摘要
根据自己提取植物基因组DNA的实际经验,改进了Clark利用CTAB提取植物基因组DNA的方法.在试验过程中发现,通过在研磨材料时加入液氮、用剪去端部的吸头转移含有DNA的溶液,并且将加入RNaseA消化RNA的步骤改在最后进行等一系列改进,可以获得高质量的DNA.本文同时就主要提取步骤进行了分析.
The CTAB method of extracting plant genomic DNA of Clark's was improved according to the experiences of ourselves. Plant materials were ground in liquid nitrogen. DNA solution was transferred with pipettetip without top. The digestion step of RNase A was carried out at the last step. High-quality DNA could be obtained on a series of these changes. In addition, major procedures of the CTAB method of extracting plant DNA of Clark's were discussed.
