摘要
本文以鸡冠为实验动物模型,通过大体、光学显微镜和透射电镜观察了PDT对真皮浅层毛细血管网和上皮组织的作用,发现PDT对真皮浅层毛细血管网有高度的选择性破坏作用,可使血管内皮细胞变性坏死,管壁破坏,但不损伤上皮组织,这种选择性破坏微血管网的作用与HpD给药量和激光能量有关。本文就PDT选择性破坏微血管网的影响因素进行了实验研究。
In order to improve the therapeutic results of port wine stains (PWS), an experimental study using chicken comb as an animal model of PWS treated by pholodynamic therapy (PDT) has been carried out to explore the possibility of selective destruction of the malformative vasculature in PWS. The influence of the following factors were investigated by gross inspection, light and electron microscopy: HpD dosage (l^20 mg/kg), intervals between HpD injection and argon laser irradiation (0~240 min), power density (50～150 mW/cm^2) and energy density (15～180 J/cm^2). Particular attention was paid to the gross color change of the comb, the morphology of endothelial cells of capillaries and the overlying epidermis of the comb at various post-irradiation intervals up to 4 weeks. The results showed that selective destruction of microvasculature in the superficial part of dermis could be achieved by PDT while the overlying epidermis remained normal. The HpD dosage and the light energy densities were found to be positively related to the degree of selective destruction. By the same HpD dose and light energy, the most prominent microvascular damage occurred in the animals irradiated at 0～30 min after HpD injection. It was demonstrated that the endothelial cells were most susceptible to photodynamic action. The possibility of selective destruction of abnormal vasculature by PDT may be of interest in clinical treatment of port wine stains.
