摘要
应用所构建的不同脊髓灰质炎病毒-痘苗病毒(下简称PV-VV)重组体,探讨脊髓灰质炎病毒(下简称PV)2A蛋白在重组病毒中的作用。所得到的四个以不同PV基因片段插入痘苗病毒(下简称VV)TK区而形成的重组病毒均有PV抗原的表达,并可诱导抗体产生,但此表达水平明显受到2A蛋白的影响。同时,重组病毒对不同细胞的适应性亦有变化。对此,本文进行了初步的探讨。
For studying the function of Poliovirus 2A proteinaes in Polio-Vaccinarecombinant virus, We constructed the various chimeric virus of Polio-Vac-cinia, and obtained 4 Polio--Vaccinia recombinant viruses which contain var-ious fragments of Polio virus I type mahoney strain gene. These fragmentswere inserted into the TK region of vaccinia gene. The antigen of Polioviruswas expressed in these 4 recombinant viruses, the antibody against Poliovirusl type was induced in the rabbit infected by these chimeric viruses. But,the expressive level of the Poliovirus I type antigen was influenced obviouslywith the 2A proteinase. The result about the suitable growth of recombinantvirus in various cell lines showed that 2A proteinase did not performed enti-rely its role that showed in Poliovirus in cell.
