BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor prognosis.Despite its clinical severity,the genetic alte...BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor prognosis.Despite its clinical severity,the genetic alterations,mechanisms,and signaling pathways underlying gallbladder NEC remain unclear.CASE SUMMARY This case study presents a rare instance of primary gallbladder NEC in a 73-year-old female patient,who underwent a radical cholecystectomy with hepatic hilar lymphadenectomy and resection of liver segments IV-B and V.Targeted gene sequencing and bioinformatics analysis tools,including STRING,GeneMANIA,Metascape,TRRUST,Sangerbox,cBioPortal and GSCA,were used to analyze the biological functions and features of mutated genes in gallbladder NEC.Twelve mutations(APC,ARID2,IFNA6,KEAP1,RB1,SMAD4,TP53,BTK,GATA1,GNAS,and PRDM3)were identified,and the tumor mutation burden was determined to be 9.52 muts/Mb via targeted gene sequencing.A protein-protein interaction network showed significant interactions among the twelve mutated genes.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were used to assess mutation functions and pathways.The results revealed 40 tumor-related pathways.A key regulatory factor for gallbladder NEC-related genes was identified,and its biological functions and features were compared with those of gallbladder carcinoma.CONCLUSION Gallbladder NEC requires standardized treatment.Comparisons with other gallbladder carcinomas revealed clinical phenotypes,molecular alterations,functional characteristics,and enriched pathways.展开更多
Background:The development of computer vision technology has enabled the use of markerless movement tracking for biomechanical analysis.Recent research has reported the feasibility of markerless systems in motion anal...Background:The development of computer vision technology has enabled the use of markerless movement tracking for biomechanical analysis.Recent research has reported the feasibility of markerless systems in motion analysis but has yet to fully explore their utility for capturing faster movements,such as running.Applied studies using markerless systems in clinical and sports settings are still lacking.Thus,the present study compared running biomechanics estimated by marker-based and markerless systems.Given running speed not only affects sports performance but is also associated with clinical injury prevention,diagnosis,and rehabilitation,we aimed to investigate the effects of speed on the comparison of estimated lower extremity joint moments and powers between markerless and marker-based technologies during treadmill running as a concurrent validating study.Methods:Kinematic data from marker-based/markerless technologies were collected,along with ground reaction force data,from 16 young adults running on an instrumented treadmill at 3 speeds:2.24 m/s,2.91 m/s,and 3.58 m/s(5.0 miles/h,6.5 miles/h,and 8.0 miles/h).Sagittal plane moments and powers of the hip,knee,and ankle were calculated by inverse dynamic methods.Time series analysis and statistical parametric mapping were used to determine system differences.Results:Compared to the marker-based system,the markerless system estimated increased lower extremity joint kinetics with faster speed during the swing phase in most cases.Conclusion:Despite the promising application of markerless technology in clinical settings,systematic markerless overestimation requires focused attention.Based on segment pose estimations,the centers of mass estimated by markerless technologies were farther away from the relevant distal joint centers,which led to greater joint moments and powers estimates by markerless vs.marker-based systems.The differences were amplified by running speed.展开更多

[Objectives]To compare the effects of traditional processing and fresh processing on the quality of Polygonatum odoratum decoction piece.[Methods]The effects of fresh processing and traditional processing on the quali...[Objectives]To compare the effects of traditional processing and fresh processing on the quality of Polygonatum odoratum decoction piece.[Methods]The effects of fresh processing and traditional processing on the quality of P.odoratum decoction piece were compared and analyzed with appearance characteristics,total ash content,extract content,total polysaccharides content,and total flavonoids content as the evaluation indexes.[Results]Fresh processing method in different production areas has different effects on P.odoratum decoction piece.P.odoratum was dried in oven of 50℃.When moisture content was 41.44%-59.67%,it was cut.After complete drying at 50℃,the moisture content of dried P.odoratum was 8.94%-9.60%,and ethanol-soluble extract content was 77.29%-78.20%,and water-soluble extract was 77.7%-78.14%.At this time,the appearance characteristics of section of P.odoratum decoction piece were better than that of traditional processing,which was yellowish white.The total polysaccharide content was higher than that of traditional processing,and the content of total flavonoids was statistically significant different from that of traditional processing.[Conclusions]The quality of P.odoratum decoction piece by fresh processing is better than that of the traditional processing,and it is feasible to use fresh processing.展开更多

Systematic review and analysis of the general information, postmortem examinations and assessments of chest injury had carried out from Feb. 2016 to Mar. 2018. <strong>Results:</strong> Average age of the victims was 52.2 years and the ratio of male to female deaths was 2.39:1. The proportion of motor-cyclists and pedestrians increased significantly. The overwhelming majority of accident vehicles were motorcycles and bicycles. Fractures of ribs and pulmonary contusion were the most common injuries. Craniocerebral and abdominal injuries were the most common associated injuries. <strong>Conclusion:</strong> Fractures of ribs and pulmonary contusion were the most common features of fatal road traffic injuries, often associated with vitreoretinal damage and serious multiple damages. These features reflect the characteristics of great violence in traffic accidents, which provides the evidence of identification of violent injuries.展开更多
China is one of most biodiverse countries in the world, containing at least 10% of all angiosperm species.Therefore, we should anticipate a diverse, pollinator fauna. China also has a long history of applied ethnobiol...China is one of most biodiverse countries in the world, containing at least 10% of all angiosperm species.Therefore, we should anticipate a diverse, pollinator fauna. China also has a long history of applied ethnobiology, including a sustainable agriculture based on apiculture and plant-pollinator interactions.However, the science of pollination ecology is a far younger sub-discipline in China, compared to in the West. Chinese studies in pollination ecology began in the 1970 s. For this review, we compiled a complete reference database(>600 publications) of pollination studies in China. Using this database, we identified and analyzed gaps and limitations in research on the pollination systems of native and naturalized species. Specifically, we asked the following questions: 1) What do we know about the pollination systems of native, Chinese species? 2) How does Chinese pollination ecology compare with the development of pollination research abroad and which aspects of research should be pursued by Chinese anthecologists in the near future? 3) What research on pollination in China will advance our understanding and contribute to our ongoing analyses of endemism and conservation? Subsequently, we segregated and identified prospective lines of future research that are unique to China and can only be done in China. Objective:Gemcitabine plus nab-paclitaxel(GnP)is the standard first-line therapy for advanced pancreatic ductal adenocarcinoma(PDAC).S-1,an oral fluoropyrimidine derivative,as compared with gemcitabine,is non-inferior...Objective:Gemcitabine plus nab-paclitaxel(GnP)is the standard first-line therapy for advanced pancreatic ductal adenocarcinoma(PDAC).S-1,an oral fluoropyrimidine derivative,as compared with gemcitabine,is non-inferior in terms of overall survival(OS)and is associated with lower hematologic toxicity.Accordingly,S-1 is a convenient oral alternative treatment for advanced PDAC.This study was aimed at comparing the efficacy and safety of gemcitabine plus S-1(GS)vs.GnP as first-line chemotherapy for advanced PDAC.Methods:Patients with advanced PDAC who received first-line GS or GnP at the Peking Union Medical College Hospital between March 2011 and November 2022 were evaluated.Results:A total of 300 patients were assessed,of whom 84 received GS and 216 received GnP.The chemotherapy completion rate was higher with GS than GnP(50.0%vs.30.3%,P=0.0028).The objective response rate(ORR)was slightly higher(14.3%vs.9.7%,P=0.35),and the median OS was significantly longer(17.9 months vs.13.3 months,P=0.0078),in the GS group than the GnP group.However,the median progression-free survival(PFS)did not significantly differ between groups.Leukopenia risk was significantly lower in the GS group than the GnP group(14.9%vs.28.1%,P=0.049).Conclusions:As first-line chemotherapy for advanced PDAC,the GS regimen led to a significantly longer OS than the GnP regimen.The PFS,ORR,and incidence of severe adverse events were comparable between the GS and GnP groups.展开更多
Natural and geological environmental conditions have an important impact on the planning and construction of sponge cities.This paper analyzes geological factors that influence the usage of natural sponge bodies,takin...Natural and geological environmental conditions have an important impact on the planning and construction of sponge cities.This paper analyzes geological factors that influence the usage of natural sponge bodies,taking the Shuanghe lake district of Zhengzhou airport zone as an example.An evaluation system with seven factors has been established and the weights of these factors are determined using the analytic hierarchy process(AHP)method.Overlay analysis is then carried out on all factors using GIS to evaluate the geological suitability of the construction of the sponge city.The results show that geologically suitable area for city construction in Shuanghe lake district accounts for 12.3%,relatively suitable area accounts for 76.1%,and relatively unsuitable area accounts for 11.6%.For suitable and relatively suitable areas,we should make full use of the advantages of surface infiltration,vadose zone transportation and aquifer storage to build a sponge city infrastructure with geological engineering as the main component,supplemented by engineering measures such as surface water storage and drainage,and jointly establish a sustainable urban hydrological cycle.For less suitable areas,artificial rain and flood control works,such as roof garden,should be considered.The findings of this paper can serve as an important reference for sponge city planning and construction not only in the research area but also in other regions with similar geological conditions.展开更多

The objective of this study was to analyze the cause of typical serious medication errors in some first-class provincial hospital to prevent medication error and improve rational use of drug and ensure patients administration security. Medication errors were collected between September 1, 2012 and August 31, 2013. 332 cases’ medication errors were categorized as harmful or not according to NCC MERP’s Index and other indexes such as type of error and causes of error. We focus on analyzing the 3 serious cases (0.9%, n = 3) among the all MEs that may have contributed to or resulted in temporary harm to the patient and required intervention, including the process and the cause and the background of the errors. The case analysis reminds us to pay more attention to prevent medication errors. The preventive medication error’s measures are explored through the analysis of the causes, such as forcing functions and constraints, automation and standardization, double-checking systems, rules and policies, information and more careful working. Conclusion: This analytical study demonstrates that medication error is an objective existent in hospital. It is especially important to ensure patient medication safety for reporting and analyzing medication error in order to explore measures preventing medication error.展开更多

Complement is...The innate immune response is a complex process involving multiple pathogen-recognition receptors, including toll-like receptors (TLRs) and nucleotide-binding oligomerization domain (NOD)-like receptors. Complement is also a critical component of innate immunity. While complement is known to interact with TLR-mediated signals, the interactions between NOD-like receptors and complement are not well understood. Here we report a synergistic interaction between C5a and Nod2 signaling in RAW 264.7 marophages. Long-term treatment with muramyl dipeptide (MDP), a NOD2 ligand, enhanced C5a-mediated expression of chemokine mRNAs in RAW 264.7 cells. This response was dependent on NOD2 expression and was associated with a decrease in expression of C5L2, a receptor for C5a which acts as a negative modulator of C5a receptor (C5aR) activity. MDP amplified C5a-mediated phosphorylation of p38 MAPK. Treatment of RAW264.7 cells with an inhibitor of p38 attenuated the synergistic effects of C5aon MDP-primed cells on MIP-2, but not MCP-1, mRNA. In contrast, inhibition of AKT prevented C5a stimulation of MCP-1, but not MIP-2, mRNA, in MDP-primed cells. Taken together, these data demonstrated a synergistic interaction between C5a and NOD2 in the regulation of chemokine expression in macrophages, associated with a down-regulation of C5L2, a negative regulator of C5a receptor activity.展开更多

BACKGROUND Colorectal cancer(CRC)is an extremely malignant tumor with a high mortality rate.Little is known about the mechanism by which forkhead Box q1(FOXQ1)causes CRC invasion and metastasis through the epidermal g...BACKGROUND Colorectal cancer(CRC)is an extremely malignant tumor with a high mortality rate.Little is known about the mechanism by which forkhead Box q1(FOXQ1)causes CRC invasion and metastasis through the epidermal growth factor receptor(EGFR)pathway.AIM To illuminate the mechanism by which FOXQ1 promotes the invasion and metastasis of CRC by activating the heparin binding epidermal growth factor(HB-EGF)/EGFR pathway.METHODS We investigated the differential expression and prognosis of FOXQ1 and HB-EGF in CRC using the Gene Expression Profiling Interactive Analysis(GEPIA)website(http://gepia.cancer-pku.cn/index.html).Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blotting were used to detect the expression of FOXQ1 and HB-EGF in cell lines and tissues,and we constructed a stable lowexpressing FOXQ1 cell line and verified it with the above method.The expression changes of membrane-bound HB-EGF(proHB-EGF)and soluble HB-EGF(sHB-EGF)in the lowexpressing FOXQ1 cell line were detected by flow cytometry and ELISA.Western blotting was used to detect changes in the expression levels of HB-EGF and EGFR pathway-related downstream genes when exogenous recombinant human HB-EGF was added to FOXQ1 knockdown cells.Proliferation experiments,transwell migration experiments,and scratch experiments were carried out to determine the mechanism by which FOXQ1 activates the EGFR signaling pathway through HB-EGF,and then to evaluate the clinical relevance of FOXQ1 and HB-EGF.RESULTS GEPIA showed that the expression of FOXQ1 in CRC tissues was relatively high and was related to a lower overall survival rate.PCR array results showed that FOXQ1 is related to the HB-EGF and EGFR pathways.Knockdown of FOXQ1 suppressed the expression of HB-EGF,and led to a decrease in EGFR and its downstream genes AKT,RAF,KRAS expression levels.After knockdown of FOXQ1 in CRC cell lines,cell proliferation,migration and invasion were attenuated.Adding HB-EGF restored the migration and invasion ability of CRC,but not the cell proliferation ability.Kaplan–Meier survival analysis results showed that the combination of FOXQ1 and HB-EGF may serve to predict CRC survival.CONCLUSION Based on these collective data,we propose that FOXQ1 promotes the invasion and metastasis of CRC via the HB-EGF/EGFR pathway.展开更多
Laterally-coupled ridge-waveguide distributed feedback lasers fabricated without epitaxial regrowth steps have the advantages of process simplification and low cost.We present a laterally coupled grating with slots.Th...Laterally-coupled ridge-waveguide distributed feedback lasers fabricated without epitaxial regrowth steps have the advantages of process simplification and low cost.We present a laterally coupled grating with slots.The slots etched between the ridge and grating area are designed to suppress the lateral diffusion of carriers and to reduce the influence of the aspect-ratio-dependent-etching effect on the grating morphology in the etching process.Moreover,the grating height in this structure can be decreased to lower the aspect ratio significantly,which is advantageous over the conventional laterally coupled ridge waveguide gratings.The effects of five main structural parameters on the coupling characteristics of gratings are studied by MODE Solutions.It is found that varying the lateral width of the grating can be used as an effective way to tune the coupling strength;narrow slots(100 nm and 300 nm)and wide ridge(2μm–4μm)promote the stability of grating coupling coefficient and device performance.It is important to note that the grating bottom should be fabricated precisely.The comparative study of carrier distribution and mode field distribution shows that the introduction of narrow slots can strengthen the competitive advantage and stability of the fundamental mode.展开更多
Tomographic perfusion imaging is a significant imaging modality for stroke diagnosis.However,the low rotational speed of the C-arm(6–8 s per circle)is a challenge for applying perfusion imaging in C-arm cone beam com...Tomographic perfusion imaging is a significant imaging modality for stroke diagnosis.However,the low rotational speed of the C-arm(6–8 s per circle)is a challenge for applying perfusion imaging in C-arm cone beam computed tomography(CBCT).Traditional reconstruction methods cannot remove the artifacts caused by the slow rotational speed or acquire enough sample points to restore the time density curve(TDC).This paper presents a dynamic rollback reconstruction method for CBCT.The proposed method can improve the temporal resolution by increasing the sample points used for calculating the TDC.Combined with existing techniques,the algorithm allows slow-rotating scanners to be used for perfusion imaging purposes.In the experiments,the proposed method was compared with other dynamic reconstruction algorithms based on standard reconstruction and the temporal interpolation approach.The presented algorithm could improve the temporal resolution without increasing the X-ray exposure time or contrast agent.展开更多
Objective:To compare the effects of clinical application of chemiluminescence and real-time,fluorescence-based quantitative PCR in the detection Epstein-Barr virus(EBV).Methods:The data of chemiluminescence and real-t...Objective:To compare the effects of clinical application of chemiluminescence and real-time,fluorescence-based quantitative PCR in the detection Epstein-Barr virus(EBV).Methods:The data of chemiluminescence and real-time fluorescent quantitative PCR.fromipaEsfwo were suspected of being infectea w1tn rito1 roro January 2016 to January 2019 in our hospital were analyzed.The specific stage of EBV infection was analyzed,and the differences in results of the two detection methods were compared.Results:Chemiluminescence method was used to detect EBV infection during the active phase.The sensitivity of the chemiluminescence method was 76.7%(56/73)and the real-time quantitative PCRmethod was 90.4%(66/73).There was a statistical difference between the two detection methods(P<0.05).Conclusion:There was no statistical difference in positive predictive values between the chemiluminescence method and the real-time,fluorescence-based quantitative PCR method in the detection of EBV infection,but the sensitivity of chemiluminescence method is slightly lower than the real-time quantitative PCRmethod.It is noteworthy that chemiluminescence method is convenient and fast while the real-time,fluorescence-based quantitative PCR method is more accurate,which can provide a more accurate reference for clinical treatment.展开更多

文摘[Objectives]To compare the effects of traditional processing and fresh processing on the quality of Polygonatum odoratum decoction piece.[Methods]The effects of fresh processing and traditional processing on the quality of P.odoratum decoction piece were compared and analyzed with appearance characteristics,total ash content,extract content,total polysaccharides content,and total flavonoids content as the evaluation indexes.[Results]Fresh processing method in different production areas has different effects on P.odoratum decoction piece.P.odoratum was dried in oven of 50℃.When moisture content was 41.44%-59.67%,it was cut.After complete drying at 50℃,the moisture content of dried P.odoratum was 8.94%-9.60%,and ethanol-soluble extract content was 77.29%-78.20%,and water-soluble extract was 77.7%-78.14%.At this time,the appearance characteristics of section of P.odoratum decoction piece were better than that of traditional processing,which was yellowish white.The total polysaccharide content was higher than that of traditional processing,and the content of total flavonoids was statistically significant different from that of traditional processing.[Conclusions]The quality of P.odoratum decoction piece by fresh processing is better than that of the traditional processing,and it is feasible to use fresh processing.
文摘<strong>Objective:</strong> To explore the characteristics and mechanisms of serious injuries of chest caused by road traffic accidents. <strong>Methods:</strong> Totally 112 autopsy cases with chest injuries in the urban of Jingzhou road traffic accidents were collected. Systematic review and analysis of the general information, postmortem examinations and assessments of chest injury had carried out from Feb. 2016 to Mar. 2018. <strong>Results:</strong> Average age of the victims was 52.2 years and the ratio of male to female deaths was 2.39:1. The proportion of motor-cyclists and pedestrians increased significantly. The overwhelming majority of accident vehicles were motorcycles and bicycles. Fractures of ribs and pulmonary contusion were the most common injuries. Craniocerebral and abdominal injuries were the most common associated injuries. <strong>Conclusion:</strong> Fractures of ribs and pulmonary contusion were the most common features of fatal road traffic injuries, often associated with vitreoretinal damage and serious multiple damages. These features reflect the characteristics of great violence in traffic accidents, which provides the evidence of identification of violent injuries.

文摘China is one of most biodiverse countries in the world, containing at least 10% of all angiosperm species.Therefore, we should anticipate a diverse, pollinator fauna. China also has a long history of applied ethnobiology, including a sustainable agriculture based on apiculture and plant-pollinator interactions.However, the science of pollination ecology is a far younger sub-discipline in China, compared to in the West. Chinese studies in pollination ecology began in the 1970 s. For this review, we compiled a complete reference database(>600 publications) of pollination studies in China. Using this database, we identified and analyzed gaps and limitations in research on the pollination systems of native and naturalized species. Specifically, we asked the following questions: 1) What do we know about the pollination systems of native, Chinese species? 2) How does Chinese pollination ecology compare with the development of pollination research abroad and which aspects of research should be pursued by Chinese anthecologists in the near future? 3) What research on pollination in China will advance our understanding and contribute to our ongoing analyses of endemism and conservation? Subsequently, we segregated and identified prospective lines of future research that are unique to China and can only be done in China. This requires discussing priorities within a systematic approach.

文摘Biliary stenosis is a common complication after liver transplantation,and has an incidence rate ranging from4.7%to 12.5%based on our previous study.Three types of biliary stenosis(anastomotic stenosis,nonanastomotic peripheral stenosis and non-anastomotic central hilar stenosis)have been identified.We report the outcome of two patients with anastomotic stricture after liver transplantation who underwent successfulcutting balloon treatment.Case 1 was a 40-year-old male transplanted due to subacute fulminant hepatitis C.Case 2 was a 57-year-old male transplanted due to hepatitis B virus-related end-stage cirrhosis associated with hepatocellular carcinoma.Both patients had similar clinical scenarios:refractory anastomotic stenosis after orthotopic liver transplantation and failure of balloon dilation of the common bile duct to alleviate biliary stricture.

文摘Objective:Gemcitabine plus nab-paclitaxel(GnP)is the standard first-line therapy for advanced pancreatic ductal adenocarcinoma(PDAC).S-1,an oral fluoropyrimidine derivative,as compared with gemcitabine,is non-inferior in terms of overall survival(OS)and is associated with lower hematologic toxicity.Accordingly,S-1 is a convenient oral alternative treatment for advanced PDAC.This study was aimed at comparing the efficacy and safety of gemcitabine plus S-1(GS)vs.GnP as first-line chemotherapy for advanced PDAC.Methods:Patients with advanced PDAC who received first-line GS or GnP at the Peking Union Medical College Hospital between March 2011 and November 2022 were evaluated.Results:A total of 300 patients were assessed,of whom 84 received GS and 216 received GnP.The chemotherapy completion rate was higher with GS than GnP(50.0%vs.30.3%,P=0.0028).The objective response rate(ORR)was slightly higher(14.3%vs.9.7%,P=0.35),and the median OS was significantly longer(17.9 months vs.13.3 months,P=0.0078),in the GS group than the GnP group.However,the median progression-free survival(PFS)did not significantly differ between groups.Leukopenia risk was significantly lower in the GS group than the GnP group(14.9%vs.28.1%,P=0.049).Conclusions:As first-line chemotherapy for advanced PDAC,the GS regimen led to a significantly longer OS than the GnP regimen.The PFS,ORR,and incidence of severe adverse events were comparable between the GS and GnP groups.

文摘Natural and geological environmental conditions have an important impact on the planning and construction of sponge cities.This paper analyzes geological factors that influence the usage of natural sponge bodies,taking the Shuanghe lake district of Zhengzhou airport zone as an example.An evaluation system with seven factors has been established and the weights of these factors are determined using the analytic hierarchy process(AHP)method.Overlay analysis is then carried out on all factors using GIS to evaluate the geological suitability of the construction of the sponge city.The results show that geologically suitable area for city construction in Shuanghe lake district accounts for 12.3%,relatively suitable area accounts for 76.1%,and relatively unsuitable area accounts for 11.6%.For suitable and relatively suitable areas,we should make full use of the advantages of surface infiltration,vadose zone transportation and aquifer storage to build a sponge city infrastructure with geological engineering as the main component,supplemented by engineering measures such as surface water storage and drainage,and jointly establish a sustainable urban hydrological cycle.For less suitable areas,artificial rain and flood control works,such as roof garden,should be considered.The findings of this paper can serve as an important reference for sponge city planning and construction not only in the research area but also in other regions with similar geological conditions.
文摘Background: Medication errors are a common problem in Chinese hospital, with potentially harmful consequences for patients. The objective of this study was to analyze the cause of typical serious medication errors in some first-class provincial hospital to prevent medication error and improve rational use of drug and ensure patients administration security. Medication errors were collected between September 1, 2012 and August 31, 2013. 332 cases’ medication errors were categorized as harmful or not according to NCC MERP’s Index and other indexes such as type of error and causes of error. We focus on analyzing the 3 serious cases (0.9%, n = 3) among the all MEs that may have contributed to or resulted in temporary harm to the patient and required intervention, including the process and the cause and the background of the errors. The case analysis reminds us to pay more attention to prevent medication errors. The preventive medication error’s measures are explored through the analysis of the causes, such as forcing functions and constraints, automation and standardization, double-checking systems, rules and policies, information and more careful working. Conclusion: This analytical study demonstrates that medication error is an objective existent in hospital. It is especially important to ensure patient medication safety for reporting and analyzing medication error in order to explore measures preventing medication error.
文摘The innate immune response is a complex process involving multiple pathogen-recognition receptors, including toll-like receptors (TLRs) and nucleotide-binding oligomerization domain (NOD)-like receptors. Complement is also a critical component of innate immunity. While complement is known to interact with TLR-mediated signals, the interactions between NOD-like receptors and complement are not well understood. Here we report a synergistic interaction between C5a and Nod2 signaling in RAW 264.7 marophages. Long-term treatment with muramyl dipeptide (MDP), a NOD2 ligand, enhanced C5a-mediated expression of chemokine mRNAs in RAW 264.7 cells. This response was dependent on NOD2 expression and was associated with a decrease in expression of C5L2, a receptor for C5a which acts as a negative modulator of C5a receptor (C5aR) activity. MDP amplified C5a-mediated phosphorylation of p38 MAPK. Treatment of RAW264.7 cells with an inhibitor of p38 attenuated the synergistic effects of C5aon MDP-primed cells on MIP-2, but not MCP-1, mRNA. In contrast, inhibition of AKT prevented C5a stimulation of MCP-1, but not MIP-2, mRNA, in MDP-primed cells. Taken together, these data demonstrated a synergistic interaction between C5a and NOD2 in the regulation of chemokine expression in macrophages, associated with a down-regulation of C5L2, a negative regulator of C5a receptor activity.

文摘Carbon-based materials are typical and commercially active electrode for supercapacitors due to their advantages such as low cost, good stability and easy availability. In the light of energy storage, supercapacitors mechanism is classified into EDLCs (electrochemical double layer capacitors) and pseudocapacitors. Multidimensional carbon nanomaterials (active carbon, carbon nanotube, graphene, etc.), carbon-based composite and corresponding electrolyte are the critical and important factor in the eyes of researcher. In this minireview, we will discuss the storage mechanism and summarize recent developed novel carbon and carbon-based materials in supercapacitors. The techniques to design the novel nanostructure and high performance electrodematerials that facilitate charge transfer to achieve high energy and power densities will also be discussed.

文摘BACKGROUND Colorectal cancer(CRC)is an extremely malignant tumor with a high mortality rate.Little is known about the mechanism by which forkhead Box q1(FOXQ1)causes CRC invasion and metastasis through the epidermal growth factor receptor(EGFR)pathway.AIM To illuminate the mechanism by which FOXQ1 promotes the invasion and metastasis of CRC by activating the heparin binding epidermal growth factor(HB-EGF)/EGFR pathway.METHODS We investigated the differential expression and prognosis of FOXQ1 and HB-EGF in CRC using the Gene Expression Profiling Interactive Analysis(GEPIA)website(http://gepia.cancer-pku.cn/index.html).Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blotting were used to detect the expression of FOXQ1 and HB-EGF in cell lines and tissues,and we constructed a stable lowexpressing FOXQ1 cell line and verified it with the above method.The expression changes of membrane-bound HB-EGF(proHB-EGF)and soluble HB-EGF(sHB-EGF)in the lowexpressing FOXQ1 cell line were detected by flow cytometry and ELISA.Western blotting was used to detect changes in the expression levels of HB-EGF and EGFR pathway-related downstream genes when exogenous recombinant human HB-EGF was added to FOXQ1 knockdown cells.Proliferation experiments,transwell migration experiments,and scratch experiments were carried out to determine the mechanism by which FOXQ1 activates the EGFR signaling pathway through HB-EGF,and then to evaluate the clinical relevance of FOXQ1 and HB-EGF.RESULTS GEPIA showed that the expression of FOXQ1 in CRC tissues was relatively high and was related to a lower overall survival rate.PCR array results showed that FOXQ1 is related to the HB-EGF and EGFR pathways.Knockdown of FOXQ1 suppressed the expression of HB-EGF,and led to a decrease in EGFR and its downstream genes AKT,RAF,KRAS expression levels.After knockdown of FOXQ1 in CRC cell lines,cell proliferation,migration and invasion were attenuated.Adding HB-EGF restored the migration and invasion ability of CRC,but not the cell proliferation ability.Kaplan–Meier survival analysis results showed that the combination of FOXQ1 and HB-EGF may serve to predict CRC survival.CONCLUSION Based on these collective data,we propose that FOXQ1 promotes the invasion and metastasis of CRC via the HB-EGF/EGFR pathway.

文摘Laterally-coupled ridge-waveguide distributed feedback lasers fabricated without epitaxial regrowth steps have the advantages of process simplification and low cost.We present a laterally coupled grating with slots.The slots etched between the ridge and grating area are designed to suppress the lateral diffusion of carriers and to reduce the influence of the aspect-ratio-dependent-etching effect on the grating morphology in the etching process.Moreover,the grating height in this structure can be decreased to lower the aspect ratio significantly,which is advantageous over the conventional laterally coupled ridge waveguide gratings.The effects of five main structural parameters on the coupling characteristics of gratings are studied by MODE Solutions.It is found that varying the lateral width of the grating can be used as an effective way to tune the coupling strength;narrow slots(100 nm and 300 nm)and wide ridge(2μm–4μm)promote the stability of grating coupling coefficient and device performance.It is important to note that the grating bottom should be fabricated precisely.The comparative study of carrier distribution and mode field distribution shows that the introduction of narrow slots can strengthen the competitive advantage and stability of the fundamental mode.

文摘Tomographic perfusion imaging is a significant imaging modality for stroke diagnosis.However,the low rotational speed of the C-arm(6–8 s per circle)is a challenge for applying perfusion imaging in C-arm cone beam computed tomography(CBCT).Traditional reconstruction methods cannot remove the artifacts caused by the slow rotational speed or acquire enough sample points to restore the time density curve(TDC).This paper presents a dynamic rollback reconstruction method for CBCT.The proposed method can improve the temporal resolution by increasing the sample points used for calculating the TDC.Combined with existing techniques,the algorithm allows slow-rotating scanners to be used for perfusion imaging purposes.In the experiments,the proposed method was compared with other dynamic reconstruction algorithms based on standard reconstruction and the temporal interpolation approach.The presented algorithm could improve the temporal resolution without increasing the X-ray exposure time or contrast agent.
文摘Objective:To compare the effects of clinical application of chemiluminescence and real-time,fluorescence-based quantitative PCR in the detection Epstein-Barr virus(EBV).Methods:The data of chemiluminescence and real-time fluorescent quantitative PCR.fromipaEsfwo were suspected of being infectea w1tn rito1 roro January 2016 to January 2019 in our hospital were analyzed.The specific stage of EBV infection was analyzed,and the differences in results of the two detection methods were compared.Results:Chemiluminescence method was used to detect EBV infection during the active phase.The sensitivity of the chemiluminescence method was 76.7%(56/73)and the real-time quantitative PCRmethod was 90.4%(66/73).There was a statistical difference between the two detection methods(P<0.05).Conclusion:There was no statistical difference in positive predictive values between the chemiluminescence method and the real-time,fluorescence-based quantitative PCR method in the detection of EBV infection,but the sensitivity of chemiluminescence method is slightly lower than the real-time quantitative PCRmethod.It is noteworthy that chemiluminescence method is convenient and fast while the real-time,fluorescence-based quantitative PCR method is more accurate,which can provide a more accurate reference for clinical treatment.

文摘The root is an important organ for plants to obtain nutrients and water,and its phenotypic characteristics are closely related to its functions.Deep-learning-based high-throughput in situ root senescence feature extraction has not yet been published.In light of this,this paper suggests a technique based on the transformer neural network for retrieving cotton's in situ root senescence properties.High-resolution in situ root pictures with various levels of senescence are the main subject of the investigation.
