盐酸氨溴索对慢性哮喘大鼠气道炎症及杯状细胞增生的影响 被引量:16

摘要 目的 :观察药物盐酸氨溴索对慢性哮喘大鼠气道炎症及杯状细胞增生 (GCH)的影响 ,探讨在哮喘防治中的作用 .方法 :用卵白蛋白致敏、激发建立大鼠哮喘模型 ,5 4只Wistar大鼠随机分为 3组 :正常对照组、哮喘组、盐酸氨溴索治疗组(简称治疗组 ) ,每组 18只 .激发哮喘后测定气道内压的变化 ;支气管肺泡灌洗液 (BALF)内细胞计数、分类计数 ,HE染色及AB PAS染色观察肺组织病理改变及杯状细胞增生状况 .结果 :治疗组气道内压增加的百分数明显低于哮喘组 (P<0 .0 1) ;治疗组BALF细胞总数及炎症细胞数明显低于哮喘组 (P <0 .0 1) ;哮喘组大鼠肺组织病理可见明显支气管收缩征象及其周围炎症细胞浸润 ,AB PAS染色显示哮喘组大鼠气道杯状细胞广泛及显著增生 ,并可见气道粘液潴留 ;治疗组大鼠气道炎症及GCH较哮喘组均明显减轻 ,GCH仅存在于大气道 ,未见粘液潴留 .结论 :盐酸氨溴索可抑制哮喘大鼠气道GCH ,改善哮喘气道炎症 。 AIM: To evaluate the protective effect of ambroxol in asthma, we observed the effect of ambroxol on airway inflammation and airway goblet cell hyperplasia (GCH) of chronic asthmatic rat. METHODS: Rat asthma models were established with the ovalbumin sensitization and provocation method. Fifty four Wistar rats were divided into three groups randomly: ① Normal control group ( n =18); ② blank asthma group ( n =18); ③ ambroxol group ( n =18). Sensitized rats were treated with ambroxol daily (10 mg·kg -1 ·d -1 ) during the period of allergen sensitization and provocation. The change in intratracheal pressure of sensitized rats was recorded. The total and differential white blood cell counting of bronchial alveolar lavage fluid (BALF) was calculated. Lung tissue sections were stained with hematoxylin and eosin for general morphology and Alician Blue Periodic Acid Schiff (AB PAS)for identification of goblet cells. The pathologic changes were observed under an optical microscope. RESULTS: Percentage of increased intratracheal pressure in ambroxol group was significantly lower than that in the blank asthma group ( P < 0.01). The total cell number was significantly increased in the asthma group than in the control group ( P <0.01). The total cell number and the number of eosinophils and neutrophils were significantly decreased in the ambroxol group than in the blank asthma group ( P <0.01). The bronchoconstriction and inflammatory cells infiltration surrounding bronchi in the asthma group were observed. A marked and extensive airway GCH and mucus retention in airway lumen in the blank asthma group were also observed. The airway inflammation and GCH were significantly alleviated and no mucus retention in the airway lumen was observed in Ambroxol group. Mildly GCH was restricted in the larger airways. CONCLUSION: Ambroxol can alleviate the airway inflammation and suppress GCH in chronic asthmatic rats. Ambroxol is valuable in the prophylaxis and treatment of asthma.
