摘要
目的 :将人肿瘤坏死因子 α(TNF α)呈现于噬菌体表面 ,为研制TNF α疫苗奠定基础 .方法 :利用噬菌体表面呈现技术 ,在噬菌体表面表达人TNF α .结果 :重组噬菌体颗粒具有TNF α的免疫反应性 ,且不具有人TNF α的细胞杀伤活性 .结论 :人TNF α在噬菌体表面得以呈现 ,呈现TNF α的噬菌体颗粒作为TNF α疫苗的成分不会引起TNF
AIM: To display human TNF-α on the phage surface for the development of TNF-α vaccine. METHODS: The technique of phage surface display was used to display human TNF-α on the phage surface. RESULTS: Particles of recombinant phage-TNF-α had immunoreaction of TNF-α. The phage displayed human TNF-α did not possess the cytotoxicity that human TNF-α has. CONCLUSION: Human TNF-α has been successfully displayed on the phage surface and the recombinant phage-TNF-α, which will be used as vaccine component of TNF-α, does not have any side effects caused by TNF-α.
