传统手工染色和现代仪器染色的对比和相关染色程序的改进研究

目的 对比传统手工染色和现代仪器染色在效率以及效果上的差异,分析改进染色程序策略。方法 随机选择广东省江门市中心医院2019年7月至2019年12月手术室、门诊标本照片,共计300例,分成手工组与仪器组,手工组标本照片采取传统手工染色,... 目的 对比传统手工染色和现代仪器染色在效率以及效果上的差异,分析改进染色程序策略。方法 随机选择广东省江门市中心医院2019年7月至2019年12月手术室、门诊标本照片,共计300例,分成手工组与仪器组,手工组标本照片采取传统手工染色,仪器组标本照片采取现代仪器染色,对比两组染色效率、染色质量情况以及醋酸化醇溶性伊红的使用时间情况,为染色程序改进提供可靠依据。结果 经过两组统计数据对比,现代仪器染色的仪器组标本照片染色质量与效率明显优于手工组,差异有统计学意义(P<0.05)。结论 利用现代仪器进行病理标本照片染色不仅能够提升染色效率、染色质量,也减少了原材料的使用,可基于现代仪器染色进行染色程序改进,减少脱水透明环节的乙醇与二甲苯环节,优化脱水透明程序,减少原材料消耗。 展开更多

On the basis of information theory and statistical methods, we use mutual information, n- tuple entropy and conditional entropy, combined with biological characteristics, to analyze the long range correlation and short range correlation in human Y chromosome palindromes. The magnitude distribution of the long range correlation which can be reflected by the mutual information is PS〉PSa〉PSb (P5a and P5b are the sequences that replace solely Alu repeats and all interspersed repeats with random uneorrelated sequences in human Y chromosome palindrome 5, respectively); and the magnitude distribution of the short range correlation which can be reflected by the n-tuple entropy and the conditional entropy is PS〉P5a〉PSb〉random uncorrelated sequence. In other words, when the Alu repeats and all interspersed repeats replace with random uneorrelated sequence, the long range and short range correlation decrease gradually. However, the random nncorrelated sequence has no correlation. This research indicates that more repeat sequences result in stronger correlation between bases in human Y chromosome. The analyses may be helpful to understand the special structures of human Y chromosome palindromes profoundly. 展开更多

Wool top and knitted fabric were treated with ammonia in the presence of sodium chloride (salt-ammonia process). The effectiveness of salt-ammonia process traditionally used as pretreatment for low temperature dyeing has been investigated for conferring shrink-resistance of wool. The pretreatmont of salt-ammonia process followed by enzymatic treatment has been proved that contribute to not only the enhancement of dyeing behavionr but also shrinkage-resistance in the present studies. A novel composition of salt-ammonia pretreatment, oxidized and reduced treatment as well as protease modification was recommended to achieve low temperature dyeing and shrinkage-resistance of wool. At the same time, the process conditions were optimized by orthogonal array and assessed by dye uptake rate, weight loss and area shrinkage. Laboratory experiments showed that the knitted fabric treated according to optimized conditions of the combined process, achieved not only improvement of dye abilities but also the strict requirement of machine-washable, representing a possible alternative to chlorination. 展开更多

Nuclear reprogramming: the zygotic transcription program is established through an“erase-and-rebuild” strategy 被引量:5

Oocytes display a maternal-specific gene expression profile, which is switched to a zygotic profile when a haploid set of chromatin is passed on to the fertilized egg that develops into an embryo. The mechanism underlying this transcription reprogramming is currently unknown. Here we demonstrate that by the time when transcription is shut down in germinal vesicle oocytes, a range of general transcription factors and transcriptional regulators are dissociated from the chromatin. The global dissociation of chromatin factors (CFs) disrupts physical contacts between the chromatin and CFs and leads to erasure of the maternal transcription program at the functional level. Critical transcription factors and regulators remain separated from chromatin for a prolonged period, and become re-associated with chromatin shortly after pronuclear formation. This is followed temporally by the re-establishment of nuclear functions such as DNA replication and transcription. We propose that the maternal transcription program is erased during oogenesis to generate a relatively naive chromatin and the zygotic transcription program is rebuilt de novo after fertilization. This process is termed as the "erase-and-rebuild" process, which is used to reset the transcription program, and most likely other nuclear processes as well, from a maternal one to that of the embryo. We further show in the accompanying paper (Gao T, et al., Cell Res 2007; 17:135-150.) that the same strategy is also employed to reprogram transcriptional profiles in somatic cell nuclear transfer and parthenogenesis, suggesting that this model is universally applicable to all forms of transcriptional reprogramming during early embryogenesis. Displacement of CFs from chromatin also offers an explanation for the phenomenon of transcription silence during the maternal to zygotic transition. 展开更多
