摘要
将番木瓜(Carica papaya L.)受精胚珠接种于MS+IAA1mg/L+BA2mg/L+GA_3 2mg/L+S(蔗糖)3%和1/2MS+谷胺酰胺400Mg/L+CW(椰子汁)20%+S6%的培养基中,诱导愈伤组织,愈伤组织继代培养于MS+CW 2%+S6%+2.4-D 2mg/L的培养基上。进行了几种激素配比和基本培养基(MS和改良MS)对诱导胚性愈伤组织、体胚发生和植物再生培养基的比较实验。结果表明:在改良MS+硫酸腺嘌呤160mg/L+NAA 1mg/L+BA 0.5mg/L+GA_3 1mg/L+S4%培养基中获得理想胚性愈伤组织和体胚,在改良MS+硫酸腺嘌呤160mg/L+NAA 1mg/L+KT 0.5mg/L+GA_3 1mg/L+S4%产生正常具三裂片和根系的小植株。进行番木瓜体胚包埋、制成人工种子,实验获得小苗,并移栽成功。
Carica papaya L. ovules calli was induced upon MS (MurashigeSkoog 1962) medium containing 1mg/L IAA, 2mg/L, 2mg/L GA_3 ann 3% sucrose, and 1/2 MS medium containing 400mg/L glutainine, 20% filter-sterilizen concnut milk (CM) and 6% sucrose. The calli was subcultured on MS medium supplemented with 20% CM, 6% sucrose and 2mg/L 2. 4-D. High-frequency embryonic calli and som-atic embryogenesis accurred on modified MS medium containing 160mg/L Adenins-ulfat Adenine Sulfate, 1mg/L NAA, 0. 5mg/L KT, 1mg/L GA_3 and 4% sucrose.Normal plantlets were obtained. Carica papaya L. artificial seeds had been formed and already germinated. Plants have been successfully established in the soil.
