摘要
目的评价米诺环素(MI)对慢性压迫性损伤(CCI)诱导大鼠神经病理性疼痛及分离小胶质细胞中IL-1浓度的影响。方法雄性SD大鼠被随机分为三组:Sham+vehicle、CCI+vehicle和CCI+MI。采用机械刺激反应和冷刺激反应评估大鼠的机械性和冷痛觉增敏。在CCI手术前及CCI手术后1、3、5、7、10和14 d进行行为学检测。CCI手术后14 d处死大鼠,从脊髓腰膨大分离小胶质细胞并进行培养。检测细胞培养24 h后上清液中白细胞介素-1(IL-1)浓度。结果与CCI+vehicle组比较,MI缓解CCI诱导的神经病理性疼痛,并且降低培养细胞的IL-1浓度。结论 MI缓解CCI诱导的神经病理性疼痛并降低小胶质细胞中IL-1浓度。
Objective To evaluated the effect of minocycline( MI) on chronic constriction injury( CCI) model of neuropathic pain in rat and assessed IL-1 concentration from cultured microglia cells. Methods Male SD rat were divided into three different groups: Sham + vehicle,CCI + vehicle,and CCI + MI. MI was injected one hour before surgery and continued daily to day 14 post ligation. Von Frey filaments and acetone were used for mechanical allodynia and cold allodynia,respectively. Experiments were performed on day 0 and days 1,3,5,7,10,and 14 post-injury. At day 14,rats were killed and microglia from lumbar part of the spinal cord were isolated and cultured in DMEM media,respectively. Interleukin-1( IL-1) concentration was evaluated in cell culture supernatant after 24 h. Results MI attenuated pain behavior,and a decrease in IL-1 concentration was observed in immune cells compared to CCI vehicle-treated rats. Conclusion MI reduced pain behavior and decreased IL-1 concentration in microglial cells.
