The SYD-2::GFP phenotypes of these mutants are mainly reflected as the changes of number, morphology, distribution of puncta and the gaps appearance. Some mutants also exhibit visible behavioral or physical phenotypes, and aldicarb resistant or sensitive phenotypes. Conclusion These mutants provide the opportunity for further systematic research on the active zone formation and the neurotransmission.展开更多
This study aims to further enhance the oil recovery of reservoirs in the Zhong-2 Block of the Gudao Oilfield by identifying the most effective microbial-flooding activator systems and applying them in the field.We beg...This study aims to further enhance the oil recovery of reservoirs in the Zhong-2 Block of the Gudao Oilfield by identifying the most effective microbial-flooding activator systems and applying them in the field.We began by analyzing the structure of the reservoirs'endogenous microbial communities to understand the potential impact of microbial flooding.This was followed by determining commonly used activator systems based on their abilities to stimulate oil-displacement functional bacteria.Through laboratory experiments on oil displacement efficiency and sweep characteristics,we determined the optimal activator injection method(injection ratio)and the requisite bacterial concentration for maximal microbial-flooding efficacy.Finally,we selected the optimal activator systems and applied them to field tests.Our findings suggest the target block is highly receptive to microbial-flooding.In terms of performance,the activator systems ranked as No.3>No.4>No.1>No.2.Interestingly,a deep activator system,when compared to the top-performing No.3 system,exhibited a higher bacterial concentration peak and longer peaking duration.Optimal oil displacement effects were observed at a 1:4 vol ratio between the No.3 activator and deep activator systems,with bacterial concentrations of up to 106 cells/mL or above.Field tests with the selected activator systems,following a specific injection protocol,demonstrated a notable increase in oil production and a reduction in water cut.展开更多

Experimental results showed that the metallurgical characteristics and hardening effect of 722M24 steel nitrided by ASPN at both floating potential and anodic (zero) potential were similar to those nitrided by DCPN. XRD and high-resolution SEM analysis indicated that iron nitride particles with sizes in sub-micron scale were deposited on the specimen surface in AS plasma nitriding. These indicate that the neutral iron nitride particles, which are sputtered from the active screen and transferred through plasma to specimen surface, are considered to be the dominant nitrogen carrier in ASPN. The OES results show that NH could not be a critical species in plasma nitriding.展开更多

Active case finding among vulnerable groups has been the key focus area of National Strategic Plan to control TB in India 2017-25. This cross-sectional study was carried out to find out the additional open cases of TB through active screening of general & rural population in selected villages of Jaipur, the capital district in Rajasthan state of India. An active screening team of trained staff visited each house-hold in selected study villages for interviewing the head of house-hold or other adult member available at the time of visit. Family members of age ≥</span><span><span><span style="font-family:;" "=""> </span></span></span><span><span><span style="font-family:;" "=""><span style="font-family:Verdana;">15 years were screened for complain of chronic cough of duration 2 weeks or more with study questionnaire. [ Method] Dilu- tion plate method and plate confrontation culture method were used to isolate and screen biocontrol bacteria from the rhizosphere soil of diseased ginseng. The strains were identified through morphology, physiological and biochemical characteristics and 16S rDNA. [ Result ] With Rhizoctonia solani, Fusarium oxysporum and Fu- sarium solani as the indicator strains, two biocontrol strains B59 and X1 with strong antagonistic effects were screened from the rhizosphere soil of diseased ginseng in Tieli farm of Heilongjiang Province, and they were identified to be Bacillus subtilis. The inhibition rates of two biocontrol strains against eight different fungi were all greater than 90%. The primary study indicated that B59 and X1 strains could secrete antifungal active substances. [ Conclusion] Two biocontrol Bacillus subti- lis strains 1359 and X1 all had strong antagonistic effect against ginseng soil-borne disease, which had certain potential for development and utilization.展开更多

With Candida albicans as the model, the secondary metabolites were carried out high-throughput screening. [ Result ] A strain of myxohacteria numbered ZJ2 was obtained after secondary screening, its secondary metabolites had activities against G. albicans. 16S rDNA cloning and sequencing results showed that ZJ2 was Myxococcus fidvus. [ Conclusion ] The secondary metabolites of ZJ2 strain had bioactive substances with significant inhibi- tion effect against the growth of C. albicans, which had potential pharmaceutical research and development value.展开更多

Although there is limited evidence that adolescents transfer PA f...Background: Given the low levels of physical activity(PA) among adolescent girls in the US, there is a need to identify tools to motivate increased PA. Although there is limited evidence that adolescents transfer PA from one context to another, exergames(i.e., video games that require gross motor activity) may act as a gateway to promote overall PA outside game play. The purpose of this study was to examine potential transfer effects(i.e., influences on external behaviors and psychological constructs) of a 12-week exergaming intervention on adolescent girls' PA, screen time,and self-efficacy toward PA, as well as the intrinsic motivation of exergaming.Methods: Participants were 37 girls aged 14–18 years(65% African American, 35% white) who were overweight or obese(body mass index ≥ 85 th percentile) and were recruited from the community via school, physicians, news media, and social media websites. Adolescents were randomly assigned to a 12-week group exergaming intervention(thirty-six 60 min sessions of group-based dance exergaming in a research laboratory using Kinect for Xbox360(Microsoft Corporation, Redmond, WA, USA)) or to a no-treatment control group. Outcome variables included objectively measured PA(total) and self-reported leisure-time PA(discretionary time only) 1 week before vs. 1 week after the intervention; selected type and intensity of PA when placed in a gym setting for 30 min("cardio free choice"); screen time; self-efficacy toward PA; and intrinsic motivation toward exergaming.Results: Attendance at the exergaming sessions was high(80%). Compared with the control group, the intervention group self-reported an increase in PA(p = 0.035) and fewer hours watching television or videos(p = 0.01) after the intervention, but there were no significant differences in sedentary, light, moderate, or vigorous PA measured by accelerometry. The intervention group significantly improved self-efficacy toward PA(p = 0.028). The intervention group highly rated intrinsic motivation toward exergaming.Conclusion: Exergaming for 12 weeks was associated with positive impacts on adolescent girls' self-reported PA, television viewing, self-efficacy,and intrinsic motivation. Future research is warranted to leverage exergames as an enjoyable, motivating, and effective PA tool.展开更多

Methods: We performed a cross-sectional survey to assess health risks from a representative sample of the population ≥3 years old in the Delta Region of Arkansas, Louisiana, and Mississippi. Linear models were utilized to examine Pediatric Quality of Life Inventory (PedsQL) scores based on reported PA and ST. Results: Median age of the 371 participants was 9.8 years, 57% were African American, and 26% had annual household income ≤$14,999. Overall, 262 (75%) were categorized as physically active and 48% reported >2 hours ST per day. Children reporting exercising 5 - 7 days per week had significantly better PedsQL total score (84.3 vs 80.8;p 2 hours per day) did not significantly impact HRQOL. Conclusions: Findings suggest that PA among children living in the Mississippi Delta was related to higher HRQOL;however, the positive effects of PA on HRQOL may be less for minority and low-income children.展开更多

Crude methanolic and water extracts of 19 marine algal species (6 Chlorophyta, 8 Phaeophyta and 5 Rhodophyta) collected from the western coast of Libya were evaluated for antibacterial activity against pathogenic bacteria (4 Gram-positive, 4 Gram-nega- tive). The extracts showed a significant antibacterial activity against Gram positive (Staphylococcus aureus, Bacillus subtilis, Bacillus spp., and Staphylococcus epidermidis) as well as Gram negative bacteria (Escherichia coli, Salmonella typhi, Klebsiella spp., and Pseudomonas aerugi- nosa). The algal aqueous and methanolic extracts displayed different degrees of antimicrobial activities against different bacteria, in some cases methanolic extracts showed higher antibacterial activity than aqueous extracts. Among tested algae, Brown algae namely Cystoseira crinite exhibited the highest antibacterial activity among tested algal species.展开更多
Purpose: To examine the associations among preschoolers fundamental motor skills, screen-time, physical activity(PA), and sedentary behavior(SB).Methods: Children ages 3à4 years were enrolled in a prospective obs...Purpose: To examine the associations among preschoolers fundamental motor skills, screen-time, physical activity(PA), and sedentary behavior(SB).Methods: Children ages 3à4 years were enrolled in a prospective observational trial of PA. Trained assessors conducted the Test of Gross Motor Development-3 rd edition(TGMD-3), and the Movement Assessment Battery for Children-2 nd edition, and parent-reported child screen-time and sociodemographic information. Children wore an accelerometer for 7 days to examine SB and total PA(TPA). TPA was further characterized as moderateto-vigorous PA(MVPA) or vigorous PA(VPA). Mixed linear models were calculated, controlling for age(for TGMD-3), sex, household income, and accelerometer wear time(for accelerometry models), with childcare center as a random effect. The primary analysis reported on the cross-sectional baseline data of 126 children with complete fundamental motor skill and screen-time data; a subanalysis included 88 children with complete accelerometry data.Results: Children were 3.4 § 0.5 years of age(54% girls; 46% white, 42% African American, 12% other). A total of 48% lived in households at or below the federal poverty level. Children engaged in 5.1 § 3.6 h/day of screen-time. Children's screen-time was inversely related to the Movement Assessment Battery for Children-2 nd edition, manual dexterity skills percentile(b(SE) = ?1.7(0.8), p = 0.049). In the accelerometry subsample,children engaged in 5.9 § 0.9 h/day of TPA of which 1.7 § 0.6 h/day was MVPA. Boys engaged in more MVPA and VPA and less SB compared with girls(all p < 0.05). A higher TGMD-3, total score(b(SE) = 0.4(0.2), p = 0.017) and locomotor score(b(SE) = 0.7(0.3), p = 0.018) were associated with more VPA but not with TPA or MVPA. Screen-time and television in the bedroom were not related to SB, TPA, MVPA, or VPA.Conclusion: Children's motor skills were positively related to VPA but inversely related to screen-time. Further inquiry into the implications of high exposure to screen-time in young children is needed.展开更多

To explore the feasibility of pharmacological correction of disrupted activation of CFTR chloride channel caused by G551D mutation, we developed a halide-sensitive fluorescence miniassay for G551D-CFTR in Fisher rat thyroid(FRT) epithelial cells for the discovery of novel activators of G551D-CFTR. A class of bicyclooctane small molecule compounds that efficiently stimulate G551D-CFTR chloride channel activity was identified by high throughput screening via the FRT cell-based assay. This class of compounds selectively activates G551D-CFTR with a high affinity, whereas little effect of the compounds on wildtype CFTR can be seen. The discovery of a class of bicyclooctane G551D-CFTR activators will permit the analysis of structure-activity relationship of the compounds to identify ideal leads for in vivo therapeutic studies.展开更多

Medicinal plants, rich in bioactive components that promote prevention and treatment, are inexpensive and no side effects. <i>Apium graveolens</i>, <i>Cleome gynandra</i>, and <i>Hibiscus sabdariffa</i> from Burkina Faso were investigated for their phytochemical profile and antioxidant activities. The high-performance thin-layer chromatography profile revealed flavonoids, tannins, and sterols in these herbaceous. The <i>Hibiscus sabdariffa</i> methanolic extract exhibited the highest total phenolic (138.4 ± 0.5 mg GAE/g DW) and flavonoid (52.8 ± 0.6 mg RuE/g DW) contents comparatively to <i>Cleome gynandra</i> and <i>Apium graveolens</i>. <i>Hibiscus sabdariffa</i> methanolic extract also presented the highest antioxidant activity (IC<sub>50</sub> = 0.31 ± 0.002 mg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay. A high correlation between flavonoid contents and hydrophilic antioxidant activities (r = 0.99) was observed, indicating that flavonoids contribute significatively to these herbaceous antioxidant properties. <i>Apium graveolens</i>, <i>Cleome gynandra, and <i>Hibiscus sabdariffa</i> constitute a natural source of phenolic compounds that could be exploited in diabetes mellitus management.</i>展开更多

Using bioinformatics approaches, this study aimed to identify antileishmanial activity in drugs that are currently used as anti-inflammatory and wound healing by such anti-Leishmania activity was validated by in vitro and in vivo assays. In silico analysis identified 153 compounds from which 87 were selected by data mining of DrugBank database, 22 and 44 were detected by PASS (http://pass.cribi.unipd.it) and BLAST (http://blast.ncbi.nlm.nih. gov/) alignment, respectively. The majority of identified drugs are used as skin protector, anti-acne, anti-ulcerative (wound healer) or anti-inflammatory and few of them had specific antileishmanial activity. The efficacy as antileishmanial was validated in vitro in 12/23 tested compounds and in all seven compounds that were evaluated in in vivo assays. Notably, this is the first report of antileishmanial activity for adapalene. In conclusion, bioinformatics tools not only can help to reduce time and cost of the drug discovery process but also may increase the chance that candidates identified in silico which have a validated antileishmanial activity by combining different biological properties.展开更多
Little is known on the association between movement behaviors including physical activity(PA),screen time(ST)and sleep(SLP)with depression in adolescents.This study aimed to explore the associations of PA,ST and SLP w...Little is known on the association between movement behaviors including physical activity(PA),screen time(ST)and sleep(SLP)with depression in adolescents.This study aimed to explore the associations of PA,ST and SLP with depressive symptoms in adolescents.A total of 1,331 middle school students participated in this survey and provided valid data pertaining to the study variables.Self-reported questionnaires were used to collect information on participants’sociodemographic parameters.The Health Behavior in School-aged Children Questionnaire was used to assess the PA(days for moderate to vigorous PA),ST(daily hours of ST)and SLP(daily hours of SLP).Study participants’depressive symptoms were assessed using the Children’s Depression Inventory.Generalized linear models were used to estimate the associations of PA,ST and SLP with depressive symptoms.Of all the 1331 study participants,boys accounted for 51.31%and percentage of 7th,8th and 9th graders were 52.44%,23.22%and 24.34%,respectively.After controlling for sex,body mass index,grade,ethnicity,residence,siblings,perceived family affluence,father and mother educational level,only ST was positively associated with depressive symptoms(beta=0.17,p=0.005).This study suggests that excessive ST would be detrimental to depressive symptoms in adolescents.It may be effective to reduce ST for depression intervention or prevention.Future studies are encouraged to use an improved study design to confirm or negate this study’s researchfindings.展开更多

Phytochemical screening of the diethyl ether, methanol and aqueous extracts of seeds confirmed the presence of alkaloids, saponins, sterols and triterpenes, carotenoids, coumarins, tannins and carbohydrates. Chloroform extract was fractionated by sing liquid solid column chromatographic technique and column fractions eluted with chloroform and methanol. The antibacterial secondary metabolites were achieved by fractionation of the active. 文摘This study aims to further enhance the oil recovery of reservoirs in the Zhong-2 Block of the Gudao Oilfield by identifying the most effective microbial-flooding activator systems and applying them in the field.We began by analyzing the structure of the reservoirs'endogenous microbial communities to understand the potential impact of microbial flooding.This was followed by determining commonly used activator systems based on their abilities to stimulate oil-displacement functional bacteria.Through laboratory experiments on oil displacement efficiency and sweep characteristics,we determined the optimal activator injection method(injection ratio)and the requisite bacterial concentration for maximal microbial-flooding efficacy.Finally,we selected the optimal activator systems and applied them to field tests.Our findings suggest the target block is highly receptive to microbial-flooding.In terms of performance,the activator systems ranked as No.3>No.4>No.1>No.2.Interestingly,a deep activator system,when compared to the top-performing No.3 system,exhibited a higher bacterial concentration peak and longer peaking duration.Optimal oil displacement effects were observed at a 1:4 vol ratio between the No.3 activator and deep activator systems,with bacterial concentrations of up to 106 cells/mL or above.Field tests with the selected activator systems,following a specific injection protocol,demonstrated a notable increase in oil production and a reduction in water cut.

文摘[Objectives]The anti-tumor activity of fractions from Buddleja officinalis Maxim.by petroleum ether,ethyl acetate,n-butanol and water solvent was studied.[Methods]The ethanol extract from B.officinalis Maxim.was extracted and then concentrated with petroleum ether,ethyl acetate,n-butanol and water,respectively,and the extracts were obtained.The inhibitory effects of the four different fractions on the growth of three tumor cell lines in vitro were detected by CCK-8 method,and the median inhibitory concentration(IC 50 value)was calculated.[Results]The four fractions inhibited the growth of the three tumor cell lines in vitro,among which the n-butanol fraction had the best anti-tumor activity.The IC 50 values of the n-butanol fraction on human gastric cancer(SGC-7901),human breast cancer(MCF-7)and human liver cancer(BEL-7404)cell lines were 0.08,1.58 and 0.12 mg/mL,respectively.[Conclusions]Petroleum ether,ethyl acetate,n-butanol and water fractions from the ethanol extract of B.officinalis Maxim.had certain anti-tumor effects,and the n-butanol fraction had the best anti-tumor activity.
文摘Active screen plasma nitriding (ASPN) is a novel nitriding process, which overcomes many of the practical problems associated with the conventional DC plasma nitriding (DCPN). Experimental results showed that the metallurgical characteristics and hardening effect of 722M24 steel nitrided by ASPN at both floating potential and anodic (zero) potential were similar to those nitrided by DCPN. XRD and high-resolution SEM analysis indicated that iron nitride particles with sizes in sub-micron scale were deposited on the specimen surface in AS plasma nitriding. These indicate that the neutral iron nitride particles, which are sputtered from the active screen and transferred through plasma to specimen surface, are considered to be the dominant nitrogen carrier in ASPN. The OES results show that NH could not be a critical species in plasma nitriding.

Active case finding among vulnerable groups has been the key focus area of National Strategic Plan to control TB in India 2017-25. This cross-sectional study was carried out to find out the additional open cases of TB through active screening of general & rural population in selected villages of Jaipur, the capital district in Rajasthan state of India. An active screening team of trained staff visited each house-hold in selected study villages for interviewing the head of house-hold or other adult member available at the time of visit. Family members of age ≥</span><span><span><span style="font-family:;" "=""> </span></span></span><span><span><span style="font-family:;" "=""><span style="font-family:Verdana;">15 years were screened for complain of chronic cough of duration 2 weeks or more with study questionnaire. Presumptive cases found at screening were referred for seeking medical consul</span><span style="font-family:Verdana;">tations from nearest designated microscopy centre. The referred patients w</span><span style="font-family:Verdana;">ere followed up on telephone up to five times for results of medical consultancies. Laboratory technicians of catchment DMCs were also followed up for verification of results. Total 19,306 individuals were covered. Study included 52.8% male, 69.3% ≥ 15 years age, and 99.3% Hindu participants. There were total 153 presumptive TB patients. Out of these, total 10 cases were confirmed as </span><span style="font-family:Verdana;">TB. The yield of detection was 0.51 cases/1000 population screened. 文摘[ Objective] The paper was to obtain biocontrol strains with good control effects against ginseng soil-borne disease through screening. [ Method] Dilu- tion plate method and plate confrontation culture method were used to isolate and screen biocontrol bacteria from the rhizosphere soil of diseased ginseng. The strains were identified through morphology, physiological and biochemical characteristics and 16S rDNA. [ Result ] With Rhizoctonia solani, Fusarium oxysporum and Fu- sarium solani as the indicator strains, two biocontrol strains B59 and X1 with strong antagonistic effects were screened from the rhizosphere soil of diseased ginseng in Tieli farm of Heilongjiang Province, and they were identified to be Bacillus subtilis. The inhibition rates of two biocontrol strains against eight different fungi were all greater than 90%. The primary study indicated that B59 and X1 strains could secrete antifungal active substances. [ Conclusion] Two biocontrol Bacillus subti- lis strains 1359 and X1 all had strong antagonistic effect against ginseng soil-borne disease, which had certain potential for development and utilization.

文摘[ Objective] The paper was to screen myxobacteria with antifungal activity, and identify its species. [ Method] 237 myxobacteria were isolated and pu- rified from 200 copies of soil samples collected from 23 provinces in China. The strains were fermented in four different media, and the fermentation products were extracted by methanol to obtain metabolites. With Candida albicans as the model, the secondary metabolites were carried out high-throughput screening. [ Result ] A strain of myxohacteria numbered ZJ2 was obtained after secondary screening, its secondary metabolites had activities against G. albicans. 16S rDNA cloning and sequencing results showed that ZJ2 was Myxococcus fidvus. [ Conclusion ] The secondary metabolites of ZJ2 strain had bioactive substances with significant inhibi- tion effect against the growth of C. albicans, which had potential pharmaceutical research and development value.

文摘Background: Given the low levels of physical activity(PA) among adolescent girls in the US, there is a need to identify tools to motivate increased PA. Although there is limited evidence that adolescents transfer PA from one context to another, exergames(i.e., video games that require gross motor activity) may act as a gateway to promote overall PA outside game play. The purpose of this study was to examine potential transfer effects(i.e., influences on external behaviors and psychological constructs) of a 12-week exergaming intervention on adolescent girls' PA, screen time,and self-efficacy toward PA, as well as the intrinsic motivation of exergaming.Methods: Participants were 37 girls aged 14–18 years(65% African American, 35% white) who were overweight or obese(body mass index ≥ 85 th percentile) and were recruited from the community via school, physicians, news media, and social media websites. Adolescents were randomly assigned to a 12-week group exergaming intervention(thirty-six 60 min sessions of group-based dance exergaming in a research laboratory using Kinect for Xbox360(Microsoft Corporation, Redmond, WA, USA)) or to a no-treatment control group. Outcome variables included objectively measured PA(total) and self-reported leisure-time PA(discretionary time only) 1 week before vs. 1 week after the intervention; selected type and intensity of PA when placed in a gym setting for 30 min("cardio free choice"); screen time; self-efficacy toward PA; and intrinsic motivation toward exergaming.Results: Attendance at the exergaming sessions was high(80%). Compared with the control group, the intervention group self-reported an increase in PA(p = 0.035) and fewer hours watching television or videos(p = 0.01) after the intervention, but there were no significant differences in sedentary, light, moderate, or vigorous PA measured by accelerometry. The intervention group significantly improved self-efficacy toward PA(p = 0.028). The intervention group highly rated intrinsic motivation toward exergaming.Conclusion: Exergaming for 12 weeks was associated with positive impacts on adolescent girls' self-reported PA, television viewing, self-efficacy,and intrinsic motivation. Future research is warranted to leverage exergames as an enjoyable, motivating, and effective PA tool.
文摘Background: The association of physical activity (PA) and screen time (ST) on health-related quality of life (HRQOL) among predominantly low-income children in the Mississippi Delta region has not been previously described. Methods: We performed a cross-sectional survey to assess health risks from a representative sample of the population ≥3 years old in the Delta Region of Arkansas, Louisiana, and Mississippi. Linear models were utilized to examine Pediatric Quality of Life Inventory (PedsQL) scores based on reported PA and ST. Results: Median age of the 371 participants was 9.8 years, 57% were African American, and 26% had annual household income ≤$14,999. Overall, 262 (75%) were categorized as physically active and 48% reported >2 hours ST per day. Children reporting exercising 5 - 7 days per week had significantly better PedsQL total score (84.3 vs 80.8;p 2 hours per day) did not significantly impact HRQOL. Conclusions: Findings suggest that PA among children living in the Mississippi Delta was related to higher HRQOL;however, the positive effects of PA on HRQOL may be less for minority and low-income children.
文摘Marine macroalgae are considered as an excellent source of bioactive compounds which has a broad range of biological activities including antibacterial and antioxidant. Crude methanolic and water extracts of 19 marine algal species (6 Chlorophyta, 8 Phaeophyta and 5 Rhodophyta) collected from the western coast of Libya were evaluated for antibacterial activity against pathogenic bacteria (4 Gram-positive, 4 Gram-nega- tive). The extracts showed a significant antibacterial activity against Gram positive (Staphylococcus aureus, Bacillus subtilis, Bacillus spp., and Staphylococcus epidermidis) as well as Gram negative bacteria (Escherichia coli, Salmonella typhi, Klebsiella spp., and Pseudomonas aerugi- nosa). The algal aqueous and methanolic extracts displayed different degrees of antimicrobial activities against different bacteria, in some cases methanolic extracts showed higher antibacterial activity than aqueous extracts. Among tested algae, Brown algae namely Cystoseira crinite exhibited the highest antibacterial activity among tested algal species.

文摘Purpose: To examine the associations among preschoolers fundamental motor skills, screen-time, physical activity(PA), and sedentary behavior(SB).Methods: Children ages 3à4 years were enrolled in a prospective observational trial of PA. Trained assessors conducted the Test of Gross Motor Development-3 rd edition(TGMD-3), and the Movement Assessment Battery for Children-2 nd edition, and parent-reported child screen-time and sociodemographic information. Children wore an accelerometer for 7 days to examine SB and total PA(TPA). TPA was further characterized as moderateto-vigorous PA(MVPA) or vigorous PA(VPA). Mixed linear models were calculated, controlling for age(for TGMD-3), sex, household income, and accelerometer wear time(for accelerometry models), with childcare center as a random effect. The primary analysis reported on the cross-sectional baseline data of 126 children with complete fundamental motor skill and screen-time data; a subanalysis included 88 children with complete accelerometry data.Results: Children were 3.4 § 0.5 years of age(54% girls; 46% white, 42% African American, 12% other). A total of 48% lived in households at or below the federal poverty level. Children engaged in 5.1 § 3.6 h/day of screen-time. Children's screen-time was inversely related to the Movement Assessment Battery for Children-2 nd edition, manual dexterity skills percentile(b(SE) = ?1.7(0.8), p = 0.049). In the accelerometry subsample,children engaged in 5.9 § 0.9 h/day of TPA of which 1.7 § 0.6 h/day was MVPA. Boys engaged in more MVPA and VPA and less SB compared with girls(all p < 0.05). A higher TGMD-3, total score(b(SE) = 0.4(0.2), p = 0.017) and locomotor score(b(SE) = 0.7(0.3), p = 0.018) were associated with more VPA but not with TPA or MVPA. Screen-time and television in the bedroom were not related to SB, TPA, MVPA, or VPA.Conclusion: Children's motor skills were positively related to VPA but inversely related to screen-time. Further inquiry into the implications of high exposure to screen-time in young children is needed.

文摘Objective:To evaluate the antioxidant activity of extracts of dried flowers of Tithonia diversifolia(Hemsl) A.Gray(T.diversifolia) dry flower-a slirubby plant belonging to the Asteraceac family and very common in Brazil,providing data to help prevent premature aging skin.Methods:The tests of phytochemical screening included total phenols,tannins,flavonoids,alkaloids and saponins.The active antioxidant was determined by 2,2-diphenyl-1-picrylhydrazyl method.Results:The phytoehemical screening of T.diversifolia dry flowers revealed the presence of phenolic compounds(tannins,flavonoids and total phenolsi,while alkaloids and saponins were not detected.The IC_(50) values showed a strong antioxidant activity of the plant extracts.Conclusions:Therefore,this study suggests the possibility of using dry flowers extracts of T.diversifolia for the prevention of cell aging,as was shown to have significant antioxidant activity.

文摘The glycine-to-aspartic acid missense mutation at the codon 551(G551D) of the cystic fibrosis transmembrane conductance regulator(CFTR) is one of the five most frequent cystic fibrosis(CF) mutations associated with a severe CF phenotype. To explore the feasibility of pharmacological correction of disrupted activation of CFTR chloride channel caused by G551D mutation, we developed a halide-sensitive fluorescence miniassay for G551D-CFTR in Fisher rat thyroid(FRT) epithelial cells for the discovery of novel activators of G551D-CFTR. A class of bicyclooctane small molecule compounds that efficiently stimulate G551D-CFTR chloride channel activity was identified by high throughput screening via the FRT cell-based assay. This class of compounds selectively activates G551D-CFTR with a high affinity, whereas little effect of the compounds on wildtype CFTR can be seen. The discovery of a class of bicyclooctane G551D-CFTR activators will permit the analysis of structure-activity relationship of the compounds to identify ideal leads for in vivo therapeutic studies.
文摘Diabetes mellitus is a socially significant disease characterized by chronic hyperglycemia and metabolic disorders of proteins, carbohydrates, and lipids due to reduced function of insulin. Medicinal plants, rich in bioactive components that promote prevention and treatment, are inexpensive and no side effects. <i>Apium graveolens</i>, <i>Cleome gynandra</i>, and <i>Hibiscus sabdariffa</i> from Burkina Faso were investigated for their phytochemical profile and antioxidant activities. The high-performance thin-layer chromatography profile revealed flavonoids, tannins, and sterols in these herbaceous. The <i>Hibiscus sabdariffa</i> methanolic extract exhibited the highest total phenolic (138.4 ± 0.5 mg GAE/g DW) and flavonoid (52.8 ± 0.6 mg RuE/g DW) contents comparatively to <i>Cleome gynandra</i> and <i>Apium graveolens</i>. <i>Hibiscus sabdariffa</i> methanolic extract also presented the highest antioxidant activity (IC<sub>50</sub> = 0.31 ± 0.002 mg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay. A high correlation between flavonoid contents and hydrophilic antioxidant activities (r = 0.99) was observed, indicating that flavonoids contribute significatively to these herbaceous antioxidant properties. <i>Apium graveolens</i>, <i>Cleome gynandra, and <i>Hibiscus sabdariffa</i> constitute a natural source of phenolic compounds that could be exploited in diabetes mellitus management.</i>
文摘The research on discovery and development of new treatments for cutaneous leishmaniasis has been declared as priority. Using bioinformatics approaches, this study aimed to identify antileishmanial activity in drugs that are currently used as anti-inflammatory and wound healing by such anti-Leishmania activity was validated by in vitro and in vivo assays. In silico analysis identified 153 compounds from which 87 were selected by data mining of DrugBank database, 22 and 44 were detected by PASS (http://pass.cribi.unipd.it) and BLAST (http://blast.ncbi.nlm.nih. gov/) alignment, respectively. The majority of identified drugs are used as skin protector, anti-acne, anti-ulcerative (wound healer) or anti-inflammatory and few of them had specific antileishmanial activity. The efficacy as antileishmanial was validated in vitro in 12/23 tested compounds and in all seven compounds that were evaluated in in vivo assays. Notably, this is the first report of antileishmanial activity for adapalene. In conclusion, bioinformatics tools not only can help to reduce time and cost of the drug discovery process but also may increase the chance that candidates identified in silico which have a validated antileishmanial activity by combining different biological properties.
文摘Little is known on the association between movement behaviors including physical activity(PA),screen time(ST)and sleep(SLP)with depression in adolescents.This study aimed to explore the associations of PA,ST and SLP with depressive symptoms in adolescents.A total of 1,331 middle school students participated in this survey and provided valid data pertaining to the study variables.Self-reported questionnaires were used to collect information on participants’sociodemographic parameters.The Health Behavior in School-aged Children Questionnaire was used to assess the PA(days for moderate to vigorous PA),ST(daily hours of ST)and SLP(daily hours of SLP).Study participants’depressive symptoms were assessed using the Children’s Depression Inventory.Generalized linear models were used to estimate the associations of PA,ST and SLP with depressive symptoms.Of all the 1331 study participants,boys accounted for 51.31%and percentage of 7th,8th and 9th graders were 52.44%,23.22%and 24.34%,respectively.After controlling for sex,body mass index,grade,ethnicity,residence,siblings,perceived family affluence,father and mother educational level,only ST was positively associated with depressive symptoms(beta=0.17,p=0.005).This study suggests that excessive ST would be detrimental to depressive symptoms in adolescents.It may be effective to reduce ST for depression intervention or prevention.Future studies are encouraged to use an improved study design to confirm or negate this study’s researchfindings.
文摘Citrus aurantifolia, the family Rutaceae, which consists of 150 genera and 900 species, is gaining grounds as important source for treatment in complementary medicine, and commonly called limone. Chloroform, methanol and ethanol extract exhibited significant antimicrobial activity and highlighted the biological monitoring of activity from the methanol extract of seeds. Phytochemical screening of the diethyl ether, methanol and aqueous extracts of seeds confirmed the presence of alkaloids, saponins, sterols and triterpenes, carotenoids, coumarins, tannins and carbohydrates. Chloroform extract was fractionated by sing liquid solid column chromatographic technique and column fractions eluted with chloroform and methanol. The antibacterial secondary metabolites were achieved by fractionation of the active. The seed extracts were monitored analytically by TLC preparative and some chemical analyses of seeds have been done such as determination of ash, nitrogen, protein content.
