD为 3.1% ;苯甲酸为 96 .3% ,RSD为 3.2 %。结论 本方法测定了 17个不同产地、不同批号赤芍样品中 3种化学成分的含量 ,该方法分离度好 ,快速 ,简便 ,重现性好。
OBJECTIVE: A reserved-phase HPLC method was developed for the determination of paeoniflorin, albiflorin and benzoic acid in red peony root and 17 samples from different habitats were measured. METHODS: A Hypersil-C 18 column (200 mm × 4.6 mm) was used with the mobile phase of acetonitrile-water (12:88), adjusted pH=3.0 by phosphoric acid. The flow rate was at 0.8 mL·min-1. The detection wavelength was 230 nm at room temperture. 17 samples were determined. RESULTS: The linear response ranged from 18.24 ∼ 273.6 μg·mL-1 for paeoniflorin (r = 0.999 4, n = 7), 4.96 ∼ 74.4 μg·mL-1 for albiflorin (r = 0.999 5, n = 7) and 2. 168 ∼ 21.68 μg·mL-1 for benzoic acid(r = 0.999 8, n = 6) respectively. The recoveries were 98.6% with RSD 3.0% for paeoniflorin, 98.6% with RSD 3.1% for albiflorin and 96.3% with RSD 3.2% for benzoic acid. CONCLUSION: The method is quick, simple and repeatable.
