碱性成纤维细胞生长因子对脊髓损伤后bcl-2及bax基因表达的影响 被引量:6

摘要 目的:研究碱性成纤维细胞生长因子(bFGF)对脊髓损伤后bcl-2和bax基因表达的影响。方法:利用AllenWD法以25gcf致伤SD大鼠脊髓制作损伤模型,经蛛网膜下腔导管于术后即刻,0.5,1,2,3,4,6,12,24,48h导管注入bFGF20μL(含bFGF200U);对照组相同时间点注入等量生理盐水作对照。术后2,6,12,24,48h取损伤脊髓组织,采用免疫组化和原位杂交方法分别检测Bcl-2、Bax蛋白和bcl-2mRNA基因的表达。结果:脊髓损伤后应用bFGF显著促进bcl-2基因的表达,2,6,12,24,48hBcl-2平均光密度值分别为0.165±0.020,0.254±0.081,0.312±0.017,0.415±0.021,0.304±0.031,(P<0.01,t=2.729~9.279),Bax平均光密度值分别为0.034±0.021,0.184±0.014,0.204±0.014,0.216±0.027,0.180±0.013(P<0.01,t=4.601~6.376),提高Bcl-2蛋白的含量,降低了Bax蛋白的水平,每组数据差异具有统计学意义。结论:bFGF通过促进bcl-2基因的表达、抑制Bax蛋白的表达抑制脊髓损伤后神经细胞的凋亡,从而保护脊髓组织,这可能是bFGF对脊髓损伤具有保护作用的机制之一。 AIM:To investigate the temporal effect of basic fibroblast growth factor (bFGF) on the expression of bcl 2 and bax gene in the injured spinal cord of rats. METHODS:Sixty Sprague Dawley rats with 25 gcf impact to T8 SCI were randomly divided into two groups, experience group and control group. A thin plastic tube was situated in subarachnoid space below the injury level for perfusion. BFGF treated animals in experience group received bFGF 20 ul u(200 liquid)from the tube at 0, 0.5, 1, 2, 3, 4, 6, 12, 24 and 48 hours after injury. The saline treated animals received the equal volume of normal saline at the same time as control. The rats were sacrificed at 2, 6, 12, 24 and 48 hours postinjury(n=6).The tissues in the injured cord were collected and made into sections. The sections were tested by immunohistochemical method for the bcl 2 and bax protein. Bcl 2 mRNA was detected by in situ hybridization. RESULTS:The levels of both bcl 2 and bcl 2 mRNA in experience group after SCI were increased obviously than that in control group. The mean optical density of bcl 2 were (0.165± 0.020), (0.254± 0.081),(0.312± 0.017), (0.415± 0.021) and (0.304?à 0.031)respectively£¨ p<0.01, t=2.729£- 9.279£(c) . The mean optical density of bcl 2 were (0.034± 0.021), (0.184± 0.014), (0.204± 0.014)£? (0.216?à 0.027) and (0.180?à 0.013) respectively£¨ P< 0.01,t=4.601£- 6.376£(c) .However, the levels of bcl 2 protein increased and Bax protein of bFGF treated animals after SCI reduced.There were statistical significance in each data.
