摘要
目的制备氧化苦参碱磷脂复合物并对其进行表征,初步考察其体外抗乙肝病毒作用。方法以综合评分法为评价标准,通过正交试验优化氧化苦参碱磷脂复合物的制备工艺,并采用紫外分光光度法、X-射线衍射分析法和红外分光光度法对其进行表征,利用酶联免疫法考察其体外抗乙肝病毒作用。结果氧化苦参碱磷脂复合物的最优制备工艺为:药脂比1∶1、反应温度40℃,主药浓度为20 mg·mL-1。体外抗HBeAg试验结果表明,氧化苦参碱磷脂复合物中剂量对HBeAg有一定的抑制作用(P<0.05)。结论确定了氧化苦参碱磷脂复合物的制备工艺,证明氧化苦参碱与大豆磷脂形成了磷脂复合物。氧化苦参碱磷脂复合物对HBeAg有抑制作用。
Objective The oxymatrine phospholipid complex was prepared and characterized,and its in vitro anti-HBV effect was preliminarily investigated.Methods Using the comprehensive scoring method as the evaluation standard,the preparation process of oxymatrine phospholipid complex was optimized by orthogonal test,and characterized by ultraviolet spectrophotometry,X-ray diffraction analysis and infrared spectrophotometry.The in vitro anti-HBV effect was examined by Enzyme-linked immunosorbent assay.Results The optimal preparation process of oxymatrine phospholipid complex are as follows:the ratio of drug to lipid is 1∶1,the reaction temperature is 40℃,and the concentration of main drug is 20 mg·mL-1.The results of in vitro anti-HBeAg test showed that the medium dose of oxymatrine phospholipid complex had a certain inhibitory effect on HBeAg(P<0.05).Conclusion The preparation process of the oxymatrine phospholipid complex was determined,and it was confirmed that the oxymatrine and the soybean phospholipid formed a phospholipid complex.The oxymatrine phospholipid complex inhibited HBeAg in vitro.
