热化疗诱导荷瘤鼠肿瘤细胞凋亡的研究 被引量:2

摘要 目的 :探讨热化疗诱导荷瘤鼠肿瘤细胞凋亡及相关机制。方法 :Balb c纯系小白鼠 4 0只 ,随机分为 4组。于右后足底部皮下注入S180腹水型瘤细胞每只 2× 10 8mL-1。 4 8h后 ,分别给予不治疗、平阳霉素治疗、高温治疗及高温联合平阳霉素治疗 2次。切取肿瘤标本 ,TUNEL染色标记凋亡细胞 ,免疫组化S P法染色检测bcl 2和Bax蛋白表达 ,流式细胞仪检测细胞凋亡率。结果 :热化疗组癌细胞凋亡明显增多 ,凋亡率明显升高 ,bcl 2蛋白表达明显降低 ,Bax蛋白表达明显升高。结论 :热化疗通过调节凋亡相关基因bcl 2和Bax的表达诱导肿瘤细胞的凋亡。 Objective To study the apoptosis induced by thermochemotherapy and its mechanism in S180 cells in vivo.Methods Forty Balb/c mice were divided into four groups randomly,those were non-treatment (NT group),pingyangmycin treatment (P group),hyperthermia treatment (H group) and pingyangmycin combined with hyperthermia treatment (PH group).Forty eight hours after implanted S180 cells in right lower foot of the mice subcutaneously,the treatments were performed for twice except NT group.Then tumor specimens were cutfor studing the tumor cell apoptosis with TUNEL technique,the bcl-2 and Bax protein expressions with immunohistochemistry and the tumor cell apoptotic rate with flow cytometry.Results Thermochemotherapy enhanced tumor cell apoptosis.It significantly increased the apoptosis rate in the HP group compared with the other groups,P<0.01.It regulated the expression of bcl-2 to low level and Bax to high level.Conclusion Thermochemotherapy may regulate the expressions of bcl-2 and Bax proteins and then induce the apoptosis in S180 cells.
