摘要
目的 观察β1整合素反义寡核苷酸对高侵袭性人卵巢癌细胞系CAOV3粘附侵袭的抑制作用。方法 合成的β1整合素反义寡核苷酸 (asODN)用脂质体转染入CAOV3细胞中 ,通过RT PCR、流式细胞术、MTT法、聚碳酸酯膜侵袭试验测定 β1整合素mRNA和细胞表面β1整合素蛋白表达及癌细胞粘附力、侵袭力的变化。结果 asODN组β1整合素 / β actinmRNA辉度值比值为 (0 .4 4 9± 0 .0 2 9) ,与对照组为 (0 .6 0 8± 0 .0 10 )相比 ,显著下降 (P <0 .0 1) ;流式细胞术检测asODN组平均荧光指数为(2 .4 30± 0 .10 4 ) ,较对照组 (6 .15 6± 0 .0 77)明显下降 (P <0 .0 1) ;asODN组粘附力为 0 .0 86± 0 .0 0 6 ,较对照组 (0 .137± 0 .0 19)显著下降 (P <0 .0 1)。asODN组穿透聚碳酸酯膜的细胞数为 16± 2 ,较对照组 (32± 4 )亦有显著差别 (P <0 .0 1)。而随机序列寡核苷酸 (rODN)组上述各指标较对照组无明显变化。结论 CAOV3细胞 β1整合素的表达与粘附侵袭能力密切相关 ,asODN能有效抑制CAOV3细胞 β1整合素基因的表达 ,从而抑制蛋白质合成 ,降低其粘附侵袭性。
ObjectiveTo observe the effect of integrin-β_ 1 antisense oligonucleotide inhibits the adhesion and invasion of the highly invasive ovarian cancer cell line CAOV3. MethodsIntegrin-β_ 1 antisense oligonucleotides were transfected into CAOV3 by liposome. The expression of integrin-β_ 1 mRNA and protein were determined by RT-PCR and FCM respectively.The changes of adhesion and invasion were measured by MTT and polycarbonate filters incorporated in modified Boyden chambers. ResultsIntegrin-β_ 1 /β-actin mRNA relative quantitative ratio of asODN group (0.449±0.029) was obviously lower than that of control group (0.608±0.010) (P<0.01). The mean fluorescent indexof asODN group (2.430±0.104) detected by FCM was evidently decreased than that of control group(6.156±0.077) (P<0.01). The adhesive ability of asODN group (0.086±0.006)obviously declined in compare with control group(0.137±0.019)(P<0.01). There was marked difference between asODN group(16±2) and control group(32±4) on the cell number that infiltrated the polycarbonate membrane(P<0.01). But there was no difference between the above-mentioned indexes of rODN group and that of control group. ConclusionsThe expression of integrin-β_ 1 in CAOV3 was related to the ability of adhesion and invasion.The asODN could effectively inhibit the expression of integrin-β_ 1 gene,thus inhibited the protein synthesis,and decreased the ability of adhesion and invasion.
