摘要
目的 研究血管紧张素Ⅱ对大鼠肝星形细胞系HSC-T6凋亡的影响。方法 采用免疫细胞化学法检测血管紧张素Ⅱ的Ⅰ型受体表达;采用流式细胞仪技术和Hoechest 33342直接染色法观察血管紧张素Ⅱ对HSC-T6凋亡的影响;采用半定量RTPCR法检测凋亡相关基因Bcl-2和Bax表达的变化。结果 无血清培养的HSC-T6自发凋亡率分别为(2.99±0.98)%(流式细胞仪检测)和(5.69±1.52)%(Hoechest 33342直接染色法),血管紧张素Ⅱ能剂量依赖性的抑制其凋亡。与对照组比较,凋亡率具有显著差异(P<0.05或P<0.01);血管紧张素Ⅱ还能上调Bcl-2/Bax比值。结论 血管紧张素Ⅱ抑制HSC-T6凋亡是其参与肝纤维化的机制之一,此作用是通过上调Bcl-2/Bax比值实现的。
Objective This study is designed to observe the effect of angiotensin Ⅱ on apoptosis of the immortalized rat hepatic stellate cell line - HSC-T6. Methods The expression of angiotensin Ⅱ receptor type Ⅰ was detected by immunohistochemical techniques; apoptosis of HSC-T6 was determined by flow cytometry and Hoechest 33342 staining. Semi-quantity RT-PCR techniques were used to determine changes of Bcl-2 and Bax expression. Results The apoptosis rate of HSC-T6 cultured in serum free medium was (2.99±0.98)% (by flow cytometry) and (5.69±1.52)% (by Hoechest 33342 staining) , respectively. Angiotensin Ⅱ could inhibit HSC-T6 apoptosis in a dose-dependent way. Angiotensin Ⅱ also could increase the Bcl-2/Bax ratio. Conclusion Inhibition apoptosis of the hepatic stellate cell was one of the way of angiotensin Ⅱ to accelerate liver fibrosis. Angiotensin II inhibited apoptosis through increasing the Bcl-2 /Bax ratio.
