摘要
对肠道病毒 71型 (enterovirus 71 ,EV71 )中国 (深圳 )分离株SHZH0 3进行了全基因组 (未包括多聚腺苷尾 )74 0 6个碱基的核苷酸序列测定。结果表明 ,SHZH0 3株与其它肠道病毒 71型毒株相比 ,在编码区没有核苷酸的缺失和插入 ,其 5′UTR和 3′UTR区的长度和序列有一定的差异。核苷酸同源性比较结果表明 ,在P1区SHZH0 3株与SHZH98株、中国台湾流行株 (TW 2 0 86、TW 2 2 72 )的同源性较高 (分别为 92 .5 % ,90 .1 %和 87.9% ) ,与新加坡流行株SIN5 6 6 6、SIN5 86 5及标准株MS、BrCr的同源性则在 81 %左右 ,而与CoxsackievirusA1 6 (Cox .A1 6 )的同源性最低 (6 3.6 % )。氨基酸同源性比较结果表明 ,在P1区SHZH0 3株与Cox .A1 6的同源性最低 ,但在P2和P3区SHZH0 3株与Cox .A1 6的同源性最高。P1区的遗传进化分析表明 ,SHZH0 3株和中国台湾 1 998年流行的EV71毒株的亲缘关系较近 ,属于同一型 (genogroup) ,而与标准株BrCr和MS的亲缘关系较远。
Eight overlapping clones covering the whole viral genome were obtained by RT PCR and then the sequences were determined by Sanger dideoxg mediated chain termination method.The results showed that the full length of enterovirus 71(EV71)SHZH03 genome(not including Poly A tail)is 7*!406bp.No insertion or deletion is detected in the coding region.There are several insertions and deletions in 5' and 3'UTR regions.In P1 region,SHZH03 strain shares high homology with SHZH98 strain and Taiwan predominant strains(TW2086,TW2272)but low homology with Cox.A16.In P2 and P3 regions,SHZH03 strain shares higher nucleotide homology with Cox.A16(80.7% and 82.6%)than that with Taiwan strains(TW2086,TW2272)and the highest amino acid homology with Cox.A16(98.3% and 96.8%).The phylogenetic analysis based on P1 region demonstrates that SHZH03 strain has the nearest genetic relationship with SHZH98 and Taiwan predominant strains(isolated in 1998).The results allow us to deduce a conclusion that SHZH03 may belong to the same genogroup with Taiwan predominant strains.
