摘要
目的 :研究五倍子提取物对内毒素 (LPS)抑制人牙周膜成纤维细胞 (PDLCs)活性及对LPS损伤PDLCs超微结构的影响 ,并初步探讨其可能的作用机理。方法 :应用细胞培养技术、3 H -TdR掺入法以及透射电子显微镜观测五倍子提取物对LPS抑制PDLCs活性及对LPS损伤PDLCs超微结构的影响。结果 :LPS在浓度为 10 0mg/L时明显抑制细胞生长 ,加入五倍子提取物后 ,细胞活性明显增强 (P <0 .0 5 ) ;LPS (10 0mg/L)对PDLCs各超微结构均有不同程度的损伤 ,特别是线粒体损伤严重。加入五倍子提取物 (10mg/L)后 ,超微结构损伤减轻 ,细胞器结构基本恢复正常。结论 :五倍子提取物对LPS抑制PDLCs活性及对LPS损伤PDLCs超微结构具保护作用 。
AIM:To observe the effects of the Chinese nutgall on LPS which lead to the decreases of activities of PDLCs and the impairments of ultrastructures of PDLCs, and try to study the mechanism of the effects.METHODS:The cell culture technique, 3H-TdR and transmission electron microscope (TEM) methods were used to observe the effects of the Chinese nutgall on LPS.RESULTS:At the concentration of 100 mg/L,LPS can inhibit the activities of PDLCs remarkably and activities of PDLCs could greatly improved after adding the Chinese nutgalls. Furthermore, LPS had significant impairmental effects on ultrastructure of PDLCs in 100 mg/L, especially to the mitochondria. After adding the Chinese nutgalls, the impairments of ultrastructures of PDLCs were decreased. CONCLUSION:The Chinese nutgalls have the protective effects on LPS.The mechanism of the effects may be related to the discomposure of LPS and the protection of the Chinese nutgall going into the interior of PDLCs.
