BACKGROUND: Tea polyphenols have been shown to protect against carbon tetrachloride ( CCl4) -induced liver injury, liver fibrosis, hepatic ischemia-reperfusion injury. In this study, we examined the effect of tea polyphenols on lipopolysaccharide ( LPS ) -induced liver injury, and explored its mechanisms. METHODS: Sprague-Dawley rats received tea polyphenols (100 mg · kg-1·d-1) or vehicle (water) intragastrically by gavage for 14 days, followed by LPS (5 mg/kg) or saline injection intraperitoneally. Liver injury was assessed by biochemical assay and pathological analysis. Serum tumor necrosis factor-α (TNF-α) levels and liver malondialdehyde (MOA) contents were determined. Inducible nitric oxide synthase (iNOS) protein and TNF-α, iNOS and en-dothelial nitric oxide synthase (eNOS) mRNA expressions in the liver were detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR), respectively. RESULTS: Administration of LPS resulted in liver injury in rats, evidenced by elevated activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), hepatocellular necrosis, and neutrophil infiltration in the liver. These responses were associated with increased serum TNF-α levels, induced iNOS protein, expressions of TNF-α, iNOS mRNA in the liver and elevated lipid peroxidation at 90 minutes or 6 hours after LPS injection. Pretreatment with tea polyphenols attenuated LPS-induced liver injury, and blunted the rises of serum TNF-α levels and lipid peroxidation and the induction of expressions of TNF-α, iNOS in the liver. CONCLUSION: Tea polyphenols prevent LPS-induced liver injury, and the mechanisms may involve the reduction of serum TNF-α levels and lipid peroxidation and the suppression of TNF-α, iNOS expressions in the liver. 展开更多

Previous studies addressing the protection of tea polyphenols against cerebral ischemia/ reperfusion injury often use focal cerebral ischemia models, and the optimal dose is not unified. In this experiment, a cerebral ischemia/reperfusion injury rat model was established using a modified four-vessel occlusion method. Rats were treated with different doses of tea polyphenols (25, 50, 100, 150, 200 mg/kg) via intraperitoneal injection. Results showed that after 2, 6, 12, 24, 48 and 72 hours of reperfusion, peroxide dismutase activity and total antioxidant capacity in brain tissue gradually increased, while malondialdehyde content gradually decreased after tea polyphenol intervention. Tea polyphenols at 200 mg/kg resulted in the most apparent changes. Terminal deoxynucleotidyl transferase-mediated nick end labeling and flow cytometry showed that 200 mg/kg tea polyphenols significantly reduced the number and percentage of apoptotJc cells in the hippocampal CA1 region of rats after cerebral ischemia/reperfusion injury. The open field test and elevated plus maze experiments showed that tea polyphenols at 200 mg/kg strengthened exploratory behavior and reduced anxiety of cerebral ischemia/reperfusion injured rats. Experimental findings indicate that tea polyphenols protected rats against cerebral ischemia/ reperfusion injury and 200 mg/kg is regarded as the optimal dose. 展开更多

To investigate their neuroprotective potential, we induced oxidative damage in spinal cord neurons using hydrogen peroxide, and applied different concentrations (50-200μg,/mL) of green tea polyphenol to the cell medium for 24 hours. Measurements of superoxide dismutase activity, malondialdehyde content, and expression of apoptosis-related genes and proteins revealed that green tea polyphenol effectively alleviated oxidative stress. Our results indicate that green tea polyphenols play a protective role in spinal cord neurons under oxidative stress. 展开更多

Purpose: To investigate the oxidative modification of water-soluble crystallins of human fetal lens with H2O2 and fourteen metal ions with or without EDTA. Tea-polyphenols (TP) was added to above solutions in order to testing their antioxidative abilities. Methods: The experiments were performed at 37℃ with final concentration of 2.5mg/ml protein, 0.1 mM metal ions, 0.3 mM EDTA and 1.0 mM H2O2. Then the TP was added to the solution with CuSO4 and H2O2, after 5 or 24 hours, the crystallins were analysed with SDS-PAGE and IEF.Results : There were marked oxidative modifications of lens protein in H2O2 and copper without EDTA. In SDS-PAGE patterns, we found an increase in those species above of bands higher than 30 kD and some diffuse bands from 30 to 17 kD after 5 hours. In IEF patterns, there were a general increase on acidity with loss of the more basic species. Surimi cake is a high value aquatic product. In this work, lipid oxidation and volatile compounds in Nemipterus virgatus surimi cake were investigated to evaluate the effectiveness of tea polyphenols in preventing or reducing lipid oxidation. Surimi cake was prepared in the presence or absence of 0.2% tea polyphenols, and was stored at 4 ℃ for 16 days. It was found that the peroxide value(POV) and thiobarbituric acid reactive substances(TBARS) values of the experimental groups(with 0.2% tea polyphenols) were much lower than those of the untreated surimi cake(p<0.05). In total, 52 compounds were presented in the volatiles of the surimi cake, such as ethanol, hexanal,(E,E)-2,4-decadienal, 2,3-octanedione, 2-hydroxy-propanamide, and 2,5-dimethyl-pyrazine. It indicated that tea polyphenols could reduce the amount of hexanal, ethanol, and(E,E)-2,4-decadienal, and delay the lipid oxidation of the surimi cake. Effects of lipid oxidation of the tea polyphenols on surimi cake were reduced during storage in ice. 展开更多

目的 探讨茶多酚(TP)调节单磷酸腺苷活化蛋白激酶(AMPK)/沉默信息调节因子1(SIRT1)/过氧化物酶体增殖物激活受体γ共激活因子1(PGC-1)信号通路对光老化小鼠模型氧化损伤的影响。方法 采用皮下注射D-半乳糖联合紫外线(UVA+UVB)照射构建... 目的 探讨茶多酚(TP)调节单磷酸腺苷活化蛋白激酶(AMPK)/沉默信息调节因子1(SIRT1)/过氧化物酶体增殖物激活受体γ共激活因子1(PGC-1)信号通路对光老化小鼠模型氧化损伤的影响。方法 采用皮下注射D-半乳糖联合紫外线(UVA+UVB)照射构建光老化小鼠模型,将造模成功小鼠按照随机数字表法分为模型组、TP低、中、高剂量组(50、100、150 mg/kg)、阳性对照组(维生素E 30 mg/kg),每组12只,另选12只作对照组在正常光照饲养,各组灌胃相应药物,1次/d,连续30 d,给药结束后对各组小鼠进行皮肤皱纹等级评分;HE、Masson染色观察小鼠皮肤组织病理损伤、表皮层厚度及纤维化程度;免疫组化法测定Ⅰ型胶原蛋白(COL-Ⅰ)、Ⅲ型胶原蛋白(COL-Ⅲ)的表达;酶生化法测定皮肤组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、羟脯氨酸(Hyp)及丙二醛(MDA)水平;酶联免疫吸附测定(ELISA)法检测小鼠皮肤组织IL-6、IL-1β、肿瘤坏死因子α(TNF-α)水平;蛋白免疫印迹法(Western blot)检测基质金属蛋白酶-1(MMP-1)、MMP-9、AMPK/SIRT1/PGC-1信号通路蛋白表达。结果 与对照组相比,模型组小鼠皮肤评分、表皮层厚度、MDA、IL-6、IL-1β、TNF-α水平、MMP-1、MMP-9表达显著升高,胶原蛋白含量、SOD、CAT、Hyp水平、COL-Ⅰ、COL-Ⅲ、p-AMPK/AMPK、SIRT1、PGC-1表达显著降低(均P<0.05);与模型组相比,TP低、中、高剂量组及阳性对照组小鼠皮肤评分、表皮层厚度、MDA、IL-6、IL-1β、TNF-α水平、MMP-1、MMP-9表达显著降低,胶原蛋白含量、SOD、CAT、Hyp水平、COL-Ⅰ、COL-Ⅲ、p-AMPK/AMPK、SIRT1、PGC-1表达显著升高(均P<0.05)。结论 TP可能通过激活AMPK/SIRT1/PGC-1信号通路,抑制氧化应激,对D-半乳糖联合UVA+UVB辐射诱导的皮肤老化具有改善作用。 展开更多

Objective Toxic metal ions have been implicated in the generation of reactive oxygen species (ROS) and nitric oxide (NO). Metallothionines (MT) and plant flavonoids have been reported in the intervention against oxidative damage. We investigated the effect of zinc induced MT and green tea polyphenol (GTP) in reducing the oxidative responses induced by nickel and platinum. Methods Zinc (10 mg/kg b. wt, sc) was administered to rats twice at a gap of 24hrs and GTP (10 mg/100 mL in drinking water) was fed ad libitum for 8 days. Nickel chloride (150 umol/kgb.wt, ip) and cisplatin (50 mmol/kg b.wt, sc) was administered to rats 24 h after Zn or GTP pre-treatment. Animals of all the groups were sacrificed 16 hrs after treatment and biochemical markers for toxicity were monitored. Results Zinc or GTP pre-treatment caused significant protection against nickel or cisplatin enhanced mortality in rats, and reduction in lipid peroxidation and NO. Conclusion It is proposed that inhibition of ROS and NO by GTP and zinc may prove useful as a selective pharmacological agent in the amelioration of metal toxicity. 展开更多

Tea (Cammelia sinensis) is the most widely consumed beverage in the world and has been reported to have unlimited health benefits due to its antioxidant properties. There is a high correlation between polyphenol compounds with antioxidant properties. Tea leaves are a major source of polyphenols. The aim of the present investigation was to determine the approximate level of polyphenols in different other parts of the tea plant to give comparative data on obtaining extracts that can be used to design products through value addition to assist in the prevention of diseases associated with oxidative stress. Twenty-one selected region specific tea varieties were used to obtain roots, flowers, leaves and barks. Leaves were prepared by microwaving and the remaining portions processed as green non-aerated teas and black aerated tea. Roots, barks, flowers were sun-dried and milled. Total polyphenol content was determined by calorimetric method using Folin-Ciocalteu reagent. The obtained results suggest that different parts of tea plant have varying numbers of total polyphenols with microwaved leaves having mean levels at 23.1%, steamed leaves (non-aerated green tea) at 22.37%, aerated leaves at 15.51%, barks at 14.92%, flowers at 10.62% and roots at 1.48%. 展开更多

目的探讨茶多酚(TP)对NLRP3炎症小体的调控作用机制以及其对脓毒症相关急性肺损伤的治疗效用。方法将60只C57BL/6小鼠随机平均分为假手术组(单纯开腹探查盲肠后即关腹)、盲肠结扎穿孔组(开腹暴露并游离盲肠后行盲肠结扎与穿孔操作后关腹... 目的探讨茶多酚(TP)对NLRP3炎症小体的调控作用机制以及其对脓毒症相关急性肺损伤的治疗效用。方法将60只C57BL/6小鼠随机平均分为假手术组(单纯开腹探查盲肠后即关腹)、盲肠结扎穿孔组(开腹暴露并游离盲肠后行盲肠结扎与穿孔操作后关腹)以及盲肠结扎穿孔+TP治疗组(盲肠结扎穿孔前1周予TP灌胃),每组20只。建模完成后绘制生存曲线评估各组小鼠对脓毒症打击的耐受情况。测定各组小鼠肺湿/干质量比值、肺组织髓过氧化物酶(MPO)水平,并对肺组织进行HE染色和急性肺损伤评分,评估肺损伤程度。ELISA法测定肺组织内IL-1β、TNF-α及IL-6表达水平;蛋白免疫印迹实验检测肺组织内NLRP3、caspase-1 p10及ASC蛋白表达水平;免疫组化染色测定肺组织内MPO、caspase-8及NLRP3蛋白表达,评估各组小鼠肺内炎症情况。检测肺组织内MDA及H2O2以评估氧化应激水平;免疫荧光共染NLRP3和NOX4蛋白以探讨二者表达及共定位。结果TP治疗组小鼠术后72 h死亡率、小鼠肺湿/干质量比值、肺组织MPO水平以及急性肺损伤评分均较盲肠结扎穿孔组小鼠显著下降(P<0.05);TP治疗组小鼠肺组织内IL-1β、TNF-α及IL-6、NLRP3、caspase-1 p10、ASC等蛋白表达水平亦较盲肠结扎穿孔组小鼠显著降低(P<0.05);氧化应激相关检测结果提示,TP治疗组小鼠肺组织内MDA及H2O2水平较盲肠结扎穿孔组小鼠显著减低(P<0.05);免疫荧光共染则提示TP治疗组小鼠NOX4蛋白与NLRP3蛋白在肺组织内的表达和共定位较盲肠结扎穿孔组小鼠减少。结论TP可通过抑制NLRP3炎症小体相关炎症改善盲肠结扎穿孔诱导的小鼠脓毒症肺损伤,且这一调控机制可能与其对肺组织内氧化应激相关蛋白NOX4的表达抑制相关。 展开更多

为降低饱和脂肪摄入过高对人体健康造成的危害,本实验选择大豆分离蛋白与大豆油为原料制备高内相Pickering乳液(High interphase Pickering emulsions,HIPEs),并以茶多酚作为活性物质进一步优化乳液性能,以替代肉丸中动物脂肪。分别选... 为降低饱和脂肪摄入过高对人体健康造成的危害,本实验选择大豆分离蛋白与大豆油为原料制备高内相Pickering乳液(High interphase Pickering emulsions,HIPEs),并以茶多酚作为活性物质进一步优化乳液性能,以替代肉丸中动物脂肪。分别选用水、HIPEs及负载茶多酚的HIPEs替代猪背脂肪,制备了6种不同的配方,并对肉丸的理化、感官指标进行表征。结果表明,相较于对照组,除pH外各指标均存在显著性差异(P<0.05),随着乳液的添加,肉丸的蒸煮得率可提高至93.59%,水分含量上升至66.91%,脂肪含量下降至8.42%,蛋白质含量上升至14.48%,肉丸的硬度、弹性、内聚性、咀嚼性均随着乳液的添加而上升,pH无明显差异,添加Pickering乳液后肉丸的L值提高,a、b值因茶多酚的添加而升高,含茶多酚的HIPEs对肉丸水分与脂肪渗出率的改善最佳,分别降低至1.57%与0.019%,硫代巴比妥酸(Thiobarbituric acid,TBA)值最低为4.12 mg/kg,添加乳液后,感官评价较对照组均有所上升。以HIPEs作为脂肪替代可有效减少肉丸中脂肪含量,提高肉丸的出品率及品质,本研究可为减脂肉制品的开发提供一定参考。 展开更多

为提高冷藏猪肉货架期品质,利用不同浓度(0.1%、0.3%和0.5%)的茶多酚、葡萄籽提取物和维生素C 3种天然抗氧化剂处理生鲜猪肉.通过测定贮藏过程中菌落总数、硫代巴比妥酸值、挥发性盐基氮值、羰基含量和总巯基含量等指标的变化,以及对采... 为提高冷藏猪肉货架期品质,利用不同浓度(0.1%、0.3%和0.5%)的茶多酚、葡萄籽提取物和维生素C 3种天然抗氧化剂处理生鲜猪肉.通过测定贮藏过程中菌落总数、硫代巴比妥酸值、挥发性盐基氮值、羰基含量和总巯基含量等指标的变化,以及对采用抗氧化剂处理后冷藏猪肉的脂肪酸含量变化进行分析,比较不同浓度3种天然抗氧化剂对猪肉品质的影响.结果表明,经3种天然抗氧化剂浸泡处理后,均能有效抑制冷藏猪肉微生物的生长繁殖及脂肪氧化,同时延缓蛋白质水解,降低饱和脂肪酸;贮藏12 d时,0.5%茶多酚组的菌落总数和挥发性盐基氮值含量分别为4.85 lg(CFU/g)和8.81 mg/100 g,明显低于对照组和其他处理组,硫代巴比妥酸值略高于0.5%葡萄籽提取物组(0.221 mg/kg),为0.225 mg/kg.综合菌落总数、硫代巴比妥酸值、挥发性盐基氮值、羰基含量、总巯基含量和脂肪酸组成分析,0.5%茶多酚组具有最优的保鲜效果. 展开更多

氧化应激会造成脂质过氧化,诱导细胞凋亡且损伤细胞遗传物质,进而影响动物正常的生长发育过程。本文对喂饲茶多酚的小鼠进行氧化应激诱导,通过监测诱导后小鼠的体重和进食量,检测其血清和肝脏抗氧化指标,研究茶多酚对小鼠氧化应激的缓... 氧化应激会造成脂质过氧化,诱导细胞凋亡且损伤细胞遗传物质,进而影响动物正常的生长发育过程。本文对喂饲茶多酚的小鼠进行氧化应激诱导,通过监测诱导后小鼠的体重和进食量,检测其血清和肝脏抗氧化指标,研究茶多酚对小鼠氧化应激的缓解效用。结果显示,氧化应激诱导前,各组小鼠的体重及进食量差异无统计学意义;氧化应激诱导后,应激组血清中的超氧化物歧化酶含量及总抗氧化水平显著下降,而茶多酚可以有效提高血清的抗氧化水平。同时,应激组小鼠肝脏过氧化氢酶及超氧化物歧化酶含量降低,丙二醛含量升高,而茶多酚处理可提高小鼠肝脏过氧化氢酶及超氧化物歧化酶含量,这表明茶多酚可以有效提高血清抗氧化水平,并保护小鼠肝脏,降低敌草快引起的氧化应激损伤。目的在于为进一步研究茶多酚缓解畜牧养殖中氧化应激提供参考。 展开更多

为了研究茶多酚对鲈鱼在冷藏期间鲜度和肌原纤维蛋白氧化的影响,将鲈鱼浸渍于0.2%的茶多酚溶液中60 min,取出后沥干,于4℃条件贮藏。以K值、TVB-N值、肌原纤维蛋白盐溶性、Ca2+-ATPase酶活性、表面疏水性、总巯基和活性巯基作为评价指标... 为了研究茶多酚对鲈鱼在冷藏期间鲜度和肌原纤维蛋白氧化的影响,将鲈鱼浸渍于0.2%的茶多酚溶液中60 min,取出后沥干,于4℃条件贮藏。以K值、TVB-N值、肌原纤维蛋白盐溶性、Ca2+-ATPase酶活性、表面疏水性、总巯基和活性巯基作为评价指标,分别于第0、2、4、6、8、10 d测定各指标。结果表明,茶多酚可以显著地抑制鲈鱼在冷藏期间K值、TVB-N值和表面疏水性的增加(p<0.05),在整个贮藏期间肌原纤维蛋白的溶出量、Ca2+-ATPase活性、总巯基和活性巯基的含量显著高于对照组(p<0.05),到贮藏末期第10 d时茶多酚处理组与对照组相比分别高出20.91%,57.14%,18.42%和89.47%。综合各指标检测结果可知茶多酚能够较好的保持鲈鱼在冷藏期间的鲜度,抑制鲈鱼在冷藏过程中肌原纤维蛋白的氧化。 展开更多

用过氧化氢(H2O2)建立体外奶牛乳腺上皮细胞的氧化应激损伤模型,研究不同质量浓度茶多酚对氧化应激所致奶牛乳腺上皮细胞损伤的影响。MTT法检测细胞存活率,比色法检测细胞培养液中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH... 用过氧化氢(H2O2)建立体外奶牛乳腺上皮细胞的氧化应激损伤模型,研究不同质量浓度茶多酚对氧化应激所致奶牛乳腺上皮细胞损伤的影响。MTT法检测细胞存活率,比色法检测细胞培养液中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和乳酸脱氢酶(LDH)活性,流式细胞术检测细胞凋亡情况,分光光度法检测天冬氨酸特异的半胱氨酸蛋白酶3(Caspase-3)的活性。结果显示:与H2O2损伤模型组相比,20~100μg.mL-1茶多酚处理组的奶牛乳腺上皮细胞存活率、SOD活性显著升高,而MDA含量、LDH与Caspase-3相对活性与细胞凋亡率均下降,其中以100μg.mL-1茶多酚处理组效果最显著,说明一定质量浓度的茶多酚可以缓解氧化应激所致的奶牛乳腺上皮细胞的损伤,提高细胞存活率,抑制乳腺细胞凋亡。结论:茶多酚对乳腺上皮细胞氧化应激损伤具有保护作用,其保护作用可能与降低MDA含量,增强SOD活性及抑制Caspase-3活性有关。 展开更多
