Acute myeloid leukemia(AML)is regarded as a stem cell disease.However,no one unique marker is expressed on leukemia stem cells(LSC)but not on leukemic blasts nor normal hematopoietic stem cells(HSC).CD34^(+)CD38^(-)with or without CD123 or CD44 subpopulations are immunophenotypically defined as putative LSC fractions in AML.Nevertheless,markers that can be effectively and simply held responsible for the intrinsical heterogeneity of LSC is still unclear.In the present study,we examined the frequency of three different LSC subtypes(CD34^(+)CD38^(-),CD34^(+)CD38^(-)CD123^(+),CD34^(+)CD38^(-)CD44^(+))in AML at diagnosis.We then validated their prognostic significance on the relevance of spectral features for diagnostic stratification,immune status,induction therapy response,treatment effect maintenance,and long^(-)term survival.In our findings,high proportions of the above three different LSC subtypes were all significantly characterized with low complete remission(CR)rate,high relapse/refractory rate,poor overall survival(OS),frequent FLT3^(-)ITD mutation,the high level of regulatory T cells(Treg)and monocytic myeloid^(-)derived suppressor cells(M^(-)MDSC).However,there was no significant statistical difference in all kinds of other clinical performance among the three different LSC groups.It was demonstrated that CD34^(+)CD38^(-)subpopulation without CD123 and CD44 might be held responsible for LSC and correlated with an imbalance of immune cell subsets in AML. 展开更多

STUDIES ON LYMPHOCYTE SUBSETS, HUMORAL IMMUNITY AND THEIR RELATIONSHIP WITH SELENIUM IN PATIENTS WITH KASHIN-BECK DISEASE

Objective To study the humoral immunity status and distribution pattern of lymphocyte subgroups of peripheral blood mononuclear cell (PBMC) in patients with Kashin-Beck Disease (KBD), and their relationship with erythrocyte selenium. Methods 23 X-ray diagnosed patients, 22 age- and sex- matched healthy children in KBD affected area (KAA), and 25 in KBD non-affected area (KNAA) were randomly selected. Immunohistochemistry with monoclonal antibodies anti-CD4, anti-CD8, anti-CD20 was conducted to analyze the lymphocyte subsets. Serum IgM, IgA, IgG, Complement C3 and C4 were assayed using rate nephelometry (Array 360 System, USA). The contents of erythrocyte selenium was determined by 2,3-diaminonaphthalene fluorescence assay. Results CD4+ and CD8+ cells percentage in PBMCs and serum IgA were significantly lower in KAA than those in KNAA(P< 0.05). CD20+ percentage in KAA displayed a decreasing trend compared to KNAA, although not statistically significantly. No statistical differences were found in CD4/CD8 ratio, serum IgG, IgM, C3 and C4 levels. Erythrocyte selenium level in KAA still showed a pronounced decrease compared to that in KNAA. Correlation analysis showed that erythrocyte selenium contents had a strong association with the CD4 cell percentage (r= 0.625, P< 0.05), as well as serum IgA (r= 0.462, P< 0.05). In addition, a moderate correlation between the serum IgA and CD4+ percentage (r= 0.130, P> 0.05) was found. Conclusion These results suggested that children in KAA had a comparably low cellular immunity level and their humoral immunity status was also in a state of moderate immune suppression. Of this immune disorder in Kashin-Beck disease patients, selenium deficiency probably played a critical role via affecting the distribution pattern of peripheral blood lymphocyte. Selenium-deficiency and immune impairment maybe both have something to do with the cause-effect chain of KBD. 展开更多

In order to meet the requirements of combustion optimization for saving energy and reducing pollutant emission simultaneously,an immune cell subsets based multiobjective optimization algorithm(ICSMOA)is proposed.In the ICSMOA,the subset division operator and the immunological tolerance operation are defined.Preference can be easily addressed by using the subset division operator,and the distribution of the solutions can be guaranteed by the immunological tolerance operation.Using the ICSMOA,a group of Pareto optimal solutions can be obtained.However,by the traditional weighting method(WM),only one solution can be obtained and it cannot be judged as Pareto optimal or not.In contrast to the solutions obtained by the repeatedly performed WM,the simulation results show that most solutions obtained by the ICSMOA are better than the solutions obtained by the WM.In addition,the Pareto front obtained by the ICSMOA is not as uniform as most classical multiobjective optimization algorithms.More optimal solutions which meet the preference set by the decision-maker can be obtained and they are very useful for industrial application. 展开更多

BACKGROUND: Besides local changes of cranial parenchymal cells, hemorrhage, etc., severe traumatic brain injuries also cause the changes of total body fluid and various functions, and the changes of lymphocytes and T lymphocyte subsets should be paid more attention to. OBJECTIVE: To reveal the changing laws of T lymphocyte subsets after severe traumatic brain injury, and compare with mild to moderate brain injury. DESIGN: A comparative observation. SETTINGS: Department of Neurosurgery, Longgang District Buji People's Hospital of Shenzhen City; Central Laboratory of Shenzhen Hospital of Prevention and Cure for Chronic Disease. PARTICIPANTS: All the subjects were selected from the Department of Neurosurgery, Longgang District Buji People's Hospital of Shenzhen City from August 2002 to August 2005. Thirty patients with severe brain injury, whose Glasgow coma score (GCS) was ≤ 8 points, were taken as the experimental group, including 21 males and 9 females, aging 16 - 62 years. Meanwhile, 30 patients with mild traumatic brain injury were taken as the control group (GCS ranged 14- 15 points), including 18 males and 12 females, aging 15 -58 years. All the subjects were in admission at 6 hours after injury, without disease of major organs before injury Informed consents were obtained from all the patients or their relatives. METHODS: (1) The T lymphocytes and the subsets in peripheral blood were detected with immunofluorescent tricolor flow cytometry at l, 3, 7 and 14 days after injury in both groups. (2) The conditions of pulmonary infections were observed at 4 days after injury. The differences of measurement data were compared with the t test. MAIN OUTCOME MEASURES: Changes of T lymphocytes subsets at 1 - 14 days after severe and mild or moderate traumatic injury. RESULTS: Finally, 28 and 25 patients with mild to moderate traumatic brain injury, whereas 25 and 21 patients with severe traumatic brain injury were analyzed at 7 and 14 days respectively, and the missed ones died due to the development of disease. (1) Changes of T lymphocyte subsets: At 1 and 3 days after injury, CD3, CD4, CD8, CD4/CD8 began to decrease, whereas CD8 increased in the experimental group, which were very significantly different from those in the control group (t =2.77 - 3.26, P 〈 0.01), and began to recover at 7 days, which were significantly different from those in the control group (t = 2.06 - 2.24, P 〈 0.05), and generally recovered to the normal levels at 14 days (P 〉 0.05). (2) Conditions of pulmonary infections: At 4 days after injury, the rate of pulmonary infection was significantly different between the experimental group and control group [73% (22/30), 0, x2=37.29, P 〈 0.01]. CONCLUSION: Patients with severe traumatic brain injury suffer from damages of cellular immune function at early period (within 7 days), and they are easily to be accompanied by pulmonary infections. 展开更多

This study examined the effect of intensive insulin therapy on immune function and inflammatory factors at the early phase after severe trauma. At day 1, 3, 5, 7 after admission, subsets of CD4+ helper T lymphocytes (Th1/Th2) and human leukocyte antigen (HLA)-DR expression on CD14+ monocytes were flow cytometrically measured. Levels of cytokines, including tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), interleukin-10 (IL-10) and other immunity markers, such as IgA, IgG, IgM, C3, C4 and C reaction protein (CRP) were examined in two groups. The results showed that TNF-α, IL-6 and CRP levels in the intensive insulin therapy group were significantly lower than those in the conventional therapy group, whereas IL-10 levels were substantially increased after intensive insulin therapy. C3 level at day 3, 5, 7 and C4 levels at day 5, 7 were lower in the intensive therapy group than in the conventional therapy group. Th1/Th2 ratios decreased gradually over time in both groups, and were much lower at day 3, 5, 7 in intensive therapy group. There were significant differences among day 3 to day 7 after admission in HLA-DR expression in CD14+ monocytes. It was concluded that the intensive insulin therapy could decrease pro-inflammatory cytokines and increase anti-inflammatory cytokines in the elderly suffering from severe trauma, at the same time, with complement recovery being delayed. Moreover, intensive insulin therapy promoted immune suppression and, therefore, measures need be taken to address the issue. 展开更多

METHODS: The levels of T lymphocyte subsets and mlL-2R in peripheral blood mononuclear cells (PBMCs) were measured by biotin-streptavidin (BSA) technique in patients with chronic hepatitis B before and after the treatment with liniment levamisole. RESULTS: After one course of treatment with liniment levamisole, the levels of CD3+, CD4+, and the ratio of CD4+/CD8+ increased as compared to those before the treatment but the level of CD8+ decreased. The total expression level of mIL-2R in PBMCs increased before and after the treatment with liniment levamisole. CONCLUSION: Liniment levamisole may reinforce cellular immune functions of patients with chronic hepatitis B. 展开更多

Objective:To evaluate humoral immunity and peripheral blood T lymphocyte subset levels after Mycoplasma pneumoniae infection. Methods:118 cases of children with Mycoplasma pneumoniae infection were selected as observation group, and 98 cases of healthy children receiving medical examination in our hospital during the same period were selected as control group. The differences in humoral immunity, T lymphocyte subsets, anti-inflammatory and pro-inflammatory factors, Th1/Th2 balance state, etc were compared between two groups of children. Results:Serum IgA level of observation group was lower than that of control group (P<0.05), and IgM level was higher than that of control group (P<0.05). Peripheral blood CD3+and CD4+T lymphocyte levels as well as CD4+/CD8+ratio were lower than those of control group (P<0.05), while CD8+T lymphocyte level was higher than that of control group (P<0.05). Serum pro-inflammatory factors IL-8 and TNF-αlevels were higher than those of control group (P<0.05), while anti-inflammatory factors IL-10 and IL-13 levels were lower than those of control group (P<0.05). Serum IL-4, IFN-γand IL-4/IFN-γlevels were higher than those of control group (P<0.05). Conclusions:Humoral immune and cellular immune suppression, high inflammatory state and so on after Mycoplasma pneumoniae infection are the important mechanisms leading to disease progression. 展开更多

目的从免疫角度探讨痰湿体质与高尿酸血症(HUA)的关系,揭示“体病相关”理论的生物学基础,为HUA及其并发症的防治提供新思路及科学依据。方法依据HUA及中医体质诊断标准及纳排标准,以2023年1—6月在北京中医药大学东方医院风湿科就诊的... 目的从免疫角度探讨痰湿体质与高尿酸血症(HUA)的关系,揭示“体病相关”理论的生物学基础,为HUA及其并发症的防治提供新思路及科学依据。方法依据HUA及中医体质诊断标准及纳排标准,以2023年1—6月在北京中医药大学东方医院风湿科就诊的平和体质HUA患者20例为平和体质HUA组,痰湿体质HUA患者20例为痰湿体质HUA组,另选择同期体检的平和体质健康者20例为平和体质健康组。比较3组人群性别、年龄、外周静脉血单个核细胞(PBMC)中抗体标记的辅助性T细胞(Th1、Th2、Th17)、调节性T细胞(Treg)比例和Th17/Treg比值、Th1/Th2比值。结果3组人群性别、年龄比较差异均无统计学意义(P均>0.05)。痰湿体质HUA组Th1细胞比例、Th17/Treg比值、Th1/Th2比值均明显高于平和体质健康组(P均<0.05),Th17细胞比例明显高于平和体质健康组和平和体质HUA组(P均<0.05),Treg细胞比例明显低于平和体质健康组(P<0.05);平和体质HUA组Th1细胞比例、Th17细胞比例、Treg细胞比例、Th17/Treg比值、Th1/Th2比值与平和体质健康组比较差异均无统计学意义(P均>0.05);3组Th2细胞比例比较差异均无统计学意义(P均>0.05)。结论痰湿体质HUA患者存在更明显的免疫失衡现象,以Th1和Th17细胞介导的免疫反应增强为主,早期调节痰湿体质或可为HUA治疗提供新方向。 展开更多

试验旨在探讨酵母细胞壁(YCW)对黄羽肉鸡生长性能、免疫、抗氧化能力和肠道菌群的影响。选取1 680只1日龄黄羽肉鸡,随机分为4组,每组6个重复,每个重复70只黄羽肉鸡,对照组(A组)饲喂基础饲粮,B组、C组、D组分别在基础饲粮中添加0.05%、0.... 试验旨在探讨酵母细胞壁(YCW)对黄羽肉鸡生长性能、免疫、抗氧化能力和肠道菌群的影响。选取1 680只1日龄黄羽肉鸡,随机分为4组,每组6个重复,每个重复70只黄羽肉鸡,对照组(A组)饲喂基础饲粮,B组、C组、D组分别在基础饲粮中添加0.05%、0.10%和0.15%的YCW。试验期63 d。结果显示,与对照组相比,1~21 d C组黄羽肉鸡ADFI显著增加(P<0.05)。与对照组相比,D组黄羽肉鸡的胸腺指数显著提高(P<0.05)。与对照组相比,C组和D组黄羽肉鸡血清中总抗氧化能力(T-AOC)显著提高(P<0.05),D组血清超氧化物歧化酶(T-SOD)活性显著提高(P<0.05),B组CD3+、 CD4+含量和CD4+/CD8+显著提高(P<0.05)。与对照组相比,C组黄羽肉鸡回肠中Toll样受体2 (TLR-2)、黏蛋白2 (MUC2)、紧密连接蛋白1(ZO-1)和闭合蛋白(Occludin)的mRNA表达量显著提高(P<0.05)。与对照组相比,D组黄羽肉鸡回肠食糜内的大肠杆菌和沙门氏菌的数量显著降低(P<0.05),各组黄羽肉鸡回肠黏膜中的总菌数量均显著增加(P<0.05),C组肉鸡回肠黏膜中的沙门氏菌数量显著降低(P<0.05)。研究表明,饲粮中添加YCW可能通过促进免疫器官发育,影响外周血T淋巴细胞亚群的平衡,增强肠道免疫屏障基因表达,调节肠道菌群结构,从而提高黄羽肉鸡的生长性能。YCW适宜的添加剂量为0.10%。 展开更多

背景:既往研究表明经验方剂活髓方对骨髓增生异常综合征患者的免疫及造血调节起到增效减毒的协同作用,但具体机制尚未明确。目的:探讨活髓方对骨髓增生异常综合征模型大鼠骨髓造血的影响及机制。方法:70只SD大鼠采用随机数字表法分为正... 背景:既往研究表明经验方剂活髓方对骨髓增生异常综合征患者的免疫及造血调节起到增效减毒的协同作用,但具体机制尚未明确。目的:探讨活髓方对骨髓增生异常综合征模型大鼠骨髓造血的影响及机制。方法:70只SD大鼠采用随机数字表法分为正常对照组10只、模型组15只、西药组15只、活髓方低剂量组15只、活髓方高剂量组15只。除正常对照组外,其余4组采用尾静脉注射二甲基苯蒽诱导建立大鼠骨髓增生异常综合征模型,造模后正常对照组、模型组给予生理盐水,西药组给予沙利度胺胶囊10 mg/kg、维A酸片4 mg/kg,活髓方低、高剂量组分别给予活髓方1.5,6 g/kg,每天灌胃1次,连续给药28 d,取外周血和股骨骨髓组织,检测外周血常规、骨髓活检造血增殖情况,采用流式细胞术检测T淋巴细胞亚群及T淋巴细胞上CTLA-4和PD-1的表达。结果与结论:(1)与正常对照组相比,模型组外周血白细胞、中性粒细胞、血红蛋白、血小板和CD4^(+)、CD4^(+)/CD8^(+)水平显著下降(P<0.05),CD4^(+)PD-1^(+)、CD8^(+)PD-1^(+)、CD4^(+)CTLA-4^(+)和CD8^(+)CTLA-4^(+)表达显著上调(P<0.05);(2)与模型组相比,各给药组的骨髓增生程度均有改善,但组间无统计学差异(P > 0.05);(3)与模型组相比,活髓方高剂量组白细胞、血红蛋白、血小板和CD4^(+)、CD4^(+)/CD8^(+)水平显著升高(P<0.05),CD8^(+)水平显著降低(P<0.05),CD4^(+)PD-1^(+)、CD8^(+)PD-1^(+)、CD4^(+)CTLA-4^(+)和CD8^(+)CTLA-4^(+)表达水平下降但无统计意义(P > 0.05);(4)西药组与活髓方高剂量组显示出相似的疗效;活髓方高剂量组较低剂量组各指标改善均有优势。结果表明,活髓方可改善骨髓增生异常综合征模型大鼠的骨髓造血,提高CD4^(+)、CD4^(+)/CD8^(+)水平,下调CD4^(+)PD-1^(+)、CD8^(+)PD-1^(+)、CD4^(+)CTLA-4^(+)和CD8^(+)CTLA-4^(+)表达,可能与免疫负调控机制相关。 展开更多

目的:探究止抽汤(Zhichou Decoction,ZCD)对多发性抽动症(Tourette syndrome,TS)模型大鼠外周血T淋巴细胞亚群及T辅助(Th)淋巴细胞Th1/Th2细胞平衡的影响,以明确该方对TS大鼠免疫调控作用。方法:建立TS模型成功后将大鼠随机分为模型组... 目的:探究止抽汤(Zhichou Decoction,ZCD)对多发性抽动症(Tourette syndrome,TS)模型大鼠外周血T淋巴细胞亚群及T辅助(Th)淋巴细胞Th1/Th2细胞平衡的影响,以明确该方对TS大鼠免疫调控作用。方法:建立TS模型成功后将大鼠随机分为模型组、氟哌啶醇组、ZCD高、中、低剂量组,同时设立对照组;连续给药4周后处死大鼠,称定大鼠脾脏、胸腺质量,计算脏器系数;苏木素-伊红(HE)染色观察大鼠脑组织形态,ELISA法检测纹状体中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)白细胞介素-6(interleukin-6,IL-6)及血清中γ干扰素(interferon-γ,IFN-γ)和白细胞介素-4(interleukin-4,IL-4)水平;流式细胞术检测外周血中CD3^(+)T、CD4^(+)T、CD8^(+)T淋巴细胞及Th1、Th2各细胞亚群比例变化。结果:与模型组比较,给药4周后ZCD高、中剂量组大鼠刻板运动评分均降低(P<0.05),脾脏、胸腺指数增加(P<0.05,P<0.01),TNF-α、IL-6及IFN-γ水平降低(P<0.05),外周血中CD3^(+)占比增加(P<0.01、P<0.05);CD8^(+)占比降低(P<0.05),CD4^(+)/CD8^(+)比例增加(P<0.05);ZCD高剂量组还可增加CD4^(+)百分比(P<0.05)并具增加血清中IL-4水平的趋势;在Th淋巴细胞亚群中,ZCD高剂量组Th1细胞占比降低,Th2细胞比例增加(P<0.05),Th1/Th2细胞比值降低(P<0.05);ZCD中剂量组Th1细胞占比显著降低,Th2细胞稍有增加,Th1/Th2细胞比值呈降低趋势;ZCD低剂量组脾脏指数增加(P<0.05),IL-6水平降低(P<0.05),但对其他指标均无明显调控作用。结论:ZCD可显著改善TS大鼠症状,降低脑内神经炎性分子水平,其机制可能与调控外周血T淋巴细胞亚群及Th1/Th2细胞平衡,改善免疫功能相关。 展开更多

Nonetheless, no creditab... BACKGROUND Hepatitis B virus (HBV) has been recognized as a leading cause of hepatocellular carcinoma (HCC). Numerous reports suggest that immune infiltration can predict the prognosis of HCC. Nonetheless, no creditable markers for prognosis of HBV-related HCC have been established by systematically assessing the immune-related markers based on tumor transcriptomes. AIM To establish an immune-related marker based on the cell compositions of immune infiltrate obtained based on tumor transcriptomes, so as to enhance the prediction accuracy of HBV-related HCC prognosis. METHODS RNA expression patterns as well as the relevant clinical data of HCC patients were obtained from The Cancer Genome Atlas. Twenty-two immunocyte fraction types were estimated by cell type identification by estimating relative subsets of RNA transcripts. Subsequently, the least absolute shrinkage and selection operator (LASSO) Cox regression model was employed to construct an immunoscore based on the immunocyte fraction types. Afterwards, the receiver operating characteristic (ROC) curve, Kaplan-Meier, and multivariate Cox analyses were performed. Additionally, a nomogram for prognosis that integrated the immunoscore as well as the clinical features was established. Meanwhile, the correlation of immunoscore with immune genes was also detected, and gene set enrichment analysis (GSEA) of the immunoscore was conducted. RESULTS A total of 22 immunocyte fraction types were predicted and compared among the tumor as well as non-tumor samples. An immunoscore was constructed through adopting the LASSO model, which contained eight immunocyte fraction types. Meanwhile, the areas under the ROC curves for the immunoscore biomarker prognostic model were 0.971, 0.912, and 0.975 for 1-, 3-, and 5-year overall survival (OS), respectively. Difference in OS between the high-immunoscore group and the low-immunoscore group was statistically significant [hazard ratio (HR)= 66.007, 95% confidence interval (CI): 8.361-521.105;P < 0.0001]. Moreover, multivariable analysis showed that the immunoscore was an independent factor for predicting the prognosis (HR = 2.997, 95%CI: 1.737-5.170). A nomogram was established, and the C-index was 0.757 (95%CI: 0.648-0.866). The immunoscore showed a significant negative correlation with the expression of PD-1 (P = 0.024), PD-L1 (P = 0.026), PD-L2 (P = 0.029), and CD27 (P = 0.033). Eight pathways were confirmed by GSEA. CONCLUSION The established immunoscore can potentially serve as a candidate marker to estimate the OS for HBV-related HCC cases. 展开更多

Objective The aim of the study was to explore the difference between immune cell subsets during the incubation of cytokine-induced kill cells (CIKs) from patients with and without hepatitis B virus (HBV). Methods Peripheral blood samples were extracted from 50 tumor patients, and were divided into two groups according to the presence or absence of HBV. The proliferation rate and activity of CIK cells were examined based on counts on days 1, 5, 7, 9, 11, 13, and 15 of culture. Additionally, the CD3+, CD4+, CD8+, CD3+CD8+, C+)3+CD4+, and CD3+CD56+ T cell populations were analyzed by flow cytometry on days 5, 7, 10, 13, and 15 of culture. Results Proliferation over a 15-day period was higher in the HBV-positive group than in the negative group (280-fold vs. 180-fold increase, respectively), but there was no significant difference between the two groups at each time point. The frequencies of CD3+, CD8+ T, CD3+CD8+, and CD3+CD56+T cells increased over time, while those of CD4+ and CD3+CD4+ T cells decreased over time, and these changes were greater in the positive group than in the negative group. The differences in CD8+ T cells and CD3+CD4+ T cells between the two groups were significant (P 〈 0.05). Conclusion The proliferative capacity of CIK cells was higher for patients in the HBV-positive group than those in the HBV-negative group, and immune cell subsets were more favorable in the HBV-positive group than the neaative arouD. 展开更多

目的观察楮芍凉血汤加减治疗寻常型银屑病的临床疗效和对免疫功能、辅助性T淋巴细胞17(Th17)相关因子的影响。方法选取2020年2月至2021年12月在本院接受治疗64例进行期PV患者,采用随机数字表法将患者分为对照组和观察组各32例。对照组... 目的观察楮芍凉血汤加减治疗寻常型银屑病的临床疗效和对免疫功能、辅助性T淋巴细胞17(Th17)相关因子的影响。方法选取2020年2月至2021年12月在本院接受治疗64例进行期PV患者,采用随机数字表法将患者分为对照组和观察组各32例。对照组患者接受常规西药治疗,观察组接受楮芍凉血汤加减治疗,治疗周期为6周。所有研究对象均检测外周血T淋巴细胞亚群、分离单个核细胞(PBMC)情况,分析PBMC中Thl7细胞相关因子白细胞介素17(IL-17)、IL-22、维甲酸相关孤儿受体γ(ROR-γ)mRNA、干扰素γ(INF-γ)mRNA相对表达量,并对比临床总疗效。结果治疗6周后,观察组总有效治疗30例(93.75%),对照组总有效治疗24例(75.00%),2组总疗效差异具有统计学意义(P<0.05)。治疗6周后,2组患者T淋巴细胞指标CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)水平均升高,CD8^(+)及Th17相关因子IL-17、IL-22、RORC mRNA、INF-γmRNA相对表达量均降低,组内不同时点和组间同时点比较差异均具有统计学意义(P<0.05)。结论楮芍凉血汤可有效治疗寻常型银屑病,且作用机制与T淋巴细胞亚群分布及Thl7细胞相关因子有关,可以提高机体免疫功能,以此减少不良症状,提高临床疗效。 展开更多

Background:Colorectal cancer(CRC)is a common gastrointestinal malignancy.The T lymphocyte subsets are important in the develop-ment,invasion and metastasis of tumors,including CRC.Nevertheless,limited research has explored the relationship between T cell subpopu-lations and the clinical characteristics of CRC.This study compared the T lymphocyte subsets in patients with CRC and healthy individuals,and assessed the relationship between these values and clinical characteristics.Methods:Peripheral blood was collected from 100 patients with CRC and 54 healthy individuals.The numbers of CD3^(+)T,CD4^(+)T,and CD8^(+)T lymphocytes,NK cells,and the CD4^(+)T/CD8^(+)T ratio in peripheral blood were measured using flow cytometry,and were compared between CRC patients and healthy individuals.Spearman's correlation analysis was performed to investigate the relationship between the T lymphocyte subsets in patients diagnosed with CRC and the levels of carcinoembryonic antigen(CEA)and thymidine kinase 1(TK1).Receiver operating characteristic(ROC)curves were utilized to evaluate the potential utility of the T lymphocyte counts in predicting lymph node metastasis,vas-cular infiltration,and high Ki-67 expression.Results:The CRC patients had lower counts of CD3^(+)T,CD4^(+)T,and CD8^(+)T lymphocytes compared to the healthy population(P<0.05).However,no significant differences were observed in the CD4^(+)/CD8^(+)ratio or NK cells(P>0.05).Notably,the CD3^(+)T,CD4^(+)T,and CD8^(+)T lym-phocyte counts were higher in patients with stageⅠ-Ⅱdisease,no lymph node metastasis,no vascular invasion,and low Ki-67 expression than in those with stageⅢ,lymph node metastasis,vascular invasion,and high Ki-67 expression(P<0.05).There was a negative association be-tween the CD3^(+)T,CD4^(+)T,and CD8^(+)T lymphocyte counts and CEA and TK1 levels in patients with CRC.The ROC curves demonstrated that CD3^(+)T,CD4^(+)T,and CD8^(+)T lymphocyte counts had significant predictive value for lymph node metastasis,vascular infiltration,and high Ki-67 expression.Conclusions:The peripheral blood CD3^(+)T,CD4^(+)T,and CD8^(+)T lymphocyte counts are related to the clinical traits of patients with CRC and can predict the prognosis of the disease. 展开更多

Effect of Astragalus Polysaccharide on the Cell-mediated Immunity of Traumatic Stress Mice

To investigate the changes of immune functions and the effects of Astragalus polysaccharide (ASP) on the cell-mediated immunity of the traumatic stress model of mouse by amputation, 50 mice were randomly divided into 5 groups for study, in which the group A and B served as the normal control (by injecton of 0.5 ml of saline intra-peritoneally daily), and as the stress control (by intra-peritoneal injecton of 0.5 ml of normal saline into mice after amputation) respectively, to the group C, D and E of mice, 1000 mg/kg (high dose), 300 mg/kg (median dose) and 250 mg/kg (low dose). The CD4 + and CD8 + T cells as well as the expression of the c-fos protein were determined by immunohistochemical techniques, and the expressions of NF-κB mRNA and IL-10 mRNA were assayed by hybridization in situ . The experimental results showed that in comparison with the normal control group of mice (group A), the expression levels of NF-κB mRNA, IL-10 mRNA and the c-fos protein in the tissues of thymus and spleen in the stress controls were significantly elevated and the CD4 + T cells and CD4/CD8 ratio were decreased. However, in comparison with the stress control of mice (group B), the expressions of NF-κB mRNA and IL-10 mRNA were inhibited by ASP, and the CD4 + T cells and CD4/CD8 ratio were increased in groups C, D and E, but the level of c-fos protein was decreased. There was no significant difference in these parameters among group C, D and E. It is concluded that the functions of cell-mediated immunity of mice were disturbed under the stress condition of the traumatic injuries after amputation. And the immune functions can be effectively restored by the use of Astragalus polysaccharide. 展开更多
