However, because DNA can exhibit marked differences among morphologically similar species, DNA barcoding offers a potential means for oyster identification. We analyzed the complete sequences of the cytochrome oxidase subunit I (COI) of five common Crassostrea species in China (including Hong Kong oyster C. hongkongensis, Jinjiang oyster C. ariakensis, Portuguese oyster C. angulata, Kumamoto oyster C. sikamea, and Pacific oyster C. gigas) and screened for distinct fragments. Using these distinct fragments on a high-resolution melting analysis platform, we developed an identification method that does not rely on species-specific PCR or fragment length polymorphism and is efficient, reliable, and easy to visualize. Using a single pair of primers (Oyster- COI-1), we were able to successfully distinguish among the five oyster species. Positive results have been achieved in submarine geological research, particularly in the investigation of marine gas hydrates. However, the amount of traveltime difference information is reduced for the velocity analysis under conditions of a shorter spread length, thus leading to poorer focusing of the velocity spectrum energy group and a lower accuracy of the velocity analysis. It is thus currently debatable whether the velocity analysis accuracy of short-arrangement multichannel seismic detection technology is able to meet the requirements of practical application in natural gas hydrate exploration. Therefore, in this study the bottom boundary of gas hydrates(Bottom Simulating Reflector, BSR) is used to conduct numerical simulation to discuss the accuracy of the velocity analysis related to such technology. Results show that a higher dominant frequency and smaller sampling interval are not only able to improve the seismic resolution, but they also compensate for the defects of the short-arrangement, thereby improving the accuracy of the velocity analysis. In conclusion, the accuracy of the velocity analysis in this small-scale, high-resolution, multi-channel seismic detection technology meets the requirements of natural gas hydrate exploration.展开更多

To quantify the mineral dissemination and pore space distribution of an ore particle, a cylindrical copper oxide ore sample (I center dot 4.6 mm x 5.6 mm) was scanned using high-resolution X-ray computed tomography (HRXCT), a nondestructive imaging technology, at a spatial resolution of 4.85 mu m. Combined with three-dimensional (3D) image analysis techniques, the main mineral phases and pore space were segmented and the volume fraction of each phase was calculated. In addition, the mass fraction of each mineral phase was estimated and the result was validated with that obtained using traditional techniques. Furthermore, the pore phase features, including the pore size distribution, pore surface area, pore fractal dimension, pore centerline, and the pore connectivity, were investigated quantitatively. The pore space analysis results indicate that the pore size distribution closely fits a log-normal distribution and that the pore space morphology is complicated, with a large surface area and low connectivity. This study demonstrates that the combination of HRXCT and 3D image analysis is an effective tool for acquiring 3D mineralogical and pore structural data.展开更多
In this paper, using the quasi-3D coupled current method, the influences of structure of cold crucible, the power frequency, the electricity property of melt, the coil position and current on the electromagnetic field...In this paper, using the quasi-3D coupled current method, the influences of structure of cold crucible, the power frequency, the electricity property of melt, the coil position and current on the electromagnetic field (EMF) and the levitation characteristics in the melting processes are analyzed. It is shown that in the processes of levitation melting with cold crucible, the power frequency and cold crucible structure are the decisive factors for the ability of magnetic flux penetrating into cold crucible. The magnetic flux density in cold crucible is reduced as the increasing of power frequency, and this tendency becomes stronger when the power frequency is higher than 100kHz. The segmented structure of cold crucible can reduce the induction eddy in itself effectively, and the higher the power frequency is, the better the result is. So, a cold crucible can be segmented into 16-20 sectors for high frequency electromagnetic field and/or 8-12 sectors for lower frequency one. It is also shown that the levitation force of melting charge is related to coil current as a parabolic function.展开更多

The thermodynam...Herein, a multi-index analysis of the nickel content of an alloy, output rate of the alloy, nickel recovery rate, and iron recovery rate during the melting of laterite metallized pellets was performed. The thermodynamic reduction behavior of oxides such as NiO, FeO, Fe_3 O_4, and Cr_2 O_3 was studied using the FactSage software, which revealed that SiO_2 is not conducive to the reduction of iron oxides, whereas the addition of basic oxides such as CaO and MgO is beneficial for the reduction of iron oxides. On the basis of a comprehensive analysis to achieve greater nickel recovery and lower iron recovery rates, the optimum experimental parameters in the orthogonal experiment were A3 B1 C3(t = 30 min, C/O = 0.4, R = 1.2); the indicators wNi, φalloy, ηNi, and ηFe had values of 15.0 wt%, 12.1%, 44.9%, and 96.4%, respectively. In single-factor experiments, increasing basicity(R) substantially improved the separation effect in the low-basicity range 0.5 ≤ R ≤ 0.8 but not in the high-basicity range 0.8 ≤ R ≤ 1.2. Similar results were obtained for the effect of the C/O ratio. Moreover, the recovery rate of nickel increased with increasing recovery rate of iron.展开更多
A 3D finite element model was established to investigate the temperature and stress fields during the selective laser melting process of Al−Mg−Sc−Zr alloy.By considering the powder−solid transformation,temperaturedepe...A 3D finite element model was established to investigate the temperature and stress fields during the selective laser melting process of Al−Mg−Sc−Zr alloy.By considering the powder−solid transformation,temperaturedependent thermal properties,latent heat of phase transformations and molten pool convection,the effects of laser power,point distance and hatch spacing on the temperature distribution,molten pool dimensions and residual stress distribution were investigated.Then,the effects of laser power,point distance and hatch spacing on the microstructure,density and hardness of the alloy were studied by the experimental method.The results show that the molten pool size gradually increases as the laser power increases and the point distance and hatch spacing decrease.The residual stress mainly concentrates in the middle of the first scanning track and the beginning and end of each scanning track.Experimental results demonstrate the accuracy of the model.The density of the samples tends to increase and then decrease with increasing laser power and decreasing point distance and hatch spacing.The optimum process parameters are laser power of 325−375 W,point distance of 80−100μm and hatch spacing of 80μm.展开更多

The model covers an initial 18-h spin-up, the 36-h rapid intensifica...A 72-h high-resolution simulation of Supertyphoon Rammasun (2014) is performed using the Advanced Research Weather Research and Forecasting model. The model covers an initial 18-h spin-up, the 36-h rapid intensification (RI) period in the northern South China Sea, and the 18-h period of weakening after landfall. The results show that the model reproduces the track, intensity, structure of the storm, and environmental circulations reasonably well. Analysis of the surface energetics under the storm indicates that the storm's intensification is closely related to the net energy gain rate (eg), defined as the difference between the energy production (PD) due to surface entropy flux and the energy dissipation (Ds) due to surface friction near the radius of maximum wind (RMW). Before and during the RI stage, the ~:g is high, indicating sufficient energy supply for the storm to intensify. However, the Sg decreases rapidly as the storm quickly intensifies, because the Ds increases more rapidly than the PD near the RMW. By the time the storm reaches its peak intensity, the Ds is about 20% larger than the PD near the RMW, leading to a local energetics deficit under the eyewall. During the mature stage, the PD and Ds can reach a balance within a radius of 86 km from the storm center (about 2.3 times the RMW). This implies that the local PD under the eyewall is not large enough to balance the Ds, and the radially inward energy transport from outside the eyewall must play an important role in maintaining the storm's intensity, as well as its intensification.展开更多

Calcium is easily brought into the widely ased brazing alloy, Ag-Cu-Zn, during the producing process. This paper aims at revealing the effect of calcium on the melting behavior of the brazing alloy. The thermal analysis kinetics of silver alloy with trace calcium was studied by using differential scanning calorimetry ( DSC ) , and the enthalpy peaks were analyzed by differential methods. The rate constant of phase transformation in the probable brazing temperature range goes up with increasing calcium content, according to the values of the apparent activation energy, E, and the frequeney constant, A. It is concluded that the calcium addition could improve the melting performance of Ag-Cu-Zn brazing alloy.展开更多

Background: Pyrazinamide (PZA) is one of the most important drugs for tuberculosis (TB) treatment, however, its susceptibility is not routinely tested. High-resolution melting (HRM) curve analysis has been widely used...Background: Pyrazinamide (PZA) is one of the most important drugs for tuberculosis (TB) treatment, however, its susceptibility is not routinely tested. High-resolution melting (HRM) curve analysis has been widely used for many applications. In this study, HRM assay was developed and evaluated for the detection of PZA resistance in Mycobacterium tuberculosis clinical isolates. Methods: Ninety five M. tuberculosis clinical isolates with different susceptibility patterns to anti-TB drugs were used to evaluate this assay. Isolates were phenotypically (Bactec MGIT 960) and genotypically (HRM and pncA gene sequencing) analysed for PZA resistance. Results: Bactec MGIT 960 analysis revealed that 29 of the 95 M. tuberculosis isolates were PZA resistant. In comparison to the Bactec MGIT 960, HRM showed a sensitivity of 47.7% and specificity of 74.6%, and the overall agreement between the two methods was 68.4%. Based on DNA sequencing, a correlation of 0.67 (significant at p-value pncA mutations was observed. PZA resistance was strongly associated with multi-drug resistant (MDR)-TB as it was shown in 79.3% of the MDR isolates included in the study. Conclusion: HRM is simple and useful for screening clinical M. tuberculosis isolates for PZA resistance, however, further modifications to improve its performance are required.展开更多

To ad...Wavelet transformation is a widely used method in high-frequency sequence stratigraphic analysis.However, the application is problematic since different wavelets always return the same sequence analysis results. To address this issue, we applied five commonly used wavelets to theoretical sequence models to document some application criteria. Five gradual scale-change sequence models were simplified from the glutenite succession deposition by gravity flows to form the fining-upwards cycle sequences(FUCS) and coarsening-upwards cycle sequences(CUCS). After conducting theoretical sequence model tests, the optimal wavelet(sym4) was selected and successfully used with actual data to identify the sequence boundaries. We also proposed a new method to optimize the scale of continuous wavelet transformation(CWT) for sequence boundary determination. We found that the balloon-like marks in scalograms of db4, sym4, and coif4 wavelet determine, respectively, the fourth-order sequence boundary, the thick succession sequence boundaries in FUCS, and the thick succession sequence in FUCS and CUCS. Comparing the sequence identification results shows that the asymmetric wavelets had an advantage in high-frequency sequence boundary determination and sedimentary cycle discrimination through the amplitude trend of the coefficient, in which the sym4 wavelet is the most effective. In conclusion, the asymmetry of wavelets is the first selection principle, of which asymmetric wavelets are more sensitive to sediment deposition by flood flows. The match of the wavelet between the sequence is the second selection principle, in which the correlation of time-frequency impacts the accuracy of sequence surface localization. However, the waveform of the wavelet is a visual and abstract parameter for sequence boundary detection. The appropriate wavelet for lacustrine sequence analysis is the asymmetric wavelet with a weak number of side lobes. The depositional flows, depositional process,and autogenic are three sedimentary factors that influence the sequence analysis results.展开更多
The melt onset dates(MOD)over Arctic sea ice plays an important role in the seasonal cycle of sea ice surface properties,which impacts Arctic surface solar radiation absorbed by the ice-ocean system.Monitoring interan...The melt onset dates(MOD)over Arctic sea ice plays an important role in the seasonal cycle of sea ice surface properties,which impacts Arctic surface solar radiation absorbed by the ice-ocean system.Monitoring interannual variations in MOD is valuable for understanding climate change.In this study,we investigated the spatio-temporal variability of MOD over Arctic sea ice and 14 Arctic sub-regions in the period of 1979 to 2017 from passive microwave satellite data.A set of mathematical and statistical methods,including the Sen’s slope and Mann-Kendall mutation tests,were used to comprehensively assess the variation trend and abrupt points of MOD during the past 39 years for different Arctic sub-regions.Additionally,the correlation between Arctic Oscillation(AO)and MOD was analyzed.The results indicate that:(1)all Arctic sub-regions show a trend toward earlier MOD except the Bering Sea and St.Lawrence Gulf.The East Siberian Sea exhibits a significantly earlier trend,with the highest rate of-9.45 d/decade;(2)the temporal variability and statistical significance of MOD trend exhibit large interannual differences with different time windows for most regions in the Arctic;(3)during the past 39 years,the MOD changed abruptly in different years for different sub-regions;(4)the seasonal AO has more influence on MOD than monthly AO.The findings in this study can improve our knowledge of MOD changes and are beneficial for further Arctic climate change study.展开更多

Current analytical techniques typically find difficulties distinguishing between closely related Prunus species which include almond (Prunus dulcis), an EU listed allergenic species. This study describes a proof of principle real-time PCR approach utilising DNA melt analyses that targets the internal transcribed spacer (ITS) sequence to differentiate between a panel of Prunus test species. The method was successfully applied to the characterisation of a commercial paprika sample suspected of having being adulterated with almond, referred to the UK Government Chemist in 2015 in its advisory capacity. Subject to further validation work, the method appears to specifically amplify Prunus species and is capable of discrimination based on the resultant melt profiles. The developed method provides analysts with a simple and broad molecular tool to identify common Prunus species for food authenticity and allergen testing purposes. Initial development work demonstrates a promising approach with the potential to improve discrimination between Prunus species not easily resolved by routine analytical methods.展开更多
In this work,the evolution of melt pool under single-point and single-line printing in the laser engineered net shaping(LENS)process is analyzed.Firstly,the basic structure of the melt pool model of the LENS process i...In this work,the evolution of melt pool under single-point and single-line printing in the laser engineered net shaping(LENS)process is analyzed.Firstly,the basic structure of the melt pool model of the LENS process is established and the necessary assumptions are made.Then,the establishment process of the multi-physical field model of the melt pool is introduced in detail.It is concluded that the simulation model results are highly consistent with the online measurement experiment results in terms of melt pool profile,space temperature gradient,and time temperature gradient.Meanwhile,some parameters,such as the 3D morphology and surface fluid field of the melt pool,which are not obtained in the online measurement experiment,are analyzed.Finally,the influence of changing the scanning speed on the profile,peak temperature,and temperature gradient of the single-line melt pool is also analyzed,and the following conclusions are obtained:With the increase in scanning speed,the profile of the melt pool gradually becomes slender;The relationship between peak temperature and scanning speed is approximately linear in a certain speed range;The space temperature gradient at the tail of the melt pool under different scanning speeds hardly changes with the scanning speed,and the time temperature gradient at the tail of the melt pool is in direct proportion to the scanning speed.展开更多

In this study, a finite-element a...Metamaterials have been receiving an increasing amount of interest in recent years. As a type of metamaterial, pentamode materials (PMs) approximate the elastic properties of liquids. In this study, a finite-element analysis was conducted to predict the mechanical properties of PM structures by altering the thin wall thicknesses and layer numbers to obtain an outstanding load-bearing capacity. It was found that as the thin wall thickness increased from 0.15 to 0.45 mm, the compressive modulus of the PM structures increased and the Poisson’s ratio decreased. As the layer number increased, the Poisson’s ratio of the PM structures increased rapidly and reaches a stable value ranging from 0.50 to 0.55. Simulation results of the stress distribution in the PM structures confirmed that stress concentrations exist at the junctions of the thin walls and weights. For validation, Ti–6Al–4V specimens were fabricated by selective laser melting (SLM), and the mechanical properties of these specimens (i.e., Poisson’s ratio and elastic modulus) were experimentally studied. Good consistency was achieved between the numerical and experimental results. This work is beneficial for the design and development of PM structures with simultaneous load-bearing capacity and pentamodal properties.展开更多

At proper imaging conditions the intensity peaks in the image have a constant spatial relationship with the projected atom columns. This allows the determination of the geometry of the projected unit cell without comparison with image simulations. The fast procedure is particularly suited for the analysis of large areas. The software package LADIA is written in the PV-WAVE code and provides all necessary tools for image processing and analysis. Image intensity peaks are determined by a cross-correlation technique, which avoids problems from noise in the low spatial frequency range. The lower limit of strain that can be detected at a sampling rate of 44 pixels/nm is≈2%.展开更多

AIM: To investigate the metabolic profiles of xenograft pancreatic cancer before and after radiotherapy by high-resolution magic angle spinning proton magnetic resonance spectroscopy (HRMAS 1H NMR) combined with princ...AIM: To investigate the metabolic profiles of xenograft pancreatic cancer before and after radiotherapy by high-resolution magic angle spinning proton magnetic resonance spectroscopy (HRMAS 1H NMR) combined with principal components analysis (PCA) and evaluate the radiotherapeutic effect. METHODS: The nude mouse xenograft model of human pancreatic cancer was established by injecting human pancreatic cancer cell SW1990 subcutaneously into the nude mice. When the tumors volume reached 800 mm3 , the mice received various radiation doses. Two weeks later, tumor tissue sections were prepared for running the NMR measurements. 1H NMR and PCA were used to determine the changes in the metabolic profiles of tumor tissues after radiotherapy. Metabolic profiles of normal pancreas, pancreatic tumor tissues, and radiationtreated pancreatic tumor tissues were compared. RESULTS: Compared with 1H NMR spectra of the normal nude mouse pancreas, the levels of choline, taurine, alanine, isoleucine, leucine, valine, lactate, and glutamic acid of the pancreatic cancer group were increased, whereas an opposite trend for phosphocholine, glycerophosphocholine, and betaine was observed. The ratio of phosphocholine to creatine, and glycerophosphocholine to creatine showed noticeable decrease in the pancreatic cancer group. After further evaluation of the tissue metabolic profile after treatment with three different radiation doses, no significant change in metabolites was observed in the 1H NMR spectra, while the inhibition of tumor growth was in proportion to the radiation doses. However, PCA results showed that the levels of choline and betaine were decreased with the increased radiation dose, and conversely, the level of acetic acid was dramatically increased. CONCLUSION: The combined methods were demonstrated to have the potential for allowing early diagnosis and assessment of pancreatic cancer response to radiotherapy.展开更多

To quantify the mineral dissemination and pore space distribution of an ore particle, a cylindrical copper oxide ore sample (I center dot 4.6 mm x 5.6 mm) was scanned using high-resolution X-ray computed tomography (HRXCT), a nondestructive imaging technology, at a spatial resolution of 4.85 mu m. Combined with three-dimensional (3D) image analysis techniques, the main mineral phases and pore space were segmented and the volume fraction of each phase was calculated. In addition, the mass fraction of each mineral phase was estimated and the result was validated with that obtained using traditional techniques. Furthermore, the pore phase features, including the pore size distribution, pore surface area, pore fractal dimension, pore centerline, and the pore connectivity, were investigated quantitatively. The pore space analysis results indicate that the pore size distribution closely fits a log-normal distribution and that the pore space morphology is complicated, with a large surface area and low connectivity. This study demonstrates that the combination of HRXCT and 3D image analysis is an effective tool for acquiring 3D mineralogical and pore structural data.
文摘In this paper, using the quasi-3D coupled current method, the influences of structure of cold crucible, the power frequency, the electricity property of melt, the coil position and current on the electromagnetic field (EMF) and the levitation characteristics in the melting processes are analyzed. It is shown that in the processes of levitation melting with cold crucible, the power frequency and cold crucible structure are the decisive factors for the ability of magnetic flux penetrating into cold crucible. The magnetic flux density in cold crucible is reduced as the increasing of power frequency, and this tendency becomes stronger when the power frequency is higher than 100kHz. The segmented structure of cold crucible can reduce the induction eddy in itself effectively, and the higher the power frequency is, the better the result is. So, a cold crucible can be segmented into 16-20 sectors for high frequency electromagnetic field and/or 8-12 sectors for lower frequency one. It is also shown that the levitation force of melting charge is related to coil current as a parabolic function.

文摘Herein, a multi-index analysis of the nickel content of an alloy, output rate of the alloy, nickel recovery rate, and iron recovery rate during the melting of laterite metallized pellets was performed. The thermodynamic reduction behavior of oxides such as NiO, FeO, Fe_3 O_4, and Cr_2 O_3 was studied using the FactSage software, which revealed that SiO_2 is not conducive to the reduction of iron oxides, whereas the addition of basic oxides such as CaO and MgO is beneficial for the reduction of iron oxides. On the basis of a comprehensive analysis to achieve greater nickel recovery and lower iron recovery rates, the optimum experimental parameters in the orthogonal experiment were A3 B1 C3(t = 30 min, C/O = 0.4, R = 1.2); the indicators wNi, φalloy, ηNi, and ηFe had values of 15.0 wt%, 12.1%, 44.9%, and 96.4%, respectively. In single-factor experiments, increasing basicity(R) substantially improved the separation effect in the low-basicity range 0.5 ≤ R ≤ 0.8 but not in the high-basicity range 0.8 ≤ R ≤ 1.2. Similar results were obtained for the effect of the C/O ratio. Moreover, the recovery rate of nickel increased with increasing recovery rate of iron.

文摘A 3D finite element model was established to investigate the temperature and stress fields during the selective laser melting process of Al−Mg−Sc−Zr alloy.By considering the powder−solid transformation,temperaturedependent thermal properties,latent heat of phase transformations and molten pool convection,the effects of laser power,point distance and hatch spacing on the temperature distribution,molten pool dimensions and residual stress distribution were investigated.Then,the effects of laser power,point distance and hatch spacing on the microstructure,density and hardness of the alloy were studied by the experimental method.The results show that the molten pool size gradually increases as the laser power increases and the point distance and hatch spacing decrease.The residual stress mainly concentrates in the middle of the first scanning track and the beginning and end of each scanning track.Experimental results demonstrate the accuracy of the model.The density of the samples tends to increase and then decrease with increasing laser power and decreasing point distance and hatch spacing.The optimum process parameters are laser power of 325−375 W,point distance of 80−100μm and hatch spacing of 80μm.

文摘A 72-h high-resolution simulation of Supertyphoon Rammasun (2014) is performed using the Advanced Research Weather Research and Forecasting model. The model covers an initial 18-h spin-up, the 36-h rapid intensification (RI) period in the northern South China Sea, and the 18-h period of weakening after landfall. The results show that the model reproduces the track, intensity, structure of the storm, and environmental circulations reasonably well. Analysis of the surface energetics under the storm indicates that the storm's intensification is closely related to the net energy gain rate (eg), defined as the difference between the energy production (PD) due to surface entropy flux and the energy dissipation (Ds) due to surface friction near the radius of maximum wind (RMW). Before and during the RI stage, the ~:g is high, indicating sufficient energy supply for the storm to intensify. However, the Sg decreases rapidly as the storm quickly intensifies, because the Ds increases more rapidly than the PD near the RMW. By the time the storm reaches its peak intensity, the Ds is about 20% larger than the PD near the RMW, leading to a local energetics deficit under the eyewall. During the mature stage, the PD and Ds can reach a balance within a radius of 86 km from the storm center (about 2.3 times the RMW). This implies that the local PD under the eyewall is not large enough to balance the Ds, and the radially inward energy transport from outside the eyewall must play an important role in maintaining the storm's intensity, as well as its intensification.
文摘The purity of the brazing alloys applied is necessary to be improved with the increasing cleanness of steel. Calcium is easily brought into the widely ased brazing alloy, Ag-Cu-Zn, during the producing process. This paper aims at revealing the effect of calcium on the melting behavior of the brazing alloy. The thermal analysis kinetics of silver alloy with trace calcium was studied by using differential scanning calorimetry ( DSC ) , and the enthalpy peaks were analyzed by differential methods. The rate constant of phase transformation in the probable brazing temperature range goes up with increasing calcium content, according to the values of the apparent activation energy, E, and the frequeney constant, A. It is concluded that the calcium addition could improve the melting performance of Ag-Cu-Zn brazing alloy.
文摘The sedimentary characteristics and their combination succession of the Permian in the Shandong nd Huainan-Huaibei coalfields are analyzed. The mid-and short-term stratigraphic base-level cycles are identified. High-resolution sequences are divided based on the above results. The study shows that the stratigraphic base-level cyclic method is an efficient way in the determination of the high-resolution sequences, especially in the classification of tbe terrestrial and transitional depositional succession.
文摘Background: Pyrazinamide (PZA) is one of the most important drugs for tuberculosis (TB) treatment, however, its susceptibility is not routinely tested. High-resolution melting (HRM) curve analysis has been widely used for many applications. In this study, HRM assay was developed and evaluated for the detection of PZA resistance in Mycobacterium tuberculosis clinical isolates. Methods: Ninety five M. tuberculosis clinical isolates with different susceptibility patterns to anti-TB drugs were used to evaluate this assay. Isolates were phenotypically (Bactec MGIT 960) and genotypically (HRM and pncA gene sequencing) analysed for PZA resistance. Results: Bactec MGIT 960 analysis revealed that 29 of the 95 M. tuberculosis isolates were PZA resistant. In comparison to the Bactec MGIT 960, HRM showed a sensitivity of 47.7% and specificity of 74.6%, and the overall agreement between the two methods was 68.4%. Based on DNA sequencing, a correlation of 0.67 (significant at p-value pncA mutations was observed. PZA resistance was strongly associated with multi-drug resistant (MDR)-TB as it was shown in 79.3% of the MDR isolates included in the study. Conclusion: HRM is simple and useful for screening clinical M. tuberculosis isolates for PZA resistance, however, further modifications to improve its performance are required.
文摘Wavelet transformation is a widely used method in high-frequency sequence stratigraphic analysis.However, the application is problematic since different wavelets always return the same sequence analysis results. To address this issue, we applied five commonly used wavelets to theoretical sequence models to document some application criteria. Five gradual scale-change sequence models were simplified from the glutenite succession deposition by gravity flows to form the fining-upwards cycle sequences(FUCS) and coarsening-upwards cycle sequences(CUCS). After conducting theoretical sequence model tests, the optimal wavelet(sym4) was selected and successfully used with actual data to identify the sequence boundaries. We also proposed a new method to optimize the scale of continuous wavelet transformation(CWT) for sequence boundary determination. We found that the balloon-like marks in scalograms of db4, sym4, and coif4 wavelet determine, respectively, the fourth-order sequence boundary, the thick succession sequence boundaries in FUCS, and the thick succession sequence in FUCS and CUCS. Comparing the sequence identification results shows that the asymmetric wavelets had an advantage in high-frequency sequence boundary determination and sedimentary cycle discrimination through the amplitude trend of the coefficient, in which the sym4 wavelet is the most effective. In conclusion, the asymmetry of wavelets is the first selection principle, of which asymmetric wavelets are more sensitive to sediment deposition by flood flows. The match of the wavelet between the sequence is the second selection principle, in which the correlation of time-frequency impacts the accuracy of sequence surface localization. However, the waveform of the wavelet is a visual and abstract parameter for sequence boundary detection. The appropriate wavelet for lacustrine sequence analysis is the asymmetric wavelet with a weak number of side lobes. The depositional flows, depositional process,and autogenic are three sedimentary factors that influence the sequence analysis results.

文摘The melt onset dates(MOD)over Arctic sea ice plays an important role in the seasonal cycle of sea ice surface properties,which impacts Arctic surface solar radiation absorbed by the ice-ocean system.Monitoring interannual variations in MOD is valuable for understanding climate change.In this study,we investigated the spatio-temporal variability of MOD over Arctic sea ice and 14 Arctic sub-regions in the period of 1979 to 2017 from passive microwave satellite data.A set of mathematical and statistical methods,including the Sen’s slope and Mann-Kendall mutation tests,were used to comprehensively assess the variation trend and abrupt points of MOD during the past 39 years for different Arctic sub-regions.Additionally,the correlation between Arctic Oscillation(AO)and MOD was analyzed.The results indicate that:(1)all Arctic sub-regions show a trend toward earlier MOD except the Bering Sea and St.Lawrence Gulf.The East Siberian Sea exhibits a significantly earlier trend,with the highest rate of-9.45 d/decade;(2)the temporal variability and statistical significance of MOD trend exhibit large interannual differences with different time windows for most regions in the Arctic;(3)during the past 39 years,the MOD changed abruptly in different years for different sub-regions;(4)the seasonal AO has more influence on MOD than monthly AO.The findings in this study can improve our knowledge of MOD changes and are beneficial for further Arctic climate change study.
文摘Food allergies represent a clear threat to the general health and wellbeing of those affected which place increasing pressure on food producers and regulatory authorities. Current analytical techniques typically find difficulties distinguishing between closely related Prunus species which include almond (Prunus dulcis), an EU listed allergenic species. This study describes a proof of principle real-time PCR approach utilising DNA melt analyses that targets the internal transcribed spacer (ITS) sequence to differentiate between a panel of Prunus test species. The method was successfully applied to the characterisation of a commercial paprika sample suspected of having being adulterated with almond, referred to the UK Government Chemist in 2015 in its advisory capacity. Subject to further validation work, the method appears to specifically amplify Prunus species and is capable of discrimination based on the resultant melt profiles. The developed method provides analysts with a simple and broad molecular tool to identify common Prunus species for food authenticity and allergen testing purposes. Initial development work demonstrates a promising approach with the potential to improve discrimination between Prunus species not easily resolved by routine analytical methods.

文摘In this work,the evolution of melt pool under single-point and single-line printing in the laser engineered net shaping(LENS)process is analyzed.Firstly,the basic structure of the melt pool model of the LENS process is established and the necessary assumptions are made.Then,the establishment process of the multi-physical field model of the melt pool is introduced in detail.It is concluded that the simulation model results are highly consistent with the online measurement experiment results in terms of melt pool profile,space temperature gradient,and time temperature gradient.Meanwhile,some parameters,such as the 3D morphology and surface fluid field of the melt pool,which are not obtained in the online measurement experiment,are analyzed.Finally,the influence of changing the scanning speed on the profile,peak temperature,and temperature gradient of the single-line melt pool is also analyzed,and the following conclusions are obtained:With the increase in scanning speed,the profile of the melt pool gradually becomes slender;The relationship between peak temperature and scanning speed is approximately linear in a certain speed range;The space temperature gradient at the tail of the melt pool under different scanning speeds hardly changes with the scanning speed,and the time temperature gradient at the tail of the melt pool is in direct proportion to the scanning speed.
文摘Metamaterials have been receiving an increasing amount of interest in recent years. As a type of metamaterial, pentamode materials (PMs) approximate the elastic properties of liquids. In this study, a finite-element analysis was conducted to predict the mechanical properties of PM structures by altering the thin wall thicknesses and layer numbers to obtain an outstanding load-bearing capacity. It was found that as the thin wall thickness increased from 0.15 to 0.45 mm, the compressive modulus of the PM structures increased and the Poisson’s ratio decreased. As the layer number increased, the Poisson’s ratio of the PM structures increased rapidly and reaches a stable value ranging from 0.50 to 0.55. Simulation results of the stress distribution in the PM structures confirmed that stress concentrations exist at the junctions of the thin walls and weights. For validation, Ti–6Al–4V specimens were fabricated by selective laser melting (SLM), and the mechanical properties of these specimens (i.e., Poisson’s ratio and elastic modulus) were experimentally studied. Good consistency was achieved between the numerical and experimental results. This work is beneficial for the design and development of PM structures with simultaneous load-bearing capacity and pentamodal properties.

文摘A thermal conduction model is applied to speed up the numerical analysis of the temperature distribution and the weld pool geometry of full penetration in gas tungsten arc (GTA) welding. With considering both top and bottom flee surface deformation of full-penetrated weld pool, three-dimensional weld pool with melting front and solidification front is predicted. Welding experiments are conducted to measure the melting front curves at the top surface and the longitudinal section of the weld. It shows that the predicted and measured results are in good agreement.
文摘Direct strain mapping from high resolution transmission electron microscopy images is possible for coherent structures. At proper imaging conditions the intensity peaks in the image have a constant spatial relationship with the projected atom columns. This allows the determination of the geometry of the projected unit cell without comparison with image simulations. The fast procedure is particularly suited for the analysis of large areas. The software package LADIA is written in the PV-WAVE code and provides all necessary tools for image processing and analysis. Image intensity peaks are determined by a cross-correlation technique, which avoids problems from noise in the low spatial frequency range. The lower limit of strain that can be detected at a sampling rate of 44 pixels/nm is≈2%.

文摘Based on the results obtained from Pg wavefront imaging in active source deep seismic sounding, we propose a new ray hit analysis method for high-resolution seismic refraction profile data processing. This method can be used to further determine possible refraction interface, especially spatial location of basement and its pattern characteristics in complex upper crustal structure region, making data processing for high-resolution refraction profiles more fine. We use this method to study the crystalline basement structure of east part of A'nyemaqên suite zone at northeast side of Qinghai-Xizang Plateau and the basement patterns as well as its spreading features at the east part of Anemaqen suite zone and its adjacent region were determined.

文摘AIM: To investigate the metabolic profiles of xenograft pancreatic cancer before and after radiotherapy by high-resolution magic angle spinning proton magnetic resonance spectroscopy (HRMAS 1H NMR) combined with principal components analysis (PCA) and evaluate the radiotherapeutic effect. METHODS: The nude mouse xenograft model of human pancreatic cancer was established by injecting human pancreatic cancer cell SW1990 subcutaneously into the nude mice. When the tumors volume reached 800 mm3 , the mice received various radiation doses. Two weeks later, tumor tissue sections were prepared for running the NMR measurements. 1H NMR and PCA were used to determine the changes in the metabolic profiles of tumor tissues after radiotherapy. Metabolic profiles of normal pancreas, pancreatic tumor tissues, and radiationtreated pancreatic tumor tissues were compared. RESULTS: Compared with 1H NMR spectra of the normal nude mouse pancreas, the levels of choline, taurine, alanine, isoleucine, leucine, valine, lactate, and glutamic acid of the pancreatic cancer group were increased, whereas an opposite trend for phosphocholine, glycerophosphocholine, and betaine was observed. The ratio of phosphocholine to creatine, and glycerophosphocholine to creatine showed noticeable decrease in the pancreatic cancer group. After further evaluation of the tissue metabolic profile after treatment with three different radiation doses, no significant change in metabolites was observed in the 1H NMR spectra, while the inhibition of tumor growth was in proportion to the radiation doses. However, PCA results showed that the levels of choline and betaine were decreased with the increased radiation dose, and conversely, the level of acetic acid was dramatically increased. CONCLUSION: The combined methods were demonstrated to have the potential for allowing early diagnosis and assessment of pancreatic cancer response to radiotherapy.
