两种小鼠异位心脏移植模型的复制及比较 被引量:4

Replication and comparison of mouse celiac and cervical heterotopic heart transplantation

摘要 目的 :研究两种小鼠心脏移植模型的优缺点 ,为不同实验中选择术式提供依据。 方法 :应用放大 16倍的显微镜和显微外科手术器械 ,单人操作完成同品系或者同种异体小鼠腹部和颈部异位心脏移植两种术式 ,比较手术成功率、手术时间以及移植心脏存活时间。 结果 :腹部和颈部异位心脏移植手术成功率分别为 85 %和 6 0 % ,前者失败原因主要为出血 ,后者主要为吻合口狭窄。两种术式同种异体移植心脏存活时间无明显差异〔(7.6±0 .9)天vs(7.5± 1.0 )天 ,P >0 .0 5〕 ,二者均明显短于同系移植心脏存活时间 (平均 >10 0天 ) ;两种术式手术时间存在明显差异 ,颈部异位心脏移植术的受者准备、动脉和静脉吻合以及总手术时间上均比腹部异位心脏移植短。结论 :应根据实验需要选择 ,如注重手术成功率时 ,应选择腹部异位心脏移植 ;注重缩短手术时间时则应选用颈部异位心脏移植。 Objectives: To study the advantage and disadvantage of two types of mouse heart transplantation, so as to instruct the selection of mouse rejection model of heart transplantation. Methods:All operations of mouse celiac or cervical heterotopic heart transplantation between iso strain or hetero strains were performed by one surgeon using common surgical microscope (max amplifying power is 16) and microsurgery tools. Comparison was made on the operation success rate, operation time and allografts survival time. Results:Success rates of mouse celiac and cervical heterotopic heart transplantation were 85% and 60%, respectively. failure in the former group were mainly due to bleeding, but stoma constriction was the most common cause of failure in the latter group. The survival times of the allografts in the two groups were not different 〔(7.6±0.9)days vs (7.5±1.0)days, P >0.05〕, but they were both shorter than that of iso strain allografts, which was longer than 100 days. However, recipients preparation time, artery and vein anastomosis time, total operating time of cervical heart transplantation were significantly shorter than celiac transplantation. Conclusions: The selection of mouse heterotopic heart transplantation under common conditions depends on the requirement of research. When success rate is emphysized, celiac heart transplantation should be considered. But when we need short operating time, cervical heart transplantation should be chosen.
