摘要
目的 探讨人胰腺癌细胞对羟基喜树碱的敏感性。方法 将人胰腺癌细胞株 (P3,SW - 1990 )接种于 96孔板 ,分别加入不同浓度的羟基喜树碱 ,作用不同时间后采用噻唑蓝法 (MTT法 )测定其抗癌敏感性。结果 羟基喜树碱对胰腺癌细胞的抑制率随药物浓度增加和作用时间的延长而增高 ,单药最大抑制率为 5 4 .6 % ;羟基喜树碱与DDP联合后产生协同作用 ,抑制率最高可达 74 .8% ,与临床常用化疗方案FEM ( 5 -氟尿嘧啶、表阿霉素、丝裂霉素 )相比无显著性差异。结论 胰腺癌细胞对羟基喜树碱敏感 ,可进一步进行胰腺癌患者的临床实验。
Objective It is to study the sensitivity of human pancreatic carcinoma cell to HCPT. Methods Human pancreatic cancer cells lines (P3, SW-1990) were incubated on a 96-well plate with addition of various concentrations of HCPT. After a certain time for reaction, the antitumor sensitivities were detected with MTT method. Results HCPT treatment resulted in a time and dose-dependent growth inhibition in vitro of pancreatic tumor cells, which with a maximal tumor growth inhibition of 54.6%. Synergistic activity was observed for DDP in combination with HCPT and the maximal tumor growth inhibition was 74.8%. Compared with FEM (5-FU, epirubicin, mitomycin), the combination of HCPT with DDP was equally active. Conclusion HCPT is a promising antitumor agent against pancreatic tumor. With good prospect, the clinical study should be engaged in.
