AIM: To explore the associations of polymorphisms of lipopolysaccharide binding protein (LBP), cluster of differentiation 14 (CD14), toll-like receptor 4 (TLR-4), interleukin-6 (IL-6) and tumor necrosis factor α (TNF...AIM: To explore the associations of polymorphisms of lipopolysaccharide binding protein (LBP), cluster of differentiation 14 (CD14), toll-like receptor 4 (TLR-4), interleukin-6 (IL-6) and tumor necrosis factor α (TNF-α) with the colorectal carcinoma (CRC) risk in Han Chinese. METHODS: Polymorphisms of LBP (rs1739654, rs223 2596, rs2232618), CD14 (rs77083413, rs4914), TLR-4 (rs5030719), IL-6 (rs13306435) and TNF-α (rs35131721) were genotyped in 479 cases of sporadic colorectal carcinoma and 486 healthy controls of Han Chinese in a case-control study. Single-nucleotide polymorphisms (SNPs) between cases and controls were analyzed by unconditional logistic regression. RESULTS: GA and GG genotypes of LBP rs2232596 were associated with a significantly increased risk ofCRC [odds ratio (OR) = 1.51, 95% confidence interval (CI) 1.15-1.99, P = 0.003; OR = 2.49, 95% CI 1.16-5.38, P = 0.016, respectively]. A similar association was also observed for the CG genotype of CD14 rs4914 (OR= 1.69, 95% CI 1.20-2.36, P = 0.002). In addition, a combination of polymorphisms in LBP rs2232596 and CD14 rs4914 led to a 3.4-fold increased risk of CRC (OR = 3.44, 95% CI 1.94-6.10, P = 0.000). CONCLUSION: This study highlights the LBP rs2232596 and CD14 rs4914 polymorphisms as biomarkers for elevated CRC susceptibility in the Chinese Han population.展开更多
目的探究肺结核伴其他肺部感染患者体内病原微生物分布特征,并分析白介素-17(interleukin-17,IL-17)、高迁移率族蛋白B1(high mobility group protein B1,HMGB1)、可溶性白细胞分化抗原14(soluble leukocyte differentiation antigen 14...目的探究肺结核伴其他肺部感染患者体内病原微生物分布特征,并分析白介素-17(interleukin-17,IL-17)、高迁移率族蛋白B1(high mobility group protein B1,HMGB1)、可溶性白细胞分化抗原14(soluble leukocyte differentiation antigen 14,sCD14)表达对疾病病情的评估意义。方法选取成都市龙泉驿区第一人民医院/四川大学华西医院龙泉医院2018年3月-2020年3月收治的543例肺结核患者为研究对象,根据是否发生其他肺部感染分为研究组(n=330)和对照组(n=213)。分析感染患者病原菌的分布特征,并对比两组患者的肺功能、IL-17、HMGB1、sCD14水平,探究以上指标对于感染患者病情的评估效果。结果330份痰液标本培养出病原菌368株,其中革兰氏阴性菌217株(58.97%),革兰氏阳性菌98株(26.63%),真菌53株(14.40%)。研究组患者肺功能指标第一秒用力呼气容积(Forced expiratory volume in the first second,FEV1)、最大呼气中段流量(Maximum middle expiratory flow,MMF)、分钟最大通气量(Min maximum ventilation,MVV)均低于对照组(t_(1)=16.3952,t_(2)=42.2180,t_(3)=6.1815,均P<0.05),血清指标IL-17、HMGB1、sCD14均高于对照组(t_(4)=59.4263,t_(5)=20.1960,t_(6)=13.3835,均P<0.05)。按照肺部感染评分(clinical pulmonary infection score,CPIS)标准将研究组患者分为轻度感染患者131例(39.70%),中度感染患者113例(34.24%),重度感染患者86例(26.06%),比较发现三组患者肺功能与血清指标之间均有明显差异,其中肺功能指标FEV1、MMF、MVV轻度感染>中度感染组>重度感染组(F_(1)=68.77,F_(2)=87.65,F_(3)=14.83,均P<0.05),血清指标IL-17、HMGB1、sCD14轻度感染<中度感染组<重度感染组(F_(4)=465.63,F_(5)=67.89,F_(6)=56.09,均P<0.05)。按照不同生存结局将研究组患者分为生存组患者293例(88.79%),死亡组患者37例(11.21%),比较发现生存组患者肺功能指标FEV1、MMF、MVV均高于死亡组(t_(1)=15.7970,t_(2)=6.5952,t_(3)=6.1373,均P<0.05),血清指标IL-17、HMGB1、sCD14均低于死亡组(t_(4)=29.7796,t_(5)=9.4618,t_(6)=10.6096,均P<0.05)。结论肺结核伴其他肺部感染病原菌多为革兰氏阴性菌,其次为革兰氏阳性菌。感染患者肺功能损伤较严重,血清中IL-17、HMGB1、sCD14均呈异常高表达,随着感染程度的加重,肺功能损伤更重,血清IL-17、HMGB1、sCD14表达更高。展开更多
Objective:To investigate the association between the cluster of differentiation 14(CD14)-I59C/T(rs2569190) gene polymorphism and susceptibility to acute brucellosis in an Iranian population.Methods:The study included ...Objective:To investigate the association between the cluster of differentiation 14(CD14)-I59C/T(rs2569190) gene polymorphism and susceptibility to acute brucellosis in an Iranian population.Methods:The study included 153 Iranian patients with active brucellosis and 128 healthy individuals as the control group.Genotyping of the CD 14 variant was performed using an amplification refractory mutation system-polymerase chain reaction method.Results:The prevalence of CD14-159 TT and CT genotypes were associated with increased risk of brucellosis[odds ratio(OR)=l.993.95%confidence interval(95%CD=1.07-3.71.P=0.03 for CT:OR=3.869.95%CI= 1.91-7.84,P=0.01 for TT genotype.Additionally,the minor allele(T) was significantly more frequently present in brucellosis patients than in controls(61%vs.45%.respectively),and was a risk factor for brucellosis(OR=3.058.95%CI= 1.507-6.315.P=0.01).Condusions:The findings provid suggestive evidence of association of the CDI4-159C/T gene polymorphism with susceptibility to acute brucellosis in the Iranian population.展开更多
