Variations in obvious form, stress-strain curve, peak strength, peak strain and elastic modulus with temperature were analyzed and the essence of rock failure modes was explored. The results indicate that, compared with granite after the high temperature treatment, the brittle-ductile transition critical temperature is lower, the densification stage is longer, the elastic modulus is smaller and the damage is larger under high temperature. In addition, the peak stress is lower and the peak strain is greater, but both of them change more obviously with the increase of temperature compared with that of granite after the high temperature treatment. Furthermore, the failure modes of granite after the high temperature treatment and under high temperature show a remarkable difference. Below 100 ℃, the failure modes of granite under both conditions are the same, presenting splitting failure. However, after 100 ℃, the failure modes of granite after the high temperature treatment and under high temperature present splitting failure and shear failure, respectively.展开更多

B...Raman spectra of purified oxygen evolution core complexes (Pd OECC) thin films on silver mirror substrates have been taken over the frequency range of 250-3100 cm -1 by surface enhanced Raman scattering (SERS). Besides the fundamental frequency modes of β_carotene in Pd OECC, many weak peaks are observed. According to the selection rules of overtone and combination bands, most of them are attributed to the second_order Raman spectra of β_carotene. Compared with the SERS of normal Pd OECC, the SERS of Pd OECC after strong illumination shows a decrease in scattering intensity and an increase in line widths, indicating changes of conformation and micro_environment of β_carotene. The results of SERS are consistent with the changes of absorption spectrum of Pd OECC induced by strong illumination. There are no changes that can be ascribed to new vibration bands, so it is deduced that Pd OECC on the silver mirror is identical to that in the solution. In summary, SERS proved a good method to study the photodamage mechanism of photosynthesis.展开更多
To investigate the mechanical properties of a dowel action under fatigue loads, nine reinforced concrete specimens were fabricated, and the monotonic and fatigue loadings were performed on these specimens, respectivel...To investigate the mechanical properties of a dowel action under fatigue loads, nine reinforced concrete specimens were fabricated, and the monotonic and fatigue loadings were performed on these specimens, respectively. All of these specimens were divided into two series. Six specimens in SeriesⅠwith different bar diameters of 12, 20 and 25 mm were subjected to monotonic loads and were used to confirm the ultimate bearing capacity. The remaining three specimens in Series Ⅱ were subjected to fatigue loads and were designed to investigate the attenuation character of dowel action and the fatigue failure modes. The test results show that the accumulated fatigue damage due to fatigue loads can reduce the ultimate bearing capacity of specimens. With the increase in fatigue loads, the failure mode can transform to fatigue rupture of the dowel bar under the serviceability loading state,i. e. 55% of the monotonic capacity. The fatigue life is determined by the fatigue properties of steel and concrete.Based on the test data, the failure process of dowel action can be divided into two stages: the accumulation of fatigue damage and the fatigue rupture of dowel bar.展开更多

Methods: Human Hepatocarcinoma cell line BEL-7402 being cultured in vitro, the effect of AT and (or) AA on its growth inhibition and its two intracellular signal molecules was evaluated separately using MTT and Western blot. Results: AT at a few μmol/L concentration could suppress abnormal proliferation of human hepatocarcinoma cells, and initiate their apoptosis by activation of caspase-3, and activate extracellular-signal regulated kinases (ERKs), which were dependent on the dosage of AT conspicuously. This form of "necrotic cell death" appears to be dependent on the generation of reactive oxygen species. Recent data have indicated that superoxide generation is dependent on the activation of NADPH oxidases, which form a complex with the adaptor molecules RIP1 and TRADD. The mechanism of superoxide generation further establishes RIP1 as the central molecule in ROS production and cell death initiated by TNFa and other death receptors. A role for the sustained JNK activation in necrotic cell death is also suggested. The sensitization of virus-infected cells to TNFα indicates that necrotic cell death may represent an alternative cell death pathway for clearance of infected cells.展开更多

Methods: Seven truncated CORE-GFP (green fluorescent protein) fusion protein expression plasmids were constructed, which contained HCV CORE sequences derived from tumor tissues (BT) and non-tumor tissues (BNT) from one patient infected with HCV. Amino acid (aa) lengths were BT: 1?172 aa, 1?126 aa, 1?58 aa, 59?126 aa, 127?172 aa; BNT: 1?172 aa and C191: 1?172 aa respectively. Subcellular localization of CORE-GFP was analyzed by con-focal laser scanning microscope. Apoptosis and necrosis were quantified by flow cytometry. Results: Different truncated CORE-GFP localized mainly in the cytoplasm, but nuclear staining was also observed. HCV CORE could induce apoptosis and necrosis, and different truncated COREs could induce cell apoptosis and necrosis at different levels. Among the same length 1?172 aa of BT, BNT and C191, the cell apoptosis and necrosis percentage of BT is highest, and C191 is the lowest (BT>BNT>C191). To the different fragment COREs of BT, N-terminal of CORE induced apoptosis and necrosis higher, compared with that of C-terminal (1?172 aa>1?126 aa>1?58 aa>127?172 aa>59?126 aa). However, experimental evidence shows that nonlinear failure criterion is able to represent fairly well the failure of almost all types of rocks, A nonlinear Hock-Brown failure criterion is employed to estimate the supporting pressures of shallow tunnels in limit analysis framework. Two failure mechanisms are proposed for calculating the work rate of extemal force and the internal energy dissipation. A tangential line to the nonlinear failure criterion is used to formulate the supporting pressure problem as a nonlinear programming problem. The objective function formulated in this way is minimized with respect to the failure mechanism and the location of tangency point. In order to assess the validity, the supporting pressures for the proposed failure mechanisms are calculated and compared with each other, and the present results are compared with previously published solutions when the nonlinear criterion is reduced to linear criterion. The agreement supports the validity of the proposed failure mechanisms. An experiment is conducted to investigate the influences of the nonlinear criterion on collapse shape and supporting pressures of shallow tunnels.展开更多

The mecha- nism by which TNF-α activates MLK3 is still not known. TNF receptor-associated factors (TRAFs) are adapter molecules that are recruited to cytoplasmic end of TNF receptor and mediate the downstream signaling, including activation of JNK. Here, we report that MLK3 associates with TRAF2, TRAF5 and TRAF6; however only TRAF2 can significantly induce the kinase activity of MLK3. The interaction domain of TRAF2 maps to the TRAF domain and for MLK3 to its C-terminal half (amino acids 511-847). Endogenous TRAF2 and MLK3 associate with each other in response to TNF-α treatment in a time-dependent manner. The association between MLK3 and TRAF2 mediates MLK3 activation and competition with the TRAF2 deletion mutant that binds to MLK3 attenuates MLK3 kinase activity in a dose-dependent manner, on TNF-α treatment. Furthermore the downstream target of MLK3, JNK was activated by TNF-α in a TRAF2-dependent manner. Hence, our data show that the direct interaction between TRAF2 and MLK3 is required for TNF-α-induced activation of MLK3 and its downstream target, JNK.展开更多

Methods Human lung adenocarcinoma c...Objective To explore the inhibitory effect of recombinant mutant human tumor necrosis factor-α(rmhTNF-α) in combination with cisplatin on human lung adenocarcinoma cell line A549. Methods Human lung adenocarcinoma cell line A549 was treated with varying concentrations of rmhTNF-α(0.38, 0.75, 1.50, 6.00 and 12.00 IU/ml) or cisplatin(3.91, 7.81, 15.63, 31.25 and 62.50 μg/ml) for 24 hours. Viable cell number was analyzed by using crystal violet staining. The inhibitory rates of A549 cells growth by the two drugs were calculated. For analyzing whether there was a synergistic effect of rmhTNF-α with cisplatin, A549 cells were treated with 0.75 IU/ml rmhTNF-α and increased concentrations of cisplatin. Results rmhTNF-α or cisplatin inhibited the growth of A549 cell lines in a dose-dependent manner. The inhibitory effect of rmhTNF-α combined with cisplatin was significantly greater than cisplatin alone at the same concentration(all P<0.01). Conclusion rmhTNF-α combined with cisplatin might have synergistic inhibitory effect on human lung adenocarcinoma cell line A549.展开更多

METHODS: Huma...AIM: To investigate the effect of bromocripUne (BCT) and tumor necrosis factor-α ClNF-α) on hepatocellular carcinoma (HCC) multidrug resistance (MDR) in nude mouse HDR model of liver neoplasm. METHODS: Human hepatocarcinoma cell line HepG2t drug resistant hepatocarcinoma cell line HepG2/adriamycin (ADM) and hepatocarcinoma cell line transfected with TNF-α gene HepG2JADM/TNF were injected into the liver of nude mice via orthotopic implantation and MDR model of liver neoplasm in vivo was established (HepG2t ADM, TNF, BCT groups). Among these groups, BCT group and TNF group were treated with BCT through gastric canal. Each group was divided into control group and chemotherapy group. Size and weight of the tumor were measured. Furthermore, tumor his^logical character and growth of the nude mice were observed and their chemosensitivity was tested. MDR-associated genes and proteins (MRP, LRP) of implanted tumors were detected by immunohistochemistry, reverse transcriptase polymerase chain reaction, and apoptosis rate of hepatocarcinoma cells was detected by TUNEL assay. RESULTS: The nude mouse model of each cell line was inoculated successfully. The tumor growth rate and weight were significantly different among groups. After chemotherapy, abdominal cavity tumor growth inhibition rate was higher in BCT group (67%) compared to ADM and TNF groups, and similar to HepG2group (54%). MDRI and LRPmRNA could be detected in all groups, but TNF-α was detected only in TNF and BCT groups. Furthermore, MDR1 and LRP protein expression of tumors in TNF and BCT groups was low similar to HepG2 group. The apoptosis rate of hepatocarcinoma cells was much higher in BCT group than in other groups with TUNEL assay. In order to study the structural response of such type of masonry, it is necessary...Historical buildings located in seismic regions have URM (unreinforced masonry) walls of considerable width as their main structure. In order to study the structural response of such type of masonry, it is necessary to know its mechanical and elastic characteristics. For such purpose, it is generally necessary to perform destructive and non-destructive tests. Other procedures, such as the study of masonry through numerical models, would make it possible to predict, with close approximation, the response of masonry to different actions. The purpose of this paper is to evaluate the behavior of URM walls subjected to axial and horizontal load by using a finite element model with the Abaqus code. Constitutive laws of materials, bricks and mortar are defined, using two constitutive models of plasticity and damage included in the materials library. The constitutive models used employ information of experimental tests performed on bricks and mortar. The results of an analysis for an URM wall subjected to axial and horizontal load are presented. Responses of walls were obtained in terms of load versus displacement for different constitutive models and for different qualities of bricks and mortar. The two constitutive models used show similar results, particularly in the response waveforms. However, the ultimate capacity values show mean variations of around 30%. The two constitutive models display high dependence upon the mechanical characteristics of bricks and mortar.展开更多

Furthermore the downstream target of MLK3, JNK was activated by TNF-α in a TRAF2-dependent manner. Hence, our data show that the direct interaction between TRAF2 and MLK3 is required for TNF-α-induced activation of MLK3 and its downstream target, JNK.

文摘Objective To explore the inhibitory effect of recombinant mutant human tumor necrosis factor-α(rmhTNF-α) in combination with cisplatin on human lung adenocarcinoma cell line A549. Methods Human lung adenocarcinoma cell line A549 was treated with varying concentrations of rmhTNF-α(0.38, 0.75, 1.50, 6.00 and 12.00 IU/ml) or cisplatin(3.91, 7.81, 15.63, 31.25 and 62.50 μg/ml) for 24 hours. Viable cell number was analyzed by using crystal violet staining. The inhibitory rates of A549 cells growth by the two drugs were calculated. For analyzing whether there was a synergistic effect of rmhTNF-α with cisplatin, A549 cells were treated with 0.75 IU/ml rmhTNF-α and increased concentrations of cisplatin. Results rmhTNF-α or cisplatin inhibited the growth of A549 cell lines in a dose-dependent manner. The inhibitory effect of rmhTNF-α combined with cisplatin was significantly greater than cisplatin alone at the same concentration(all P<0.01). Conclusion rmhTNF-α combined with cisplatin might have synergistic inhibitory effect on human lung adenocarcinoma cell line A549.

文摘AIM: To investigate the effect of bromocripUne (BCT) and tumor necrosis factor-α ClNF-α) on hepatocellular carcinoma (HCC) multidrug resistance (MDR) in nude mouse HDR model of liver neoplasm. METHODS: Human hepatocarcinoma cell line HepG2t drug resistant hepatocarcinoma cell line HepG2/adriamycin (ADM) and hepatocarcinoma cell line transfected with TNF-α gene HepG2JADM/TNF were injected into the liver of nude mice via orthotopic implantation and MDR model of liver neoplasm in vivo was established (HepG2t ADM, TNF, BCT groups). Among these groups, BCT group and TNF group were treated with BCT through gastric canal. Each group was divided into control group and chemotherapy group. Size and weight of the tumor were measured. Furthermore, tumor his^logical character and growth of the nude mice were observed and their chemosensitivity was tested. MDR-associated genes and proteins (MRP, LRP) of implanted tumors were detected by immunohistochemistry, reverse transcriptase polymerase chain reaction, and apoptosis rate of hepatocarcinoma cells was detected by TUNEL assay. RESULTS: The nude mouse model of each cell line was inoculated successfully. The tumor growth rate and weight were significantly different among groups. After chemotherapy, abdominal cavity tumor growth inhibition rate was higher in BCT group (67%) compared to ADM and TNF groups, and similar to HepG2group (54%). MDRI and LRPmRNA could be detected in all groups, but TNF-α was detected only in TNF and BCT groups. Furthermore, MDR1 and LRP protein expression of tumors in TNF and BCT groups was low similar to HepG2 group. The apoptosis rate of hepatocarcinoma cells was much higher in BCT group than in other groups with TUNEL assay. CONCLUSION: BCT and TNF-a can reverse HCC MDR in nude mouse MDR1 model of liver neoplasm. 2005 The WJG Press and Elsevier Inc. All rights reserved
文摘Historical buildings located in seismic regions have URM (unreinforced masonry) walls of considerable width as their main structure. In order to study the structural response of such type of masonry, it is necessary to know its mechanical and elastic characteristics. For such purpose, it is generally necessary to perform destructive and non-destructive tests. Other procedures, such as the study of masonry through numerical models, would make it possible to predict, with close approximation, the response of masonry to different actions. The purpose of this paper is to evaluate the behavior of URM walls subjected to axial and horizontal load by using a finite element model with the Abaqus code. Constitutive laws of materials, bricks and mortar are defined, using two constitutive models of plasticity and damage included in the materials library. The constitutive models used employ information of experimental tests performed on bricks and mortar. The results of an analysis for an URM wall subjected to axial and horizontal load are presented. Responses of walls were obtained in terms of load versus displacement for different constitutive models and for different qualities of bricks and mortar. The two constitutive models used show similar results, particularly in the response waveforms. However, the ultimate capacity values show mean variations of around 30%. The two constitutive models display high dependence upon the mechanical characteristics of bricks and mortar.
